i think it depends on what kit you use...
sometimes, the reader you use to read ELISA plate comes with some kind of
software (logistic regression)
if not, just read at XXXnm, minus YYYnm (background),
get the raw data (X-Y),
substract the blank,
and calculate in excel manually,
比如,把Standard的濃度和讀數都取對數(如果你做LOGISTIC回歸)
把UNKNOWN的讀數也取對數,再用EXCEL的FORECAST功能得到UNKNOWN的濃度
再把濃度“反對數”……

you may want to test linear range for your signal first.

compare them w/ your sample.
if need to be accurate, may need to consider: 1. the linear range;
2. incubation time;
3. sample condition;
etc
you can also use scatchard plot to get linear curves and use ANOVA
