做TF的CHIP本身就是很难。你的TFbinding确实很低，所以你需要做一个IgG control,
或者NO AB CONTROL。
还有你现在这种比较方法不是很科学，因为你在比较2个SAMPLE出来的DNA。 每个
SAMPLE 的handling都会造成很大的误差，所以你需要normalize到一个control DNA
region.
1/9*1/(2^6)
which is extremely low.
这个数值你现在show的其实没有任何意义，你这个数值如果跟SAMPLE B比较，其实很高
，但是不能证明这个TF就bind这个地方。
你需要screen其他的region在SAMPLE A里面, 如果你其他的region出来的都是1/9*1/(2
^6) 这个值，那么说明你这个值没有任何意义。
CHIP绝对值的YIELD比较没有什么意义，不同人做CHIP的方法不同，你可以用histone
H3 antibody做个CHIP，看看yield。

面粉做的，和一般店里卖的玉米面chips不一样，但蘸了酱吃起来味道也很好。而且可
以自己控制油和盐的量，比较健康。
tortilla chips
原料：4cups面粉，1tsp盐，2tsp泡打粉,4tsp橄榄油。1cup热水
1. 将除水意外的原料混合，然后逐渐加入水，揉成一个面团。醒15分钟。
2. 将面团分成20个小球。每个球擀成一个薄饼。
3. 将薄饼放平底锅上用中火烙，每面30秒钟的样子，等泡打粉冒起来一点小泡泡，并稍
微有一点点焦痕即可。这就是tortilla了。
4. 将将烙好的tortilla两面抹上一点点橄榄油，若干个叠一块十字切开成成90角扇形。
也可以切成条状。
5. 烤箱预热400度，把切好的tortilla片平铺好放进去烤。大约10分钟的样子。等

快变成浅棕色的时候立即取出。这个时间最难掌握，等烤到六七分钟的时候就得不断观
察。火候未到的话烤出来的chips不脆，而chips变脆到烤焦也就是几秒之间。

用鼠单抗，似乎非特异性的binding特别多，Negative control 用鼠的IgG也一样，有
非特异性binding。
但是同样的系统，Rabbit的抗体，和Rabbit的IgG就没有任何问题。
希望高手能指教一二。
看了cell signaling的ChIP grade抗体，无论是鼠抗还是兔抗，都是用Normal Rabbit
IgG，如果这样都可以的话，任何位点的chip，我都可以做得到了。
看了一些paper，例如用beta－catenin （BD 鼠抗），也是用Normal Rabbit IgG，这
个似乎就不合理了。另外，我看了同一篇paper里面的ChIP－seq Sample prep，作者说
能chip下来350ng的DNA。我也做了大概五六十个chip－seq的样品了，好的抗体也就能
chip下来10ng左右。
是不是protein G有问题啊？ |

想请教大家为了减免生产时除开自己商业保险以外仍有的2-3k的费用，可以（应该）去
申请chip perinatal或者 chip perinatal-emergency medical services
certification (Form H3038-P)吗？
chip perinatal 和 chip perinatal-emergency medical services有什么区别？两个
都能cover生产时医院的费用吗？
这种补助会如何影响以后带宝宝进出美国（海关查问）以及申绿卡呢？

跟大家请教一个CHIP跟绿卡的问题。就是申请CHIP会不会对以后的绿卡申请有不好影响
，如果有的话，影响有多大？
我现在刚工作不久，还在OPT，公司保险不太好，跟老婆计划生小孩子，跟保险代理打
电话咨询后，说我这个保险，生孩子的话至少自己掏3000，最多5000。另外老婆的保险
每个月自己要掏200多。
所以想计划在保险renew的时候让老婆退出保险，然后申请这个CHIP，CHIP是类似于
MedicalAid的东西，是针对没有social security number的人的。
由于我收入一年在40000左右，所以是qualify这个CHIP的，只需要每个月交50块钱就行
了。生孩子基本就不用自己掏其他的钱了。
大家觉得我要是申请这个CHIP的话会不会对以后的绿卡申请有不良影响啊？如果有的话
，大概会有啥影响呢？

先说为什么要有这个CHIP. AF CONFIRM SIGNAL 本来是机身对焦系统的功能, 根本不需
要镜头给个什么信号. NIKON/PENTAX 机身装没有CPU 的镜头仍然有FOCUC
CONFRIMATION. 只有CANON/MINOLTA/SONY 是故意把这SIGNAL DISABLE 掉, 除非机身探
测到有CPU 的镜头装着.
这CHIP 做的就是骗机身说有个50MM, 光圈F1.4固定不变的镜头装上了, 机身就给了这
信号.
FOCUS CONFIRMATION 跟AF微调一样,不只是牵涉到某个COPY的机身和某个COPY的镜头,
而是对某个特定机身/镜头的组合, 变焦的话不同焦距还可能不一样, 甚至同一镜头对
焦距离不一样, 调整还不一样. 你个同一个CHIP 是不可能普适于不同的镜头. 你测试
准焦的方法本身就不严格, 也不知道光圈多少, 收两档BACK/FRONT 就被DOF 掩盖. 退
一步讲, 就算是准的, 也只是运气, 换个别的BODY/LENS 就不定准.

是被具有激活功能的H3K36me3修饰的，看了一些别人在其它非表达的组织的ChIP-seq结
果，在非表达组织中我的基因没有被H3K36me3修饰。所以我想多做几个不同的组织器官
ChIP-PCR，如果所有的非表达组织内我的基因都没有被修饰，而只在表达组织内被修饰
了，从而可以验证我的特异表达基因是受H3K36me3调控。想到的方案有：
1. 每一个组织的ChIP DNA和INPUT比较。如果ChIP相对input在表达组织中是enrich
的话，应该可以说明问题。但似乎无法找出合适的内参
2. 所有组织的ChIP DNA之间互相比较。同样也是存在着内参的问题，植物中我似乎
没有找到一个基因被报道说在不同的组织中甲基化修饰程度相同的。
请教各位有如何选内参？也有看到文献说直接用picogreen测浓度调节模板量的做法，
但不知好一点的杂志是否接受这种做法？ |

但被拒绝，理由是当时我已经有私人保险。
现在因为是保险续期的时候，
想请问，如果我秋季开学的时候不续保险(我们学校不强制要求学生买保险)，
然后我再申请chip，这样的机会大不大呢？
因为现在资金紧张，实在是不想再买保险了。

1. chip前瞄好球，估计好距离，然后chip的时候眼睛一定要盯着球，打完球不要急于
看球奔向目标，不然头动了，容易chip坏。
2. chip时候手臂和球杆成一条直线，chip之前和之后都尽量保持，手腕不要用力。
供参考，也请大牛指教。 |

和CHIP 都拿下来,上环氧, 将干未干是(还能动),把环跟CHIP 一起在装到几身, MAKE
SURE 对准了(开机看读数), 没完全对准的话这时CHIP 还能推动一点. 等干OVERNIGHT.
装过起码一打没废过一个CHIP.

现在做的是RNAPol2的ChIP，大家都说RNApol2的这个antibody做ChIP很不错，但我步步
按照Millipore ChIP Kit做下来，elution之后用nanodrop测了一下DNA浓度，发现特低
。请问版上ChIP大牛们，要想得到个decent qPCR结果，ChIP下来的DNA浓度有个限制么？

程做完之后由于DNA浓度太低（0.1-0.2ng/ul by Nanodrop），不知道整个ChIP过程是
否成功，是否拿到有效的DNA片段。这个转录因子没有非常确定的target gene，所以也
无法设计引物做ChIP-PCR。请问各位，还有什么方法可以确定产物中含有富集的DNA片

TAG，就用TAG做CHIP. 省了抗体的钱了。 注意要一个non-sepcific recruitment
control. TF overexpression的话哪里都bind,你的negative control region可能
background也会很高。要是做转染，在转一个DNA binding domain amino acid 突变体
，然后再做CHIP,这样就能控制你的结果。 |

protenase K treatment(1/10-1/20 depend on the expression level of your
protein of interest). If you pulled down a lot of your protein of interest,
and if your procedure is clean, I see no reason the CHIP-Seq wouldn't work
if your protein is indeed a DNA binding protein.
In worst case scenario you should still be able to get lots of strong peaks
which is particularly useful to you since your problem is that you didn't

Kit，挺好的，因为当时那种细胞长的好，初始Chromatin充足。现在用的一种细胞长的
特别稀，15cm盘的铺满了不到1 million，然后还要做Transcription factor的ChIP,
根本拿不到之后需要做sequencing的量。
做qPCR确定ChIP本身work，只是浓度太低，都几十pg/ul，超级郁闷。

So far QCOM's high end chip are all SoC, which means the AP is included.That
's not what AAPL wants, especially considering AAPL's A4.
I guess it might be that QCOM will supply a combo (CDMA+WCDMA) BP chip,
modem only to AAPL, that will be a very low margin chip and QCOM will not
like it.
So far even the CDMA only version has many many bugs,not even to mention the
stability of the so called combo chip...
