我现在主要做代谢工程（不要脸的说，阿猫阿狗都做metabolic engineering,
synthetic biology）,这些circuit够fancy,但是我在想一个能够够好的调控metabolic
engineering里的基因表达的，要够fancy,有模型，有产量。
我总不能说，嘿，我的产物（酶）是oscillation的吧。
感觉吧，synthetic biology有点问题，又说不出来。 |

还有就是 研究的问题不通 使用的数学工具也不同，但是基本的数学工具还是应该有的
，analysis algebra probability
stochastic process 是最基本的吧， 至于要学什么数学知识，需要看遇到什么问题
吧，
而且，数学在生物学的研究里面 目前还只是 作为工具使用的阶段
我觉得 如果想进入一个领域 应该了解这个领域的目标和methodology 如果说这个
都没有搞清楚 就照葫芦画瓢 看到别人
用什么数学工具就跟风学什么 是永远也做不出有用的东西来的 不管怎样，你
做什么研究都是问题导向的 不会是我
要学哪些技术去做类似的研究，结果只会repeat 一堆 junk paper 出来
如果说严格意义上的synthetic biology 跟 bioengineering 没有特别大的区别，我个
人意见 认为它算是systems biology

LZ 是计算数学出身的，难道就不应该看一看 elowitz的文章么
noise in biology 这个可以连接或者引出好几个问题specificity, phenotypic
switching, bistability, bacteria persistence, pattern formation, evolution..
... 也跟可能跟 network theory 有关，
这些算不算biological science的大问题，莫非synthetic biology 不用去理会这些问
题？ 只是一心build networks？ build networks 或者 做 single molecule
imaging 不用考虑 specificity 和 noise么 不用考虑phenotypic switching？
不用考虑 pattern formation么？ 那你做的是不是 synthetic biology？ 还是
仅仅是 hypothetical network study？

不过估计lz看不上我们老板 毕竟发文章水平不高 和做的偏应用也有关系。我们主要
方向之一就是发展genome wide editing tool，然后结合robot-assisited high
throughput screening筛选一些高产或者高耐受性yeast strain 也做一些cell line和
stem cell 但毕竟不是做这个出身 主要靠合作
如果要回国的话lz可以考虑下 国内大部分synthetic biology center都是他帮忙组建
的 貌似国内的synthetic biology特偏应用 和metabolic engineering结合比较多
lz有兴趣的话 可以细聊

发帖数: 36 | 12 楼主感兴趣的话可以看看UCSF Jonathan Weissman实验室，听说他们实验室根据cas9搞
的cas9a（activation)和cas9i (repression)效果还不错，暂时主要应用在genomic
screen上。我觉得以后用来做真核的synthetic biology非常好。同样在UCSF 的
Wendell Lim前一阵子把一些基本的基因调控原件应用在免疫细胞里面，效果还不错，
以后基于synthetic biology的基因治疗可能也有很好的发展....
搬到mit的james colins 也很好。
纯从转录调控说，个人感觉e coli 和 yeast 差别不是特别大，从 yeast 到其他的真
和模式生物差别非常大。虽然yeast同样有histone,但是对转录调控的影响比较简单。
在yeast里大概影响转录调控的variables都了解的差不多了，直接manipulate这些
variables的工具也相对齐全。在其他真核系统里，定量的研究感觉上还都是停留在对
一个gene的特定promoter的研究，从这个gene上发现的规律放到其他gene上面基本... 阅读全帖 |

的表型，因为存在另一个基因，或通路什么的，可做功能补充。只有当两条通路都断掉
，个体马上灭掉。

发动机机油是由基础油和添加剂两部分组成。其中添加剂一般包括：粘度指数改进剂、
清净分散剂、抗磨剂、抗氧化剂、降凝剂、摩擦改进剂、防锈剂、抗泡剂、其他添加剂。
基础油分合成油与矿物油两种，合成油由原油中的瓦斯气或天然气分散出来乙烯、
丙烯后，再经聚合、催化等繁杂的化学反应炼制而成。矿物油则在原油提炼过程中，分
离出有用的轻物质（如航空油、汽油等）之后，利用残留在分馏塔内的塔底油提炼而成
的。就油的本质而言，矿物油用的是原油中较差的成份，提炼技术再精进也无法将其中
杂志清楚完全；而合成油则是原油中较好的成份，加以化学反应并通过人为控制，可以
达到预期的分子形态，分子排列整齐，抵抗外界变数的能力强，故合成油质量较好，在
