想在一个fungus里用CRISPR系统，非model system。
在该fungus中还没发现homologous recombination，so till now gene-knock-out is
not accessible.
current status:
1) Cas9 gets expressed by using GFP fused Cas9。
2)NLS works，在hyphae 中观察到 nuclear localization of GFP fused Cas9 and
NLS。 没有做western blot，因为GFP fuse 在Cas9 的 C末端，NLS 在Cas9的N末端，
这样理论上可以validate Cas9 能表达吧
2)U6 promoter，是个问题，由于该物种中U6 没有define，PCR克隆的U6 promoter不

内科第一年intern
从去年开始，traditional call system（每3、4、5天call一次）不符合现在的要求，
现在新规定出来后intern连续工作不能超过16or24小时（不确定）
自己所在program，有night float，从前天晚上（偶不确定是晚上还是早上。。。）六点到第二天早上七点半，每人每年轮一个月
不值night float的时候，就值白班
一、Intern之前的准备：
（一）Limited license：
根据所在州的决定，工作一、两年后，可以申请full license，正式去program上岗前，必须拿到limited license，最好在7月1日前， 有了这个之后，才能有以下的两个号：NPI（你的全国的编号）、DEA（你签名开药需要）
tele所在州board of medicine一个月开一次会，review一次你所交的资料
tele是七年制：毕业证书是五年的时候拿到，但在五年之前没有实习，是七年的后两年实习，类似这种情况，任何时候有discrepency，就要自己所在的医学院出具文件说明

transcription 的工作，总会找到合适的工作。选择跟着医生的那种。如果你没有
scribe的经验，不要紧。考版的经验足以让你成为还过得去的员工，选择在一个忙碌的
私人诊所开始，因为只有那里给你培训的机会，也给你工资，还有更重要的就是跟着医
生学习的宝贵机会。。。。十分锻炼人的。
我当时是选择了半职，干了1年，十分喜欢这个工作。并且打算如果没有买掉，就换个
医院继续做medical scribe。在面试的时候也被问到我的临床经验是交钱的嘛？ 我说
是我为它工作了，我需要收入维持我的住院医申请啊。。后来就买到这家医院了。 |

Pol II moves only in one direction on one strand. It can transcribe from
different strands – if the sense transcript is produced from the top strand

当有限的经验来评Android十大必不可少的app，GVoice当占头三名。Tech geeks和
Google员工可以略过……
一开始用主要是好玩，可以选号。很快发现这个对我没啥意义，我最恨的就是换电话号
码。然而很快就到了要换电话号码的时候了。打算抛弃我用了五年的老smartphone，上
Android。换号貌似成了麻烦：ATT网站上check portability的时候说，你这个号不是
同一个metro area的，换起来有困难……我相信肯定还是能port的，只不过看到这一条
我开始有点觉得麻烦。研究以后发现，貌似可以把号转到Google Voice上，我想，那就
先拿它做个过渡吧，反正这个号转到google以后就丢不了了。花了20刀转号（这是我唯
一不满的……）。
转完以后开始研究Google Voice app，发现了蛮多的好处，下面列一下我知道的：
1. 永远不用再转号了。为啥？接着往下看。
