用的饵是plastic minnow 和berkley gulp,rock bass都是看着上钩的,想用senko钓
大bass结果甩到手抽筋也没鱼咬钩,还试了一些其他的,都没bite。下午订到了
camping site,转战到湖南边沿路的石头堆里钓,结果试了两三个小时,没有一个鱼咬
钩,旁边的一个老头,用很大minnow,大概2-3”,钓了一些和我上午钓的差不多大
小的bass,而且他说他在那儿钓了35年,每天都去钓,说的很严肃,不像开玩笑,还
让我试他的活minnow,我谢绝了,觉得没什么意思,用那么大的minnow就拉些比minnow

看了一下这几篇文章,很有可能NEDD4不是PTEN的E3,但是那篇CELL paper的方法和数
据又没有什么问题?jiang是从xiaodong wang实验室出来的,他从in vitro assay的建立
,到寻找组分的方法,和 xiaodong wang的一摸一样,但是为什么最后找到的蛋白为什
么不对呢?我感觉是in vitro assay的问题,虽然E3有很好的substrate specifictiy
,但是overexpressed 或者是in vitro assay的时候就不是这样了,过表达的情况下就
没有底物特异性了,所以在他们的assay 中能看到PTEN ubiquitination.后面可能选择
了符合预期的实验结果。一些相互作用和WB,多做几次总能出现一次想要的。junjie
chen的文章提到nedd4和pten没有相互作用,可以解释为实验条件不一样,比如 buffer
的离子强度。
我也是做生化的,对一些生化的结果总是很困惑。比如做个screening,曾经做过

我不同意楼主的观点...wang xiaodong绝对是凋亡领域最重要的巨人之一。
但是我的观点是如果凋亡给炸药奖,小东得概率未必最高,另外这个领域我也不看好给
炸药奖,因为2002年给了horvtitz,虽然那年颁奖思路很混乱,三个人都是做线虫得,
但是有做基因组测序得,凋亡顺着给了。
这个noble一给出来,kerr, xiaodong,korsmeyer,vaux估计就知道没有希望了...
小东在凋亡领域得贡献大家都知道,但是给炸药的话不好说就稳给小东...nobel那帮评
奖老头经常犯迷糊,但是如果按诺奖得一贯注重最早的原创文章/工作的话,我觉得
xiao dong显然得不到。
另外我对小东促进国内科学发展和研究很是佩服,相比某人的高调和某人貌似低调但是

你是黑我呢吧..我一吊丝哪里摸过普林的门框...
xiao dong 自己的独门生化绝技+眼光/运气在合适的时期进入了细胞凋亡这个领域(
原来的领域SREBP已经很牛了,但是那个领域想出更大的突破很难了,毕竟薄后祖师爷
压着呢),体外系统对于整个领域对于凋亡的机制理解有很大的促进作用。
但是我个人对于把xiao dong吹上天什么诺奖囊中之物/必然会得之类的言论觉得比较
好笑,因为这种话大多都是国人口中出来得。这个相关领域贡献跟xiaodong差不多的最
起码有好几位,诺奖最多给三个,这个谁能保证xiaodong 一定得呢..
“国内90%的课题组做无用功,国际上根本无人知道,这个也有小东的很大的责任吧。”
--这个我不知道你的证据和结论都是哪里来的...国内研究水平水涨船高,也作出了
不少国际知名得成果。
你要是非较真说诺奖级别的才算“成功”,那国内现在达到这个水准得确实没有,但是
罗马不是一日建成得,总有个过程对吧?

这又是咱们华人在mitochondrial领域具有巨大贡献的例子~
Yufeng 通过观察现象和深厚的知识背景首先意识到mitochondrial和apoptosis可能的
潜在关系;
Guido Kroemer观察到mitochondria会在细胞凋亡前失活;
Don Newmeyer在cell-free system 发现mitochondria的作用。
而真正在细胞上最早把Cytochrome c释放和激活capsase关联起来直接证明线粒体参与
凋亡的还是Xiaodong同志以及后来的Ruth Kluck:(一个Cell,一个science,引用都
是4000+)

年不间断)的研究,持续推动整个领域的发展。比如brian kobilka是研究心脏病,比如
xiaodong wang是研究程序化死亡,什么生化,结构,细胞都是手段,都是为了解决具
体问题所用的方法,既可以为解决问题在实验室里建立这些方法(比如roderick
mackinnon和brian kobilka),也可以找人合作(比如xiaodong wang),但最重要的是解
决生物学问题,持续推动一个重要生物学问题/领域的发展。当然有些大牛也可以从技
术入手,那么目标就是在技术领域在方法学上突破性的创新,比如谢晓亮, 比如Lee
Hood。
拿着一门做熟的手艺,东打一锤,西来一棒,什么热做什么,发的文章是不错,但一
直都是跟在人家屁股后面。人家说这重要就做这个,人家说那个重要就做那个,没有自
己的生物学问题,没有自己建立或持续推动深入研究的领域,也没有在自己擅长的技术
上划时代的创新,还敢叫大牛?大牛是要第一个告诉别人什么重要,而不是知道了什么
重要,就做什么。

“信息”“润色”的畸形怪胎。一般以所谓的Meta分析或者随便下载一套狗屎基因芯片
数据,然后做一些似是而非的烂分析,发一堆垃圾,一篇文章卖给忙于召妓的医生5万
到10万。
