在有的sample总是有很强的信号，当然也有的 sample IgG pull down 下来的很弱的。
我想请教一下如何降低这种non/specific binding, 我用的是 invitrogen dynabead。
以前用过 millipore agrose bead ,记得 agrose bead 要用 salmon sperm DNA block
的，我想请问一下有没有人用salmon sperm DNA block dynabead。 问过 invitrogen
technical support，他们不推荐，但是也说不出个理由，只是没有人这么做过而已。
如果有人试过不行，那我也就不必试了。多谢 |

This topic has been studied already. There was a Nature paper about it.
Briefly, the egg will scan the genetic information of all the sperm cells
that are attached to the surface of the egg, then pick the sperm cell that
has the biggest difference from the egg's own genetic sequence. |

sperms are fine. he has done semen analysis several times at different labs.

以前ob让lg在其clinic做sperm analysis， 说是low normal可以怀孕， 结果我7个月
没怀上， 在这之前已经试了10个月。 这个ob真是害死人， 当然也怪我们太相信美国
医生了。 以后凡是都问问2nd opinion很重要， 希望看到此贴的jm千万要吸取我们的
教训， 不要浪费宝贵时间啊。
在我妈的建议下， 逼着lg去看urologist， 做全面sperm analysis， 结果打击巨大，
合格精子头only13%根本怀不上， 诊断是精索静脉曲张， 2个建议做IUI/IVF或做静脉
手术。
想问问有没有类似经验的jm的lg是做手术呢， 还是就直接上IUI/IVF了？
我的想法是先做手术以提高合格精子头到正常的30%， 然后试自然孕， 怀不上再做IUI
/IVF。 做手术后恢复可能会耽误点时间， 可我觉得值得， 不知对不对？
lg今年40了， 我下个月34.

我不敢多买。
一是没try过不知道这一批的质量如何。 第一次的两管都低于2milion. 第二次换了
donor, 每管都有30million. 要再买时，发现sold out了。
还有总是幻想着也许这次就成了， 那多出的sperm vials退回bank 要损失好多钱。 唉
，没钱人的烦恼。
想再试试IUI. If I am not successful this year, I will definitely want to
start IVF this year. My insurance covers $20,000 for infertility. I only
need to pay for the donor sperms, lab handling fee and all the drugs. so I
can still afford for a few tries. |

我也不想整啥虚头八脑的，这也不是啥medical research，找sperm donor的目的实话实说就是想生个娃。打算以artificial insemination或者IVF的方式受孕，不是自然受孕。男欢女爱需要很多机缘因果，两情相悦其实不易，我又很固执，还是相信爱情，所以也是无可奈何。
