家的基础,在性选择理论上讲是r selection,就是追求快的种群规模扩大速度。既然是
强调种群规模,对于种群里个体,肯定是忽视的。就好比男人也是进化出r selection,
所以精子动辄上亿的随便浪费射在墙壁上;而女人进化出K selection的策略,就是注
重个体质量而不是群体数量,那么才会小心翼翼的选择对方。
西方因为地缘关系,没有足够大的辽阔土地来进化r seletion, 他们的帝国也好,王国
也好,人口规模总是很小。当然罗马和拜占庭都很大过,但是跟中国的大一统王朝比起
来,它们其实是地域异质性很高的,分裂成很多小的文化区域,互相之间没有太多人口
杂交或者文化认同。这种情况下,个体的重要性就升高了,所以个人主义,以及民主起
源于西方也不奇怪了。
从近现代的历史看,西方文明先是通过武力、然后是文化科技和经济软实力主导了东方
,崇洋媚外自然变成一种主流心态。一开始只是在军事、技术和体制上的,久而久之连
文化、审美也被“扭曲”,放弃了对东亚面孔平面而精细的欣赏,转而接受西方对挺拔

Quadra-Drive II 也是用cluch pack。如果quadra drive II 算 full time,selet
track II 也算
jeep 的4WD 就算是不用硬齿轮连接,也应该是做的很耐操。 这两个jeep 的
differential 都在trail 上被人狂操,complain 的人不太多
另外还有2.71:1 低速档同时锁中差,扁死subaru没问题

为了我家草坪,去图书馆借一本专门的书,
书上说:杂草如果占草坪%30,你几乎就没救了,再怎么也是斗不过,
还不如全部从来。
你的照片看来,杂草很多,要我就从来。
我的草坪杂草没有》20%,但是因为要换成更加hardy,可以经常
让小孩踩踏的草种,所以就重新来。
具体步骤: 买non-seletive 除草剂,杀一遍,
最好等一阵子,里面草再长出来,再一遍,因为除草剂只能除
长出叶子, 土里面好多原来草,杂草种子,一下杀不掉。
我时间紧迫,跳过这步,后来还要费时间处理没杀死的杂草,得不偿失。
然后翻土加氮肥
然后让土settle down 4-5天。
然后挑个今后几天(最好5-7天,看你草种发芽时间)是阴雨天,撒草种,加
层top soil. 最好密点,比intrustion更密点,可以挤掉很多杂草,同时如果有什么害
虫,还是突然大太阳暴晒,不会出现草体秃头一块。
种子发芽的关键在于水分加和土壤接触。
然后就是发芽长大到1-2寸之前要勤浇水,少让太阳暴晒,
到1-2寸后以后就容易了。 |

分型就有好多种,而pathology的分型是多么的粗。说一句负
责任的话,每个人得的癌症都不同,即使是同样的病理分型是
一样,它们也是不同的癌症,(尽管它们有共性)。这个原因
很简单,癌细胞积累的突变的过程是个随机的过程,所以最终
产生的癌症必然是unique的combination,复发后和复发前
比又是一种新的疾病(当然有共性)。所以如果人类能够所谓
的“攻克”癌症,必然要走个体化的治疗。我的envision将来
的癌症治疗,必须先要对癌症样品进行whole genome
sequencing,然后看有些什么gene mutation,再相应的
选择治疗方案。当然这个前提是我们要先找到针对特定
mutation的治疗方案。
我的这个预测还是低估了癌症的复杂性,因为已有大量的证据
表明即使是一块癌症组织内部都有非常大heterogeneity,
就是说有好多的不同的癌细胞,当然某个population是

的seletion,然后挑克隆,虽然没有看到GFP的信号,但是我还是冒险试了一下EB的培养,
结果发现EB的表型和预期的一样,但是用WB检测的时候,看不到融合蛋白的表达.老板看
了不是很相信,非要做出wb,检测到蛋白,才能说明gene 的功能和你的EB表型的关系.我
搞的那个很郁闷呀, 构建, 转了8个gene呀,3个多月的工作呀,伤不起呀.
要是能检测到蛋白,这个project都差不多可以close了. |

如果经费不是问题,当然可以制两种library。
对已知物种一般没必要normalization。基因表达量太低,生物学意义也不明了。
对未知物种,当ribosomal depletion 或polyA seletion 成功,并且sequencing
depth足够高,绝大部分有表达的基因都能测到。
normalization可以降低高表达基因的读数,但我也不确定会不会提高低表达基因的读
数。你可以先拿public data做research看变化有多大。无责任推测不会有太大帮助,
因为被测出来的基因都起码有一定的表达量。 |

的修饰等各种可能性。出来的抗体就会有一小部分对某种抗原有一定的结合,这个是
clonal seletion。 被selected clone就有机会产生变异(mutation),有一些变异会
使特异性更高,所以可以下一轮被clonal selected。这样几轮下来最后产生的就是特
异性抗体。
