Title: Small regulatory RNAs regulate the biocontrol property of Pseudomonas
fluorescens strain Psd
ABSTRACT:
Abstract
The production of biocontrol factors by many pseudomonads is reported to be
controlled at post-
transcriptional level by the GacS/GacA signal transduction pathway. This
involves RNA- binding translation repressor proteins, RsmA and RsmE, and the
small regulatory RNA(sRNAs) RsmX, RsmY and RsmZ .While the former represses
some of the genes involved in secondary metabolites production, the latter
relievs this repression at the end of exponential growth. We have studied
Pseudomonas fluorescens strain Psd, possessing good biocontrol and plant
growth promoting properties, and confirmed the presence of rsmY and rsmZ
genes by PCR amplification. Gene constructs for all the three small RNAs
carried on broad host-range plasmid, pME 6032 were transformed into strain
Psd. The recombinant strains, Psd-pME7320, pME6359 and- pME6918 were found
to produce comparatively higher levels of phenazine (PCA), IAA to some
extent, and strain PsdpME6918 also showed slightly greater amount of HCN.
The tomato seedlings infected with Fusarium and Verticillum in the presence
of culture filtrate of
these strains showed better plant protection response as compared to that
from strain Psd. The results obtained so far suggested that in Ps.
fluorescens strain Psd, regulatory RNAs play an important role in regulating
