raw没有任何修改(饱和,锐化均没有)直接输出到16bit TIFF,为了做灰度渐变效果
分别0ev和-3ev输出各一张.后面所有的调整全部在TIFF的基础上完成

花了我大概30min,这个不是吹牛,我承认我用了ps的方法制造了灰度渐变的效果(单画幅压
暗天空),但是8bit下无论如何做不到这种影调的变化,拿那个原图拉一下饱和度或者
curve
就知道了.这里再打击一下curve万能学说,经过我这半年的ps再深造,我深深的感觉调
片子,图层叠加和mask的结合或者和history brush的结合比curve牛b多了.
(再附1个case,不过是8bit TIFF输入ps调整结果

cycle是两倍。如果你的efficiency不是2,比如是1.85,那么3个cycle就是2^3和1.85^
3的差别,也就是说实际6.33倍的差别被当成了8倍的差别。这是你的internal control
。如果用这个去adjust你的target的差别,加上target本身的efficiency如果也不是
100%,这个真正的差别就会非常大。
第二个问题是amplification是S型曲线,就是说,在开始阶段,或者在东西很少的不在
中间一段linear范围内的时候不能用delta delta Ct。
我的意思是,cell number不是可信与否的关键,关键是amplification efficiency。
在你怀疑你的结果是否可信的时候,建议用standard curve instead of delta delta
Ct来计算。只要你的sample的Ct落在你的standard curve里面,而你的standard curve
的correlation ef |
