发帖数: 1312 | 1 我也没做过，想想就觉得不容易。RNA-seq大多数基因都有明确的boundary，很好统计
每个gene/transcript的count。但是ChIP-seq就太复杂了。是按照每个binding site在
不同条件下来比较呢？还是调节某个基因的所有binding sites来比较？后者不但要考
虑gene length，还要考虑如何定义哪些binding sites是调节哪个基因的，这个现有知
识估计还不完全吧。
简单查了一下，目前differential binding analysis（我之前叫DE其实不对），几乎
都是比较单个的binding site/peak的count差异（如果是这样的话我觉得应该不用考虑
基因长度的影响）。下边篇文章比较了十几个ChIP-seq differential analysis的软件
，简单的提到了gene ontology，找nearest gene。所以应该还是可以做GO的。里边也
提到length of differential region, 不过我就没有细看了。希望能有点帮助。

电话。但是现在我从sipgate的另外一个号码call这个650XXXXX2的电话得到如下的log
，给人的感觉就是进了outbound的了，好奇怪；看了一下externsion。conf，应该是
come in phone call会自动dial default的呀？
其他的都很正常：从1001往外打电话，接gtalk来的电话，但是没有办法接从sipgate过
来的电话。

来请教一下。
我现在只有一副死沉的downhill ski，以前几次都是穿着climbing boots，然后把
downhill ski+boots背上山去滑下来。
上课再这么干估计希望不大，估计还是得买新东西。
简单看了一下，这个telemark和randonee（又称AT ski）上山的时候都是脚后跟可以抬
起来的，区别在于下山的时候telemark仍然是脚后跟可以抬起来，转弯的时候一跪一跪
的那种，而randonee可以下山的时候锁住脚跟，就跟一般的downhill ski没太大区别了
。这个理解是不是正确？
现在考虑下来，有这么几种方案，请指教。当然钱花得越少越好，穷人嘛。
1、老一套，还是象以前一样把整个downhill ski/boots啥的都背上去，这个最省钱，
啥都不用买，（可能需要snowshoe）。不过太重了，估计instructor看着也不爽。

全了朋友半天他还是买了，让他别买burton bindings，可最后图便宜还买了个est的
bindings
（120刀），，这下以后只能买burton的板了！

发帖数: 97 | 24 不要灰心, snowboard是不符合人的自然运动习惯的(身体正面向前v.s.侧面向前),需要
时间适应调整.这也是一个原因为什么snowboard刚开始出现的时候,一般的雪场是禁止
的,ski only. 我在摸索中得到一些体会, 和你分享一下(高手pls correct me if I'm
wrong).
1.鞋带一定要系紧, 非常非常紧,目的是保证身体stable. BOA钢丝鞋带系统就是为此原
因发明的.
2.binding的strap要系对, 前面那根是紧在toe那里还是在脚背要弄对. 我的新burton
custom binding是紧toe那里的, 被我弄在脚背, kirkwood的coach眼尖指出来了.
3.binding angle. duck stance比forward stance容易保持身体平衡, 15/-15或者18/-
6. 这个要自己试. forward stance不适合新手.
4.binding在板上的位置用centered,不要off center
5.身体的姿势: 弯腿不弯腰(从我乒乓coach那里学来的).膝盖一定要弯下来, 降低身体

，也是盡量把板子打平不讓自己滑下來
穿鞋的話還是要看不同的snowboard bindings
一小部份牌子的bindings腳後跟那塊是可以解下來平攤在地上的，我的那個就是，所以
我平時前面的bindings是固定的，要脫的話就是把後面的扣子解開。要穿的話套進去就
行了，再把後面的扣上。唯一不方便的是，既然要把後面放平，我穿還是脫都必須是要
站著呢，所以我一般是站著扣後面的扣子。在山坡上也是用推土機方式站著穿，可能边
穿边滑一點，不過穿的很快～所以我還蠻喜歡我的bindings

boots卡上去后，只有左右扭和往前使劲倒时才会分离

expert水平。当年的新雪板也旧了。想在雪季来到之前换付新的。现在是淘deal的好时

对于我来说，滑雪是主要带儿子滑，有空的时候练练mogul，滑滑powder。我身高1.73m
,体重161 lb，现在的雪板长1.65m。如果选1.63m的板子会不会嫌短？

1.上面大家的讨论都是基于SQL Server吗？
2.rebuild index为什么不需要update stats呢？我理解的rebuild index只是重建索引
结构，对stats啥影响。即使rebuild index有一个默认的update stats选项，我感觉应
该只是update index关联列的stats。其他非索引列的stats还需要update.
导入数据后是否update stats，我认为要case by case分析。一个要考虑有没有足够长
的维护窗口来做一次足够强度的采样，二是考虑业务需求。有些业务是锁定统计数据的
，比方说有的表尺寸变化比较剧烈，这样的表不适合定期更新统计数据，需要锁定统计
数据防止在不合适的时间点做统计收集。
下面说Oracle。
Oracle也有根据数据变化百分比的threshold来触发统计数据收集作业。但是仍然导致

。Angular和jquery不兼容的。我看了看Angularui-bootstrap，就是把bootstrap-ui用
Angular重新实现了一遍。Angular连直接bind to DOM都是要禁止的。要bind to DOM，
就要使用ng-bind，否则你不能trigger two-way binding。jQuery是基于event
listener的，而Angular是基于dirty checking的，不过Angular2.0要使用Observer,这

JQuery 做 DOM 操作的语法糖.
因为太基本了, 用的人太多了,
所以我觉得即使以后 ES6 了,
整个 community 也能想法继续兼容它们.
有人说 ES6 以后 data binding 好了, 不需要 JQuery 这种,
作为业余前端, 我一直没搞懂,
因为 DOM操作 是比 data binding 更低级更强的抽象,

React 回到了以Javascript为中心看待问题，HTML 只是用于解决界面问题。
很显然，以Javascript 为中心更符合看待问题本质，尤其对码农来说。
但是回到现实世界，HTML 这种 declarative 看问题的方式更符合人类这么多年描述事
情。
Two Way Binding 是Angular1 年代的一个 东西。Angular2 里面本质就两种单向
Binding。一个是，UI的某个属性一直等于某个变量。另一个是，UI 的某个事件输出到
某个变量。
为了达到Two Way Binding的类似的效果，可以同时标示这两种。也可以用一个新发明
的Syntax ［（）］。

Proteins and column both have charges.
Proteins can bind to the column by electrostatic interaction (so-called
non-specific binding). Of course this is not what you expect.
Assume Protein 1 is what you want, protein 2 is impurity.
If there is sodium chloride in the solution, sodium or chloride ion can
compete
with proteins to bind to column. So protein 2 will be removed from the column.

easily predicted with its structure. For example, if it bind to some samll
ligands, then there should be a binding pocket. Or if binds with DNA/RNA(?),
then there should be highly charged part on the surface.. Protein-potein
binding sites are more difficult to be predicted.
There should be other info. to be used. If you get a protein structure right,

1. 直接binding，yeast-two hybrid，或通过complex中找到。这些报道很少，一般
是找到后用来确证的确直接binding。
2. 根据已知的substrate中磷酸化位点序列的保守性来scan，推测，猜。
3. 猜：两个蛋白功能很接近，看是否直接binding，是否磷酸化。
4. 用peptide array：体外纯化的kinase，看对哪些peptide的磷酸化活性高，根据
peptide的序列scan，再体内验证。
5. 2-D胶。敲除某个kinase，看哪些蛋白的磷酸化发生变化。再质谱鉴定。
6. inhibitor array：现在有很多专门针对某个kinase的小分子inhibitor，把这些
inhibitor收齐全了，如果有某个substrate的磷酸化抗体，就用inhibitor筛一遍，看
哪个抑制强。

但是对于probe 200bp应该是可以的，我用的是400bp，因为我不太确定具体的binding
位点。另外，如果probe的条带和加蛋白以后一样只能说明没有bind的吧。我刚开始也
一直找不到binding的条带，后来配了几种不同的buffer，发现有一种效果好一点。这
个binding reaction的条件可能需要自己试一下，怎么样最合适。我用的是RNA，操作
过程中很容易降解，所以到现在也没一张我老板满意的picture。

用一段PROMOTER 序列（200bp) 做probe, 能binding 到体外转录的蛋白上，但背景比
较高然后用两个片段来做probe, 有一段能binding, 。奇怪的是，当把这个片段再弄成
两段（直接合成PRIMER, ANNEAL)就没有binding了，难道是这个binding 需要二级结构

发帖数: 903 | 49 在revision，review建议endogenous的coip检验是否binding，表明真实的相互作用状态，我们文章里面，overexpression可以检测到binding，但内源的试了几次，摸索了不同buffer条件没有成功，以前就看相关文献，一篇cell里面可以内源检测到该蛋白和一个底物binding，但我们没有重复出来，就连overexpression这2个蛋白，我们也没检测到binding，哎，十分frustrated啊，coip就是很tricky。 |
