要想美观， 那就RAISE BED， 当然前期投入大，但为了美，花钱是值得的。 这道很适
合我家情况，我们家菜地一开始没选好，有早晨阳光，中午的时候正在树荫下，现在沦
落成孩子的试验田，孩子也不太去，因为菜长不太好。 比如下面，好看是好看，就别
指望出菜供全家吃了。

要想吃菜，就要WIDE BED（起垄）， 好看在其次， 省钱出菜。这个是适合地大的人家
。我觉得游学士家菜地就类似下面的图， 当然游大的草地要漂亮得多还种了好多的菜
：
我理解的对么？ 谁家有那种省钱，省事情，还美丽好看的菜地吗?
加州大家的后园都不大， 好像RAISED BED更适合我们。而且买菜太便宜了，种东西就
是个爱好，不指望吃。 看我周末买的： 葱一刀4把， 韭菜一刀5把，姜一刀两磅， 黄
金瓜白瓜西瓜都是一刀以下一磅， 50刀买一大堆菜啊牛肉啊。 我真觉得自己种菜太亏

发帖数: 1627 | 9 我也用WIDE GAMUT，自己试出一套流程，可以不留副本，不减饱和，输出后在其他显示
器上观看，和在自己机器上基本一样。中间步骤可能有不对的地方，但是我自己觉得方
便好用。
先用SPYDER校正显示器，得到ICC文件A，在WINDOWS的色彩设置里，设置A为显示器默认
色彩配置文件。
如果要先用LR修RAW，则在启动LR之前，用SPYDER带的那个色彩文件管理软件，选择
SRGB，这时桌面显示颜色会不正常，无视。 启动LR。 等LR启动结束后，发现LR内图片
显示也不正常，继续无视。 再用SPYDER带的色彩管理软件，选择回色彩文件A。 这时
就会发现LR和桌面的颜色都正常，可以干活了。
从LR直接导入到PS，扔掉色彩管理（提示色彩配置文件不匹配的那个对话框里选第三个
）。这样在PS里的颜色和LR里一样，显示正常。
PS结束后，最后一步ASSIGN COLOR PROFILE，选SRGB。 指定好后，会发现PS里图片颜
色不正常，无视。保存文件，同时记得指定用SRGB色彩空间。
这样保存出来的文件，在其他人的机器上显示正常，在我的机器上，WINDOWS和ACDSEE

，但是看片子或者电影或者任何东西就是过于饱和。
无奈把显示器设为模拟sRGB模式，然后再calibrate，gamma变成2.5而不是正常的2.2，
但是品片子颜色正常了。
还有，LR或者NX2下面颜色看上去还是过饱和，但在picasa下看或者上传到网上以后就
正常了。
我对色彩管理一窍不通…… |

there may be some spacing problems at page breaks.
You also lose some float functionality because of the use
of the multicol package. See the multicol documentation
for details. The definitions between \usepackage{amsmath}

loaded using \usepackage{...}.

对这个很感兴趣。读了几篇相关的文章和screening,前面的library--infection--drug
screening--genomic DNA extraction都懂。后面的deep sequencing部分就糊涂了。
理论上讲，resistant的细胞一定是有基因改变了的，我一直以为挑单克隆，一个一个
去作整个genome测序。仔细读了文章，是药物筛选几天后，细胞pool一起，再作deep
sequence；对照没有drug筛选（Non-resistant)的组，找出那些被CRISPR改变了的gene
，再确认。
几个问题：
1。这个deep sequencing是测（整个）genomic DNA，还是测留下来的guide sequence
（在lentiviral vector上面）？
2。可以测mRNA/cDNA序列来代替genomic DNA吗？突变的基因总该表现在mRNA上（先不
考虑其他类型的RNA）。
