sequential 曝光非常方便。Bio-rad 的software 做的最好。但是 syngene的camera
和镜头好。最后定了个syngene pxi6.

瑞士诺华公司 CAR-T 疗法 Tisagenlecleucel（CTL-019）上市。FDA 将在 10 月 3 日
前依据专家意见做出最终审批决定，这意味着 CTL-019 将大概率成为全球首个获批上
市的 CRA-T 疗法。
CAR-T 疗法的成功研制让癌症不再是不可治愈，是癌症治疗届的重大突破，意味着
癌症治疗的里程碑式发展。诺华 CTL-019 申请的适应症是复发或难治性儿童和年轻成
人急性淋巴细胞白血病（ALL）。此前 FDA 对外发布的审评文件显示，Study B2202 研
究入组的 88 例患者中有 63 例在美国完成的 CAR-T 制备和输注，其中 52 例实现应
答（CR 或 CRi），40 例在输注后的前 3 个月内实现完全缓解（观察到肿瘤消失）。
目前医疗行业去除癌细胞的方式主要为手术切除和放疗化疗，但都存在弊端：手术
切除存在很大的复发风险且受制于癌症时期；放射疗法和化学疗法在杀死癌细胞的同时
往往也会对大量杀死正常细胞，对患者造成很大不良反应。

我在文章上看到用129Sv老鼠做syngeneic model(把细胞打到老鼠里长肿瘤), 到NCI-
frederick买老鼠的网站上一看, 发现有129S1/SvImJ/Cr, 这可以用吗?

科普一下：
The cancer cells you injected carry allogeneic antigens, and these antigens
could be recognized by immune cells and will be quickly rejected, just like
the rejection of kidney in the transplantation. The number is not an issue
because the rejection for allogeneic antigens is very strong, even entire
kidney, liver etc.
If you injected your own tumor cells, that will be different issue (but it

发帖数: 1874 | 8 人是outbred的，所以人和人直接的MHC 差别很大的。实验室用的老鼠多为inbred，遗传
背景很纯，所以同系之间是属于syngenic，或者没有MHC差别，属于autologous。
我的理解。不知道对不对。 |

我用的骨髓是syngeneic，所用的irradiation的dosage是常规的80%，而且是分两次放
射的。另外老鼠骨髓移植后要喝四周的四环素水。另外我用一般的老鼠作recipient一
点问题都没有。所以真不知道是哪里出了问题。
我一般是转3 million BM cells/recipient。你们一般转多少细胞？ |
