一类系统的共性的，才能够挂 systems 的名头。
不过么，总是有人把 systematic analysis of a biosystem 挂上 systems biology
的名头，而且用得还挺多。正如你所说的啊，SysBio 现在就是一个筐，一个名头，做
systems biology 的主流之一，还是那帮做 computational biology 的，很多对
systems science 基本不了解。

全的很。
有人提到这里面system biology很强，其实不是一般的强。感觉美国这边做system
biology的也有不少Tel毕业的.(犹太美女给人印象最深，09，10，11发了不少牛文)
最近粗略看了几篇关于network modeling的文章，看的不是很懂，大致就是一些flux
balance analysis 还有bernhard palsson的那本新书。
虽然不是很懂，但感觉也不像是有人说的一个大坑，只是还处在初级阶段而已。
但是我也不是很清楚目前能解决什么实际问题，bernhard在书的开头说，可以提出
model指导实验，感觉这个很玄乎。
来版上向各位请教下，system biology未来都能干些啥。 |

什么时候能够根据各种数据Infer network from nuclear gene expression to
cytosolic metabolism 那就算是开始有用了。
找biomarker貌似用不着system biology吧，proteomics or microarray就能提供一些
信息了。
颜色，轮廓等这些数据我觉得都是一些浅层的数据，如果能profile细胞内各种代谢物
浓度等等啥的就好了。

science 一作文章的LG，坚持转行，把解决身份的事推给我。 不是我不想挑这个担

publication，citation就那么低。 就算是简单的文章review，快一年了，才review了

我是phd第四年，现在有一篇S是minor revision，还有一篇质量应该还可以的文章在写
。现在想的是找到合适的postdoc后年底或者明年毕业。 另外我个人情况是想做三年
postdoc后回国，这个是确定的（我不想在美国待下去了）。
我之前做的是e. coli里的synthetic biology, 如果继续自己phd方向或者相关方向，
是有几个可选实验室的，导师也有几个推荐，这个好处是轻车熟路，文章可能容易发（
我也有几个有一定挑战性但也十分可行的想法）。但是我想自己是否应该进入一个新的
领域进行尝试，刚和导师谈话，他也是这么建议，但是我感兴趣的方向，他所了解的情
况不是很多。 所以来这里问问大家，也想听下过来人对于postdoc的意见。
来这里发帖想咨询下关于synthetic biology在真核系统中的进展情况，以及平台是否
成熟。比如在E.coli里面，基因表达在特定情况下已经是高度可控制的了。从promoter
到5’UTR, RBS，蛋白起始序列，到最后的terminator.都有很多研究如何控制这些序列

不过估计lz看不上我们老板 毕竟发文章水平不高 和做的偏应用也有关系。我们主要
方向之一就是发展genome wide editing tool，然后结合robot-assisited high
throughput screening筛选一些高产或者高耐受性yeast strain 也做一些cell line和
stem cell 但毕竟不是做这个出身 主要靠合作
如果要回国的话lz可以考虑下 国内大部分synthetic biology center都是他帮忙组建
的 貌似国内的synthetic biology特偏应用 和metabolic engineering结合比较多
lz有兴趣的话 可以细聊

发帖数: 1 | 37 最近做luciferase assay，就是把含有疾病相关的snp的promoter跟luciferase连接在
一起，看snp对promoter的影响。假设snp的ref allele是A, 疾病相关的allele是B
我首先在hek293里做，用lipofectamine3000转染。铺板子（12-well）铺了10个孔，5
个用于allele-a, 5个用于allele－b。先把lipo和dna混合好，然后把mix分给每一个孔。
以上是第一次实验，过两天我会一模一样的重复上述实验。
我想请问这里的5个孔中的每一个孔算是biological replicate还是technical
replicate呢？
而这两次实验算是biological replicate还是technical replicate呢？
之所以问这个问题是因为后来面对转染neuron的实验。neuron比较难转染所以用的是
nucleofection，一次性用cuvette转染100万个细胞，然后分到12-well的3个孔中。
同样的实验过两天我也会一模一样的再重复一次。

biological replicate，实验本身也不需要biological replicate。
在neuron中做的话，如果只有一个来源的细胞系，也是只需要technical replicate即
可；如果在不同细胞系或者原代细胞中重复的话，才叫biological replicate吧？ |

需要你纠结的时候，就说明这种提法不够准确了。
其实考虑这些问题非常的直接，你的统计学结论的scope是什么。你一次
铺5个，最后能做出的结论就是“9月12日我的实验结果说明a和b不一样“；
你第二天再搞一次，结论就是”9月12日和13日的实验结果说明a和b不一样“。
其实你想要的结论是“a和b不一样”，你所有的实验都没法推出这个结果，
你从前面两个实验的结果来推，这叫做extrapolation. 要注意，extrapolation
都是不靠谱的，当然你做两次试验比只做一次相对靠谱一点。至于啥biological/
technical都没啥区别，你第一次的实验想对mix好了测5次也算是biological，
你第二次试验相对于重新买一批hek293重做也算technical, 程度不同而已

than 10 questions on molecular biology, even 5. Even if I did lab work, my
knowledge about molecular biology is really poor. Even if I spent 5-10 mins
on one molecular biology question I met in step 1, I do not think I got it
right. I even did not have more than 10 questions in pharmacology, or
biochemistry.

对于生物版所争论的版务管理及讨论话题，几点意见：
1、话题讨论自由，人身攻击严禁：不建议限制话题，无论退或不退，均可讨论，但拒绝人身攻击行为；
2、版主可视情况对版面话题进行适度引导，尽量维护与维持版面的讨论方向；
3、对好的话题、好的帖子，版主可多鼓励并及时加入精华区；
针对以上原则，仍倾向于原版主Oncogene继续留任Biology版版主一职。
考虑到Biology话题较多，讨论也多，两个版主（Oncogene、pigskin）可能精力有限，版主可以再多添加2名版副，协助处理版务及整理精华区。
望同样有兴趣并愿意为Biology服务的ID与版主联络，申请版副一职。谢谢！

投诉理由及证据:
1， 封人理由是“干扰版面讨论秩序 行为”，难道该版猪不同意的意见就是干扰版面？
lllleaf 发文抱怨demoner无故删不同意见的贴子，由于前几天在我身上也发生了同样
的事情，我一发不同意见的帖子就被demoner删掉，我跟lllleaf的贴子表示同意，我认
为要允许不同的意见发表，该人立刻封我3天。这是滥用权力。
2，在该版猪上任后，biology版的讨论越来越没有深度，与广大生物人关心的职业前途
问题讨论越来越少，我认为与demoner滥用权力删贴有关，因为以前这种情况很少，前
后对比差异显著。 |
