什么吃太多都不好啊， 但钾相对钠好一点。 高钠是心脏病的一个重要因素。 但高
钾危害好像少点。 如果你家里的水是“硬水”， 你可能要用软水器， 其实是钾钠互
换， 建议不要换太多。

缺不缺乏也很难说。。。weruva的猫罐头和狗罐头实际是一样的，只是大小不同，但是

自己给肉吃的话，一定一定要确保补充钙和taurine，可以直接买supplement，也可以
分别用磨碎的骨头和心脏
生鱼不要吃太多，有thiaminase，容易造成thiamine deficiency
维生素B，E，和碘也要补充。但碘不能太多，容易甲亢
另外orijen有多贵？我家一袋wellness core三十多块钱，吃了半年还没吃完呢，我觉得纯吃干粮也可以顶俩月，比起罐头来每月七八十块钱真是便宜啊。。。我也是买罐头买的肉痛打算自己做猫食了。。 |

时咪宝还是能动的，就是非常的痛，所以我摸她后腿，尤其是关节，她还是会缩。小花
貌似右后腿完全没感觉了……
不知道现在如何。我没找到咪宝的全部用药记录，应该就是一般光谱抗生素。

我用的菌株是：B834（DE3）
培养基：M9+19个氨基酸+0.5%glycerol+ 5%glucose +Thiamine+1mM MgSO4+0.1mM
CaCl2+0.05mg/ml selenomethionine+amp
温度：37°
培养时间：24小时
用这样的方法，没4L菌可以大概拿到3毫克的蛋白。
现在问题是3毫克蛋白根本不够我用来长晶体。
问题：
1.在养细胞的时候我发现离心以后上清还是很浑浊，还有大量的菌在上清中无法被离下
来。我
加大了离心的速度和时间，还是无法离下来。有好多菌附着在离心管壁上。不知道这是
为什
么？
2.离下来的菌有一些黄黄的东西，而且是比较多的。通过我的纯化，我发现这些黄黄的
东西是
不能被detergent溶解的，里面没有我的目的蛋白。不知道这些黄黄的东西是什么？
3.离下来的菌在悬在buffer的时候，我发现很粘稠，和我用与表达非标记蛋白用到的菌
的时候
是完全不一样的。是不是菌破裂了，dna跑了出来。很奇怪。

Material: MTB3 and UptoDate. Step 1/Step2: 250/249 but those were 3 years

成绩出来了，涉险挤入了９９队伍。总的来说，准备的不是很充份，详见以前贴的＂我
的step ３经历＂，主要是在吃CK的老本；其他乏善可陈，成绩单里唯一的一个星号是
给了CCS，这里就说一说CCS好了。
以前总把CCS和CS混淆，其实这两个有一个共同点就是open choices和uncertainty。在
这总场合，最重要的可能就是要有一个严格的protocol，这样可以尽量避免遗漏。考过
CS的同学都知道LIQOR AAA和PAMHUGSFOSS的重要性。CCS也一样。网上比较流行的是
Neeraj的protocol，用google一搜就能找到。非常有用。我把他的protocol稍微整理了
一下，自以为是更加简洁有条理一些。给几个朋友用了用，有的朋友觉的没有时间应用
，有的朋友用了觉的还成，让我贴出来，最后的决定是如果考出来还行就贴，要不就不
出来吓人了。
象CS一样这个protocol充满了mnemonics，开始考试之前第一件事就是把所有的

culprit for CPM. Thiamine deficiency, for instance, in Alcoholics causes
Wernicke's encephalopathy but this patient did not have wernicke's lesions
in the brain, thanks to banana bags. |

我们这里icu常规高剂量thiamine up to 300mg tds iv for everyone 确诊的病例在普
通病房的也是这样给。
七十年代有一篇文章检查尸检病例发现大多数都有wernicke encephalopathy 病变但临
床没症状，也就没查vitamin b1所以其实有很多隐性病例，而这些人出现应激情况下或
大量用糖就会发病。
不过我倒没有想到hypothermia. 还有我会好奇MRI是不是完全正常，would be
interesting look hard in mamillary body and periventricular area... |
