0/2/5/20/50/100 ug/ml 的target protein，RNA MW ~30kD，protein MW ~200kD，PI
~5.3。用的是6% PAGE gel，没调pH。
主要有两大问题：1. free RNA band（bottom bands）是smear，而且band intensity
非常
低。不知是不是因为gel conc.太低的缘故。
2. 如果只是这一点也就罢了，从RNA-protein complex band应该就能得到kD，可是
0protein
smaple就有两条band，杯具。。。
之后做了点troubleshooting: 1. run了其他两个RNA sample，也都有两条band，所以
不是某一
个sample screw up；2. 怀疑protein-RNA complex没有入胶，改用4%gel（以前做过
staining，free protein 4％ gel之下肯定能入胶），结果和附件基本一样。

拉下来，在银染上看到带，才切下来去打，然后一系列实验，比如gel filtration，体
外重组蛋白重组这个complex等, 验证了，才敢说是complex吗。
现在很多文章所谓的纯化蛋白 complex，都是不需要看到清晰的条带，直接把elution
全部去打质谱（或者在切整个胶），看到三四十个蛋白，随便找几个自己想要的，就说
是interaction。

现在住的地方fedex和ups都送到家门口的porch，我只要把东西拉进门、该寄件时贴上
标签推出门让fedex来收
新城市的apt complex设计感觉fedex ups应该只能把东西送到在主要大门外？没有住过
这种complex，所以不清楚fedex ups会不会帮忙搬到公寓门前.....
假设fedex ups只会送到大楼门口，大楼门口到公寓门口还是需要自己搬，因为东西很
重，目前想到的几个办法是：
1. ups mail box: 但是因为都是fedex寄出，所以还是要把东西搬上车、载到fedex寄出
2. fedex hold mail: 只能暂存五天，通常inventory寄出会超过五天，还是要搬回家
、载到fedex寄出
请问有没有更好的方法？ |

这个想法我在2005年1月之前，换实验室时，和PI说了，他听不懂这张PPT意味什么。
后来我又换了实验室，2007年（2008？）某天新老板和我听完某个不相关的seminar，
和我说起这个，他才开口说了2句，我就知道他想说什么，
他的反应是，i have not finished my briefing yet, you really understand my
point? 于是就和他说，of course i know what you want to do,然后讨论了半天，原
来他的朋友有这个想法 etc etc，后来我又做了一些Homework，还是决定放弃。这个很
不好做，估计是个死途。
这个技术最大意义是：能找到locus specific DNA-protein complexes。目前的通行做
法是，已知某个protein, 你用CHIP，证明在某个Locus上面，然后开始making story，
编剧。但其实，那个Locus上面可能有几十种特殊蛋白在调控！！！

those found in computer science. From an information processing point of
view, human mind is an adpative, complex algorithm. In fact, all of modern
cogntive science is trying to do is to work out this algorithm.
The problem, I guess, is that most non-trival problems human mind is trying to
solve are NP-complete. So, a direct application of complexity definition in
computer science won't get you very far.

感觉拍得不错。风格有点像斯皮尔伯格的慕尼黑。视觉主要是从红军派（RAF)，所以给
