别？
sulfo 除了增加水溶性，能促进反应吗？ pH也要和NHS一样调到8吗？

发帖数: 1187 | 3 就是pierce的那个。想请教一下通常的yield有多高？第一次pilot，biotinylate后
跟streptavidin beads incubate，然后测bead上protein跟original input比，
只有10％，比预想低很多。biotin-avidin interaction应该不可能差。想到的只有
biotinylation本身出了问题。

发帖数: 1604 | 4 Pierce专门有测biotin labeling efficiency的kit，不过我嫌麻烦没买。
我做fluorophore labeling也是用它家的kit，反应的chemistry都差不多，那个的
efficiency不错，我就将就认为biotin label的也不错了。
biotin-streptavidin的binding很好的，我用biotin label的peptide和streptavidin
beads incubate，前后能相差80%。
还有一个可能，就是你的protein太多，beads太少，beads饱和了。 |

peptide you put in? that's actually lower than I thought hehe. How
did you get the result? measure the peptide concentration in supernatant
b4 & after incubation? could u pls let me know which kit you use?

efficiency:)

never really tested the efficiency (in fact now I think of it,
it'll be hard to measure how much free biotin is left... do you
just do multiple column purification to make sure of that?)
2. sorry can you elaborate on the elution part? I don't quite
understand the purpose for eluting after binding w/ beads. |

发帖数: 1604 | 8 嗯，就是incubation前后各取样测浓度。我是用Pierce的BCA assay kit做的，因为我
的peptide没有280吸收峰不能直接nanodrop测浓度。但是因为我样品少，用的是
microscale做的，所以incubation之后的浓度实际上已经很接近noise水平了,真正的
binding efficiency我感觉应该会超过80%的。
Streptavidin beads的instruction上应该有说它的binding capacity，一般用的标准
是mg biotin单抗/mg beads，你得用你的蛋白的size和单抗的size大概齐毛估估一下，
估计一个适当的蛋白浓度来做incubation，beads多一点没关系，蛋白用多了就可惜了。
嗯，不过beads也不便宜就是了。:)

关于protein-beads ratio，我labmate用的方法是：先用少量beads，
bind protein at highest coverage，这个batch用来做beads可能带来background
的实验。然后recycle supernatant，加过量beads，把能bind的protein都bind上。
感觉比较make sense，就是有点麻烦呵呵。
btw，以前用M-270 carboxylic beads，现在改用这个，发现capacity低很多，我
随便就要用几百个microliter。。。

发帖数: 1604 | 11 只要ratio对了，蛋白总是能bind完全的。我不觉得你会有90%的蛋白都label不上
biotin。这个biotin label是通过和蛋白的Lysine或者Arginine反应结合上去的，反应
效率很高。除非你的蛋白里根本没有或者很少K和R残基，否则应该考虑的是over-
labeling的问题而不是label不上的问题。 |
