请问这段文字如何运用到具体中，还有，我用2011年初BECKER， 现在UPDATE里说这个
LIMIT IS 500，000。
AMT 的数字从800，000变成了2000，000。现在考试到底用哪个数字啊。。。。救命 |

Basically,
1. snap-freeze the tissue (presumably unfixed) in Tissue-Tek by liquid
nitrogen.
2. cut sections(I prefer 5-7um). It seems the thinner, the better the
sections stay on the slide. We use plus slide from fisher
3. air-dry at room temperature for 3-5 hours.
4. 2xPBS 3min each to clear tissue-tek
5. Postfix in 2% to 4% PFA in 1xPBS for 5-10 min
6. Wash away PFA by 3xPBS 3 min each
7.PBS+Triton x-100 0.05% 5 min(optional)

incubation 过夜70度,发现tissue结构收缩,如附件所示,图示是我已经完成整个in
situ的结果,事实上我在incubation以后就已经发现tissue结构收缩了.
我问了一些人说需要在fixation的时候做decalcification,我做了decalcification,结
果in situ 的时候tissue还是会收缩,事实上看起来更严重.
我不知道这究竟问题出在什么地方,看起来像是tissue attach不牢固,导致容易变形.请
问有什么方法可以使tissue attach牢固么?
我是根据下面的步骤做的:
1. cut the arms and legs
2. 4% PFA (pH 7.1-7.4) overnight at 4C.
3. PBS three times for 5 minutes
4. 14% EDTA for decal 3 days.
5. wash with PBS

latter one gives you much better resolution.
For frozen section, if the tissue is embedded in OCT or other fixative, you
may store them in -80'C. No experience with fresh frozen samples. |

$5400的费用不cover。收到邮件之前毫不知情，不知道CFA是out-of-network，当时催
产中途胎儿心跳减弱，护士联系Ob后说马上做手术，很快就呼啦啦地被推进手术室，也
没人征询我和LG任何事项。邮件里还说医院appeal要求支付，但被驳回，这种情况我
该怎么办？ |

定命运的roster们套瓷有没有什么正面作用。当然直接讨论proposal是非法的。但是写
个EMAIL问问好，会议上主动套套瓷，或者邀请他们做个seminar什么的。这样的方式这
样的时候可能会比较敏感。不知道作这些会不会有负面作用或者正面作用。
特别想听听那些曾经做过panel roster的老师们怎么看这个问题。

吃顿饭不算什么。你可以照实跟PO讲: 你们最近一篇合作文章在x年前，现在没有on-
going collaboration, 没有合作的grant。 不过我觉得这PO的问题也有点儿奇怪，因
为即使一个reviewer和你有personal tie，他在讨论你的proposal时recuse自己就好了
，也不至于由此决定study section啊？ |

the 6/7 reviewer better have a very good reason. My old PI's grant high
score reviewer has no reason and it shows in the study section. I think the
reviewer actually made the PO mad, because something was mentioned even in
the counsel meeting.

And we are in a very small field. Their proposals are in different study
section though.

PO说这个要把自己想象成applicant的lawyer或者是prosecutor, 去study section就像
去court 一样。可是如果没有lawyer那么好的口才怎么办呢？我也很不喜欢一直抢话那
