endothelin-1的含量。现在的问题是——standard老是做的很烂。我也不知道哪里出了
问题。我前后用了两个Kit，第一个Kit的standard没问题，但是第二个（我用这个做了2批）怎么做
standard都不行。
附件中是manual中给出的样品。把浓度取log10做X轴；Luminescence取log10做Y轴，应
该是线性。我做的线性非常差，而且低浓度的standard的deviation很大（duplicate）
1）会不会是第二个Kit的antibody公司coat的不好？或者质量有问题？
2）log后才线性，那这个kit是不是本身就很容易出问题？Assay Design有个Kit，是直
接用原始数据划线，就是线性。这个会不会更精准一点？我一直不喜欢4 parameter或者其他方式
处理原始数据后才线性的kit。
3）最后一步incubation（产生luminescence的步骤），manual上说要Incubate 5-20分钟，而

IHC, ChIP, ChIP-seq, Fluorescence Microscopy.......如果抗体好，手笨点，硬件
差点，都是能做出来的。如果抗体不行，上帝他老人家来了也没辙。
其实生物男很多时间都花在尝试不同的抗体，求老板买更多抗体，以及向抗体公司投诉
上了。
可是如何选抗体呢？哥根据自己的经验体会，给大家提点建议。
在美国有300多家卖抗体的公司，全世界范围就更多了。可是我们常接触的就那么几家
。我把他们分成两类："包罗万象型"和"少而精型"。
最著名的“包罗万象型”公司有三家：AbCam, Sigma和Santa Cruz。这三位的产品也太
全了，无论你需要什么抗体，他们都有。至于是否能用，那就再说了。 我个人的经验
是：能用率大约有20％。 所以我很奇怪楼主会觉得AbCam抗体质量好。这三位大哥中
SantaCruz还算好点，毕竟便宜得多，给的量又足。万一赶上你走运遇到一支好的抗体
，够你用到当正教授了。

从我有限的经验，在研发的层面，外包的基本是点，很少涉及到面。而核心项目，基本
不会外包。
把整个rnd center砍掉，不光是涉及到人员，还有放弃整个center 硬件的setup，以及
和local的方方面面connection。
这样的决定，是strategic上的。公司报表，需要做漂亮。 |

股价是裁员裁掉研发等运营成本做出来的，但后续没有研发，药厂也终究是无源之水。
专利药的专利期到了，新药研发太贵，FDA的批准要求越来越高，医保改革导致整体药
疗市场萎缩，药厂非常难过，RND也就只能放一边。 |

lab那种rnd，确实拿不到什么钱我还是有自知之明的。何况还是小地方。也许那地方就
是花不了什么钱吧，不过还是觉得有点少。主要是我又没成家，那个地方基本就是一毛
钱的亚裔都找不到。不过估计比较适合40岁以上的人，有pension的。我在考虑等我干
不动了再去应聘一次。 |

的，把责任都推得干干净净，把功劳都揽得彻彻底底。RND就是给人家卖命干活的，最
容易被欺负。

RND不稳定，也有可能是所有的model parameter都是stochastic的，不过你说的数据真
的变化太大，我也不知道怎么搞。
我记得有一篇文章用了两个distribution function来fit的。好像没有对strike求倒，
不过我记得不是特别清楚了。你可以看看下面这个文章

，熟悉产品标准的管理等。
另外RA工作经常要和各个部门沟通的，你可以展示你在以前的工作中是怎么和生产，RA
,QA，RND,supply chain，marketing等其他部门的人合作的，有没有曾将参加过什么特
别的project, 你有没有strategical thingking, how did influence other
teammembers, 等等。
再有，估计你是博士吧，show你的学习能力，适应新环境，面对新问题解决问题的能力
。做RA也是要不断学习，适应法规变化的。
最后，不知道他们公司是不是有中国亚洲的业务，如果有，你的中文就更是优势了。
