Evaluation and Research. One thing she mentioned was interesting. It
sounded like that FDA was challenging NIH on the reliability of academic
research data. A huge amount of money was wasted because many researches on
drug development in industry were misled by unreliable data from academic
research groups. NIH fought back calling FDA's lack of scientific awareness.

做分子生物学好些年了，我觉得从转染的细胞上提取RNA一直提不好。按照标准的
Trizol protocol，但是我的问题：
1、操作非常慢，2个24孔的板子，我差不多要半天才能提到粗制的RNA (异丙醇沉淀得
到的）
2、RNA提取的质量不好，260/280低于1.9， 260/230低于1.5
3、由此得到的qPCR效果，也不是很好。同个处理的3个孔之间，variation很大，那个
误差线可以让人上吊。
4、找不到一个好的方法，去掉基因组。
哪位能提供一个reliable的, DNA free的RNA提取Protocol？ （不要说kit：因为50个
样品的kit，只能做一次实验）
有文献支持最好。

gas, 平均一年维护800刀左右?
97年前的有线路过热问题, 所以想考虑一下97-99的.
据说s320也不是很费油, 只是有点underpower, 所以transmission容易出问题?
但是我这里mid-west觉得很难找到s420, 看了一个月, craigslist没有97~99的, 93~97
也只有几辆...s320比较多, 有10个左右吧...s420以上的空气悬挂容易坏...纠结中...
坛上考古没找到资料, 想问问各位有没有开过或者了解w140这车, 有什么建议呢? |

几周前看了几辆 VW 的车，比较喜欢Tiguan，后来觉得可靠性不太有保证，修起来又贵
，所以没买。dealer 发信来问，我就说了我的疑问，他给我回了很长一封信。总结来
说：
1. VW很多技术是从Audi等高档车移植过来的，很多年纪大的人不太理解，导致 JD
Power 等可靠性评价低。
2. 2011年VW做了些调整，降低部件的价格。具体怎么降低的没说，我正等回答。
3. 很多人到 VW trade in的车都是100k以上，然后因为觉得可靠，又都买新的VW。
我自己开 Golf， 2年多没什么问题，很喜欢，但是以后怎么样心里没数。
这个VW可靠性低的reputation都是网上看来的，好像是电子部件容易出问题，机械部件
还好。

AT是靠齿轮，容易理解。以前scooter之类小engine
用橡胶皮带CVT还比较容易理解一点，马力小，橡胶不太滑。
汽车CVT就靠两个光滑如镜的斜面夹着一个光滑如镜的
金属链，而且不是夹死，还能随着宽度随时调整上下位置，
这东西怎么能挤住的同时还能灵活上下走还能高效率呢？我
主观想象一下觉得很容易打滑，时间长松了岂不是效率更会

下面是一封申请EB2 NIW I-140的petition letter的范例。这是多年前公开发表在
MITBBS“落地生根”版上的一个帖子。现在重贴于此，供大家参考。
个人点评：EB2 NIW的申请是技术活，满足NIW的三个方面都必须论证到位，否则就通过
不了；而且论证过程中，有些关键词必须出现在移民官的视野里。这封petition
letter基本上达到了这几方面的要求。更难能可贵的是，这封petition letter的后面
，还列出了所有的exhibit，值得参考。

的petition letter，开始准备NIW。看了几篇模板，发现有些疑问。
1. NIW 不需要列表格，油灯图等吗？为什么我看到的模板都没有这些，只有Exhibit？
2. EB1A的petition条理比较清楚，比方说我要claim三项，就从1，2，3几方面阐述。
但是NIW怎么感觉条理比较混乱呀？

reliability这东西有稳定性，或者在相当一段时间内是稳定的，过去相当一段时间内
某品牌的车reliable，那么同品牌的新车reliability肯定也会不错
你无非就是对jetta等车的reliability有想当然的好感，想当然地觉得这车虽然过去
reliability不咋的，但近两年应该会很好（虽然这根本不合逻辑），然后拼命为自己
找支持自己观点的理由，哪怕这理由再荒唐
一张图也许说明不了啥，很多图加无数客观的历史评价加无数当事人的反馈就能说明问

要非要买日本车么？reliable？那是10几年前的情况了吧。这都2012年了，信息革命都
不少年了，车场造车技术控制都不错了，当年美国车给人笨重费油的感觉在现款车上早
已不在了。稍微懂车的把日车和其它车比一比就知道，日车设计跟不上，技术也不占优
势，安全性也打马虎眼，就拿个reliable当幌子做卖点骗老中（非小留）。

这个也太显stereotype了。ps.烤肉什么时候能把那恶心的4速变速换了，看看人家
focus。好吧有钱了，咱别总lexus讴歌行么，买车前多看看，做做功课，别局限在自己
的思维定势里觉得就是日本车好，这也太out了。我知道一人开了4年的sienna发动机出
问题了结果trade in又换了辆新的sienna。。。比较无语，4年发动机就坏了，不悔改
啊。
还有论价格，日车不占优势，跟同级车比，两田卖的死贵还压不下价，图便宜更不能买

其实今天没想写这个帖子的,但看了wanttobefree发的问rav4的帖子,越回复越长,所以
干脆就另起个帖子说说我开rav4的感受.
我自己觉得2013新出的样子比2012和之前的样子要好看. 前些日子看了rav4 13的样子,
真有把我现在的09年的卖了或者trade in, 然后换成13年的冲动, 但后来一想要因此破
费个1万刀, 还是算了吧. 而且开suv倒车rear view camera还是有帮助的. rav4后边的
窗户本来就小, 还有个破备胎在哪里挡着,所以几乎就是个摆式. 光靠2个side mirror,
倒车有些时候挺不方便的. 而且车的底盘高,副驾驶前边的bumper几乎就看不到.
我现在开的就是09年的rav4, 11年11月thanxgiving那会儿买的. 当时跑了28k mi, 4wd

介绍一下需求。最近由于家里要添一部车子，主要用途是我上班以及一家三口周末的出
游。家里现在的crv我老婆平时带孩子开。由于她驾照还没考好，我从8月开始到年底都
可以用来挑选一部车子。
需求优先级：
1. sportiness 本人不算专业车迷但是很喜欢车，开车。
2. safety (IIHS recommended) and back seat comfy 老婆孩子周末要坐，但是腿部
空间无所谓，老婆很瘦。
3. price <= 30000 before TTL 还要买房子
4. reliability above average 理由同上
选了两个月左右，基本上还确定不了。参考了consumer report以及kbb还有truecar和
很多dealer的报价，基本上如下几个option
1. New 2013 G37 Journey without Premium pacakge (less point going for an
used G37 while a new one is this cheap right?)
