因为这个病是病毒性的。所以也没有特别有效的药治疗。只能控制。现在正在滴眼药水
，吃lysine和口服的一种药。
问题是，好好的一个白猫，被stain搞得脏兮兮的。已经每天拿棉球蘸水擦多次了。还
是不好清干净。有经验的朋友推荐个好用的产品吧？
另外，如果是可以分享治疗和控制这个病的经验就更好了！！ |

For ideal flow cytometry, you have to change your filter system to get
optimal result. For confocal microscopy, try to put in approiate filter
(usually it's hard), or you might need to do some real good compensation
(sometime
difficult, too). So, the third choise for confocal will be monitoring only one
channel (decrease of orange or increase of green)
If the nuclear is stained, that means your JC-1 concentration in the cel |

here is what we do: 30 um microtome brain section and focus on Hippocampus:
we mount the section and dry for several hrs and 0.01M(PH6) Citric acid
for 15 min at 95-100, and then
cool down twice in TBS for first time 2-3 min, second time 10 mins.
blocking with 3% DNS plus 0.3% Tween-20 for 1 hr and primary ON in
blocking solution,

你们做这个干吗用啊？过去很多人做sp是因为在老鼠上没有好的stem cell marker，而
sp有stem cell的properties，所以用来做stem cell selection。看你要研究的问题，
现在有很多markers可以用了，为啥一定要做sp？

做一个实验，需要co-stain cell surface marker 和intracellular protein,然后拍
照。细胞是adherent 的。
因为担心fixation和permeabilization会影响cell surface marker,所以准备先活细胞
染 cell surface marker，然后在fix，permeabilize染intracellular protein.
我的问题是可以活细胞染 cell surface marker， 马上fix，然后再染 2nd antibody
anti-cell surface marker吗？
还是必须活细胞染 cell surface marker，再染2nd antibody anti-cell surface
marker， 完了才能fix？

大致是这样的，我有个cell sorter，能把某种特定类型细菌抓住，不过可能对细菌有
些损伤。我希望抓住细菌后，用某种dye染色，看看抓住的菌是否有原本是alive的。也
就是说，不管我的sorter对它产生了什么damage，只要进入sorter前是活的，我就希望
输出positive结果。
不过听说live/dead stain机理有好几种，有的依赖膜完整性，有的依赖内部的代谢反
应，我不知道选哪种对我的要求更契合，请大牛给指点一下，多谢了！我目前考虑
Calcein AM，不知道这个适合我的要求么？谢谢！ |

发帖数: 79 | 35 最近在用DAPI染丝状真菌的细胞核，步骤完全是照着教科书或者paper做的。突然发现
DAPI可能会导致细胞核破裂，造成了严重的fake结果。
比如without DAPI staining, 用显微镜观察NLS-GFP是细胞核定位的；但是在DAPI染色
（10min）后观察，GFP的定位变成 cytoplasmic了。然后我做了一个time elapse的实
验，很诡异的发现在某一个时间点细胞核突然间破裂了，定位在细胞核里的GFP突然
distribute to the entire hyphae。
请教各位大虾这是因为DAPI的毒性照成的吗？因为sample比较多，实在不想fix以后再
immunostaining 观察，除此之外有什么好方法吗？十分感谢！ |

centriole 信号很强，细胞核中gamma-tubulin信号很弱，细胞质中的信号强一些。看
文献好像是这样。
但是换成PFA固定后centriole信号比较弱，还是能看到了。奇怪的是细胞核中的信号强
一些。到底哪一个染色的结果真实一些？
我还共染了另一个蛋白，带FLAG tag不同的固定方法，得到的结果也不一样。多谢！ |
