得到tubules的数量会最多，而且好切。 |

program（miranda，pita）但是结果都很大不同，而且用script run出来的和网页上现
成的也不一样。自己跑script 给出来的结果很多，不知道什么是合适的cut off。我主
要想比较sister species的difference，所以不想强调conservation。
不知道有没有大侠有没有经验，指点我一下，我没有任何mirna的经验，只是在狂读
paper。
还有就是想看gonad tissues的miRNA profile，好结合起来predict这几个基因是ovary
/testis-specific express) 上可能的functional targets。没有找到现成的文章，找
到了一些sra上的illumina data，modencode的还有没有发表文章吧。我只是相看看表
达水平，试了miRExpress，最后一步怎么都不没法让它work，其它的程序好像看起来很

首先，免疫系统需要区分Self and Non-self，也就是自身的各种物质不会激活免疫系
统，这里面涉及到在胸腺里面清除掉会攻击自身的免疫细胞，以及在血液和淋巴系统里
抑制能够攻击自身的免疫细胞，涉及许多机制，不详细讲。而肿瘤从根本上说属于Self
，因此人体对肿瘤的免疫反应，就具有先天性的弱势。
第二， 免疫系统有自动回调的默认机制，当免疫反应达到一定得强度，并持续一段时
间以后，免疫系统会自动回调，通过多种机制来抑制免疫反应，原因就是长期的高强度
免疫反应必然导致对自身组织器官的损伤，这是自身保护所必需的。
第三，肿瘤在形成过程中，成功逃避免疫系统的监控，并且充分利用免疫系统的各种回
调机制，形成一个强烈的抑制性环境，抑制针对肿瘤的免疫反应。因此在肿瘤病人体内
，肿瘤特异性的免疫反应基本是被抑制住了的，尤其是后期肿瘤，几乎所有在肿瘤中的
免疫细胞都是失去功能的。
那么免疫治疗的希望在那里呢？
首先，虽然由于免疫系统为了防止自我攻击，清除了大部分针对自身抗原的免疫细胞，

发帖数: 34 | 33 这两个文章结果不错，但是病人样本太小，病人可能都是精挑细选，更倾向对免疫疗法
有反应的（这个是根据以前一些一期试验的经验，没有依据）。个人对cancer vaccine
持怀疑态度，主要是因为：
1. 这个mutational load theory本身就很有瑕疵，如果看看几篇N Eng J Med的文章，
有疗效和没有疗效的患者mutational burden本身重叠部分就很大，加上现在很多被认
为是low mutational burden的肿瘤类型，对checkpoint反应率也没差太多。所以
antigen response 到了一定程度后，就是看microenvironment的抑制情况了。如果只
是作为maintenance therapy，那就回到20年前了，很难和新一代的制剂抗衡。
2. 即便是早期的cancer testis antigen vaccine, long peptide vaccine 也可以产
生类似于这个文章里的T 细胞反应，不过疗效在established tumour里微乎其微。这两

About 5 years ago, the alive sperm can be seen, although rarely, in my
husband's semen analysis. but last year, there is no alive sperm at all. So
either the alive sperms they detected 5 years ago are caused by
contaminations under the microscope, or the sperms-producing ability got
worse during these years.
Last year, my husband did the testis biopsy and found no sperm produced.
Your case is much better than ours. So you can try IVF, and at the same time
