1, sucrose脱水过夜是不够的,我记得我是10%,20%,30%梯度脱的
好像有点多余,不过直接用30%的脱,中间也要换液。
2, 冷冻很讲究,乙醇干冰最好,实在不行液氮也凑合了,冻得不好就碎了
3, 起皱是贴片的功夫,这个得练,熟了就不起了
4, 本底高看你的操作可能是wash的不够,应该是摇床5min,三次
5, 掉片子是载片处理的不好。先泡酸,然后泡乙醇,最后泡明胶或防脱片剂 |

Because radioactive detection is very sensitive.
If you use cell-free in vitro translation kit, you will stick a lot of
ribosomes on the NTA-resin, and the unfinished peptides will show up as
smear on the gel.
What you need to do, is to use high speed centrifugation (at least 144,000
xg) on a 20% sucrose cushion to separate the ribosome fraction (go to bottom
) from the complete proteins (remain on the supernatant).
Then, you can use the NTA resin on the supernatant. |

without sucrose) following by gel filtration to remove free labeled AAs.
the problem is the pulldown assay signal is very low. I may try both NTA-Ni
pulldown and IP to see which condition give me a better result.

secondly, when I extract nuclear protein, I just use 250M sucrose in the
extraction buffer for nuclei and no further gradient centrifuge thereafter.

If you want to simply seperate the agregate from soluble protein,
you can buy those 10 cm disposable columns and pack the resin by yourself.
It is not too difficult.
Or, you can just do a ultracentrifugation with a sucrose cushion to seperate
the aggregate. |

透析会产生好多沉淀啊,但是直接稀释却不会。
