M****e 发帖数: 178 | 1 Hello,
I have a quick question about the terms used in the field of analytical
chemistry. Any input is really appreciated.
We are trying to determine whether a patient has taken a certain drug (
compliance) by examining the existence of drug metabolite in patient's
plasma via mass spectrometry. To evaluate how accurately this MS procedure
predict patient's compliance, we treat MS as an assay and patient's
compliance as gold standard to calculate sensitivity and specificity. But
reviewers said 'sensitivity' and 'specificity' are not proper terms used in
this field. I just wonder if there are some other quantity/term to measure
the procedure performance in this case.
Thanks! |

It's a manuscript "An LC-MS assay for the semi-quantitative screening of
cardiovascular medications in human serum", submitted to Journal of
Chromatography A.
One of the reviewers gave the comments:
If the manuscript should be published in an analytical journal some
modifications have to be done. Sensitivity, specificity and accuracy are not
used as typical in analytical terms. Please rephrase.
I'm clueless about what the reviewer meant to say.

go get several just publish paper in that paper which use these terms.
go find FDA guidance for industry for bioanalytical method validation, use
it as reference
not

t******t 发帖数: 3045 | 9 In your case, sensitivity is the ability of your assay to tell "positive"
from "negative" and specificity is the confidence you have on your "positive
" to be a "true positive". Explain it to the reviewers if they do not
understand.

specificity' themselves. Even the definitions from Wikipedia will do the
work. Thanks for the nice suggestion!

t*******h 发帖数: 51 | 17 this sounds correct. BTW, not LOL, should be LOD, limit of detection.
Sensitivity and specificiy have very different defination in othet fields.
especially, you were talking about patients, should check out the exact
definitation in medical statistics. google ROC curve.
In the field of sensor, transducer and signal processing, sensitivity is
defined as the first derivative of the transfer function (output readout vs
input signal).

M****e 发帖数: 178 | 19 I think this is the point. We certainly use these terms as statistical
measures of performance, and we mentioned this in Statistical Analysis part.
I guess I need to somehow highlight the definitions... Thanks!
