is dominated by aptamer as affinity agent as the stability &
reusability is a big deal for sensors.
back to your questions, you can do thiol-gold, EDC-NHS, biotin-SA...
all you need is to synthesize the aptamer w/ desired functional group.
so are you doing arrays? or beads, like luminex? you may want to check
out larry gold's recent publication
I'd certainly like to, if it doesn't take too much time. any suggestions?

maleimide very efficiently. tri-maleimide is cheap. you have to synthesize
peptide anyway.
Just a reminder: the N-termini of proteins in vivo (bacteria or mammalian
cell) are complicated. The majority of them have the met cleaved upon
translation. Also the majority of them are N-terminally acetylated
cotranslationally or post-translationally.
Be careful! Just want to remind you not to use wrong peptides for Ab
generation. |

这些invitrogen卖的probe，我们都研究过，而且我也和invitrogen R&D联系过，
invitrogen他们承认他们的probe都不能在live cell里面定量，所以对我们的probe很
感兴趣。
我先讲一下为什么commercially available的probe不能很好的在live cell里面定量：
1.现在卖的所有的GSH或者thiol probe都是基于不可逆反应的，例如monochlorobimane
。一般按照invitrogen的protocol，加10-20 uM的probe和GSH反应，反应后，产物发出
荧光signal。这里的主要challenge是GSH的浓度非常高，一般是1-10 mM。如果是uM的
probe和mM的GSH反应，如果给足够的反应时间的话，不论你是1 mM GSH，还是10 mM
GSH，所有的probe都会被consume掉，所以得到同样的荧光output。
2. one can argue，如果GSH和probe反应足够慢的话，低浓度的GSH反应就会更慢，高
浓度GSH反应快，这样就能区分出GSH浓度... 阅读全帖 |

法是用pierce 的SMCC, 一端连抗体的amino group, 一端连thiol modified oligo；另
外一个方法是click reaction。用DBCO NHS 连上抗体，和Azide modified oligo 反应
。不知道哪种方法效率高些？
还有就是纯化，dialysis 好像找不到合适的MWCO。有什么好建议吗？oligo 大约15kD
，抗体150kD。

做label，这组实验我们是打算作为一种“indirect”验证方法的，因为我这个蛋白有
两个subunit，一共好几十个Cys，如果用手工的方法来找，真心太麻烦了，现在有不少
检查二硫键的软件可以使用，但是其accuracy和precision还真的有些难说。

做过磷酸化蛋白质组学的应该都知道，TiO2针对磷酸化基团具有很高的亲和性和特异性
，因此TiO2被广泛应用于富集磷酸化肽段的磷酸化蛋白质组研究，以分析磷酸化介导的
信号通路.TiO2富集磷酸化peptide的方法由我博后老板Martin R. Larsen 2005年发表
于MCP,引用有大约1500多次，相关专利被Thermo收购。
在本工作中，我们通过化学合成一种phosphanate tag,该tag可以特异性与Cys的free
thiol group反应，从而用于标记Cys peptide,由于tag含有phosphanate group, 从而
标记的Cys peptide可以用TiO2富集，原理类似于富集磷酸化peptide。更进一步，我们
的方法可以从同一样品同时富集cys peptides和Phosphopeptides, 从而可用于研究两
种PTM的相互作用the cross-talk between Cys PTMs and phosphorylation。 |

AuNPs的基团。所以估计这个ligand两端都可以连在AuNPs上，形成network，分散度会
很差，甚至连成一块。交换的话也难以避免这样的问题。
比较有效的办法是先找个别的反应将amino保护起来，然后attach ligand在AuNPs上，
随后将保护撤去，amino暴露出来，再进行后续反应。
一般在reduce HAuCl4时，为了保护thiol ligand的mercapto，要加一点acetic acid，
所以不存在你提到的问题。 |
