缺少这方面经验。所以我在想是否有必要使用lamellipodia, filopodia的marker进行
特异的标记,以得到更明显的效果?
我查了一下,有的文章用p16-arc (arp2/3 subunit)标记lamellipodia,用fascin标记
filopodia,然后用immunofluorence对细胞进行标记,观察荧光变化。但是不知道这样
是否可行?是否必要?还是仅用phalloidin染色就可以达到目的? |

我是从C.D.Allis,Danny Reinberg,Tony Kouzarides,Yi Zhang, Yang Shi这几个
人发的文章看起的,这几个人基本上就是目前做表观遗传最牛的一批人了。然后你看看
他们文章citation里面出现频率比较高的人名,那往往就是大牛了,然后一点一点扩展
。个人感觉看CNS及其子刊还是王道。另外推荐一个很好的网站,http://www.epidna.com/ 可以查这个领域最top的lab,也可以查该领域最新的论文。

必乐意去搀和,只是碰巧路过的时候被扯了进去,从此一发不可收拾。记得念书的时候

被发现在dna repair 里有功能,觉得很纠结,怎么会这样,人物啊,地点啊,时间啊
都对不上哈。后来见有个理论讲,其实人家天生就是repair protein,碰巧大小合适被
ribosome收容去撑场面了。。

话说我的两个实验室合作的题目,第一件事其实就是提蛋白,要大量的蛋白(做过
稳态前动力学/用过quench flow的同学都知道呵),但是那个蛋白其实一直是老板的
一大头疼, 虽然我们当时号称是唯一一个能提取表达提取那个重组蛋白的实验室(多
subunit,且不能用e coli表达,又不能加任何tag),其实整个过程很多时候靠运气,
投入大,产量很小,且不好重复,不好控制。不过说来,但凡是大重组蛋白都会有这个
问题,我后来跟老板说,您看就算是大药厂上市20几年的重组蛋白药,头疼的也还是这
些,您就那么几个人,已经很牛了。
但是想想,我当时其实处境很危险,这个题目对大小老板都不是太重要的东东,
成功了锦上添花,不成还可以赖到另一个人头上,而且他们各自都不清楚该对我有多少
指导,毕竟还有另一个pi 在,管得太多也不好,所以我基本处于3不管地段,不去实验
室都没人知道。如果我是个postdoc,那可能会是很理想的状态,但我当时还是个刚知

s******y 发帖数: 28562 | 46 蛋白降解一般而言可以至少有两个途径:proteasome, lysosome, 再加上limited
proteolysis by calpain or other proteases.
前者可以用MG132 or beta-lactone
中间可以用100mM NH4Cl
后者可以用calpain inhibitors
MG132 本身是一个小多肽模拟物,一般认为是堵塞chymotrypsin-like cleavage
activity. 所以其实也不是一个很干净的inhibitor, cross-react with calpain and
other protease. 但是它在浓度比较高的时候(20uM) 对proteasome 抑制
是非常明显的。
beta-lactone 主要通过和proteasome subunit crosslinking 而起作用,比
MG132 专一性稍微好一点点,但是抑制作用不是特别明显。
不管哪个inhibitor 都不能完全解决问题。我不能说你的导师到底是对还是错,
但是你应该找其他方法来佐证。比方说,你可以看看蛋白用这个东西处理的时候,

么生物方向的insight。我从一年级开始就找了隔壁另外一个晶体大牛挂靠,现在已经
解了一个结构,老板一直对我用结晶做他的project非常支持。
之前还好,老板虽然没什么专业上的帮助,不过也不添乱,我在隔壁大牛指导下做的很
顺。现在文章送了出去,明显老板对于后续计划有了一些想法,不过这些想法实在是让
我哭笑不得。
本来那个晶体结构一出来,我就觉得其中一个功能域还有东西可挖。我当时的建议是继
续做那个功能域和另外一个分子的共结晶。这本来是一个热点,近几年cns上相关的文
章不少,结果老板第一反应是觉得风险太大,不想花太多时间和精力又要冒被人scoop
的风险,他说基本上一两个月之内就要把文章送出去。当时我用了两个星期已经纯化出
了complex,不过要我在两个月之内搞出晶体再解了结构那是不可能的,而且觉得他小
心谨慎也是对的,于是就说好,我马上写吧。
结果初稿交给他后,他老人家磨磨蹭蹭用了半年才定稿把文章送出去。这半年里面,按
照他本来的说法,是要找我们的合作者做bio的部分。但是不知道怎么搞的,过了一个

有快20篇了。其中有cell, neuron, Journal of neuroscience, PNAS 好多篇。而且他
1995年就回国了,那个时候国内的环境有多差不用说了吧。这也快20年了。比饶益2007
年才全职回去多了12年。比起培养的中国学生也多多了,算国内贡献肯定不会小。如果
他一直留在国外,成就未必就比饶差。谁把饶益的文章列一列。国内多少篇,总共多少
篇,培养了多少中国学生?
