address the same question: why chemotherapy drug,tamoxifen, suppressed
breast cancer cell growth, but promotes endometrial tumorigenesis. Shang's
paper showed that tamoxifen could trans-activate PAX2 gene expression in
endometrial cancer cells, and the activation of PAX2 resulted in the

光有RT-PCR or promoter reporter assay 是不行的,因为还有一个alternative
splicing的可能性你们还没有排除。在完全澄清之前最好不要贸然往translation regulation 做。
第二,如果你们的结果是对的话,下面会有很多有趣的东西,但是难度也不小。
你们的蛋白最有可能的是一个释放压制RNA表达的因子。
比方说 zip code binding protein(ZBP)这个蛋白是抑制RNA translation 的。然后
有其他蛋白可以释放这个ZBP而让蛋白开始表达。你们的这个蛋白A就有可能是类似这个
释放因子。
类似的还有一些蛋白,可以释放miRNA而使得某些蛋白可以进行翻译。
要知道更多的信息的话可以去查"release of translation repression"
很可能涉及翻译调控的比较初级的问题。

我听过他的演讲,一个大胖子,来势汹汹,语速非常快。要知道他做很多DHS啊TF啊的
binding site,涉及非常复杂的transcriptional network;但他对几乎所有的
transcription factor啊,各种cell line啊几乎是如数家珍;你随便问他任何问题,
比如how much percentage,他都可以立刻回答出来。。讲到最后大家都感觉他是在上
演精彩的talk show,已经不想去听他的演讲内容了(反正那么多detail也听不懂),
都是去看他的个人演讲风采了。
相反同一天,有个哈佛著名牛B实验室的一个中国postdoc来应征faculty的位置,。。
本身我们中国人口语就比不上老外,他还无精打采豪无气势的在讲。。。讲到最后大家
人都走光了。。。老外会不会进一步加深对我们中国人“smart silent mathmatician
with 5000 years of sexual repression"的印象呢。。 |

negative?) did not; it only suggests that the endogenous Hes1 in the cell
line you tested was not invoved in maintaining the basal activity of that

在早期的promoter methylation repress gene expression的问题上搞得有点over
simplified.现在整个field都在拓展到whole genome,发现了gene body的methylation
其实是促进表达的。而且新的DNA modification比如hydroxylmethylation的发现完全
改变了对一部分DNA methylation数据的解读。
这种因为技术本身限制的问题并不能说以前的research都是trash.我也知道bisulfite
降解DNA但不能说因为降解DNA就回家睡觉啥也不干了...听蝲蝲蛄叫还不种地了么?而
且这不就是知道这个问题的人找出新方法大显身手的时候么? |

t*******y 发帖数: 36 | 48 楼主感兴趣的话可以看看UCSF Jonathan Weissman实验室,听说他们实验室根据cas9搞
的cas9a(activation)和cas9i (repression)效果还不错,暂时主要应用在genomic
screen上。我觉得以后用来做真核的synthetic biology非常好。同样在UCSF 的
Wendell Lim前一阵子把一些基本的基因调控原件应用在免疫细胞里面,效果还不错,
以后基于synthetic biology的基因治疗可能也有很好的发展....
搬到mit的james colins 也很好。
纯从转录调控说,个人感觉e coli 和 yeast 差别不是特别大,从 yeast 到其他的真
和模式生物差别非常大。虽然yeast同样有histone,但是对转录调控的影响比较简单。
在yeast里大概影响转录调控的variables都了解的差不多了,直接manipulate这些
variables的工具也相对齐全。在其他真核系统里,定量的研究感觉上还都是停留在对

王盛毕业于北京协和医学院,获得博士学位,到美国后在哥伦比亚大学和纽约大学做过
博士后,2000年受聘到波士顿大学医学院癌症研究中心工作。他在乳腺癌研究方面有不
少成果。
王盛涉嫌造假的这两篇论文,第一作者均是张葆华(Baohua Zhang,音译)。张葆华是
王盛的妻子,1984年北京医学院牙科专业毕业,其后在北京协和医学院获得硕士学位。
不但是“协和有人造假”,而且居然是“协和毕业的一对夫妻合伙造假”,奇闻也!
现在韩春雨和沈啸两个作者也是协和人,成双出没是协和人造假的特征?需要N+1的例
