说到男性更年期。我虽然在雌激素这个领域作了20年研究，和雄激素相关的研究就少多
了，大约接触过4-5年吧，当时是实验室一个学生的课题和雄激素有关，就读过一些相
关的文献。总起来讲，两者都是steroid这个种类的激素，分子水平上的机制是很相似
的。
男女都有更年期。这是由于身体里雌雄激素水平随年龄变化而出现的生理现象。但男女
有不同：男性的雄激素水平在20多就到了顶峰，然后就慢慢下降；女性的雌激素水平下
降则在45岁前后出现快速下降。所以，这个不同导致的症状就很不一样：男性更年期症
状不明显，女性的就反过来了。所以（加上乳腺癌），针对雌激素的研究就比雄激素的
研究多得多。
其实，仅仅是激素水平下降，补一点不就行了？是，很多症状在补一点激素后就会好转
或消除。这就是当年女性Hormone-replacement-therapy的基本思路。这个流行了几十
年，一直到后来发现更年期后女性乳腺癌和雌激素有关，才在2008（可能有误）前后正
式停止使用。

误提示，当时
没在意，不记得写什么了。后来发现台式机的速度越来越慢。好象是移动硬盘自我寻找

动硬盘断开就快了，后来重启机子，准备再考数据的时候，发现机子重启的速度狂慢，

（windows xp）发现原来移动硬盘的logo不见了（WD的logo不见了），能看见盘符，但
是进不去，
而且一点移动硬盘盘符，电脑好像就是死在了那里。cmd进入dos，进入移动硬盘提示

动硬
盘。但是就是进不去。
从wd官网下了检测软件，说是硬盘正常，但是从computer management里看硬盘是正常

硬盘，打开，说不能读，要 format, 点右键进 properties 显示 used, free space
都是 o bytes. 进 commmand prompt 进不去这个硬盘， 说是 data error cyclic
redundancy check。
硬盘要不要到无所谓了。怎么才能把里面的文件搞出来？请支招。 多谢 |

比较稳定，没有出过什么意外。
最近换x230了，然后买了expresscard to esata的卡，用esata连接这个硬盘，每次使
用win7的backup程序备份数据的时候，到最后一步都会显示data error : cyclic
redundancy check
我低格了一次磁盘，也用windows自带的磁盘检测，都通过了。但是用esata备份还是有
这个问题。今天用usb2.0备份了一次，居然通过了。。。然后再用esata增量备份，也
没问题。

For example, when you call setText for instance, it will try to repaint,
and painting itself will trigger your action. Those all take a while.
There are many things you could try to optimize
1. obviously find a way to update the data w/o triggering redisplay
until ready for update.
2. Use immutable objects that eliminate the need for synchronization. |

这些利用的就是 32-bit int/pointer 在 32-bit 机器上是 atomic 的。
你最好看下书。这东西挺简单，但是得有概念。 |

Anything private within a class (never shared with outside) should use
unique_ptr.
Cyclic pointer problem is solved by deliberate design effort on the class
interaction diagram (unrelated to inheritance). As long as there is a "top
level object" which has ownership, a solution can be found.
RAII 就是所谓 Linear Stack ，因此不能支持 capture by reference 的 closure（
但仍支持capture by value (copy) ）。 用 C++11 lambda 的时候还得用 shared_ptr。

纯化胰岛素受体的模块结束之后，我们这一组人进入到最后一个模块，
做钙调蛋白的纯化和分析。钙调蛋白(Calmodulin)十分重要，几乎在所有
真核细胞里面都能找到。这个蛋白只有148个氨基酸，大概16KD，比较
小。有Ca2+结合的情况下，钙调蛋白的构象可以发生较大的改变，从而激
活一系列酶的活性，比如cyclic nucleotide phosphodiesterase, 一些
protein kinases, phosphatases 和ATPase等等。这一个模块的实验中，
我们是从鸡胗里面提取和纯化钙调蛋白，然后再做一些分析。
这一个模块的纯化设计，可以说是最大限度的利用了钙调蛋白的特殊
性质，我觉得设计的很精彩。不过我对这个模块的实验经历并不是很满意。
教员的实验助手只有一个人，而且非常不称职，对实验本身似乎不太了解，
也不是很helpful，感觉比前几个模块的实验助手差多了。教员叫Constantin，
是一个俄罗斯人，在rutgers当教授。我起初对他印象不佳，后来发现这个人

afaik, the peptide w/ best kd is some RGD variant--there are a loooot
of variants, such as dimer/multimer of RGD, cyclic RGD, peptidomimetic
RGD, etc. It seems to me that no linear peptides has very good kd (again
from very small sample size), since peptide chain is too flexible.

写点自己的浅见来表示对这个领域的大牛的仰慕之情。
就先从这两篇science说起吧。确实这是该领域的突破性进展，发S也是当之无愧。一篇
文章阐述了对于DNA virus 引起的antiviral interferin response的全新机理。以前
该领域一直致力寻找直接结合双链DNA的受体，也找到了好几个，但是在KO小鼠上面表
型并不十分明显。
而Dallas的james chen领导的小组发现DNA virus or ds DNA经常在细胞里诱导核苷酸
的转化，其产物cGAMP则能够直接激活下游的interferin responses. 所以这个cGAMP
就是在这个天然免疫反应里面一个secondary messager。当病毒或者细菌DNA进入细胞
被降解转换的时候，cGAMP 的产生能激活抗病毒免疫反应来诱导干扰素反应和阻止病毒

但是看到Christopher K. Glass 和Michael G. Rosenfeld 这种大牛的paper上那么好
的结果我都不知道他们是怎么做出来的，他们的所有的paper中 ChIP 的结果也都是那
么漂亮，可是我用的同样的抗体，同样的方法，根本重复不出来，总不成写EMAIL 去质
问他们，你们他妈的是怎么弄出来的。

crystal structure resolve了！ 另一片纠正了陈志坚science paper里面的一个小错
误，2‘-5’和3‘-5’ phosphodiester bonds！
仔细看看crystal structure的文章，protein production和purification都是他们自
己做的，不是找陈志坚拿的！ 他们3月21号投的nature，陈志坚的cGAS发现
publication最早在sciencemag online刊登于12年12月底！ 这就意味着，这个
structure组从自己clone，protein production，purification，crystalization，
resolve，writing一共90天完成，太amazing了！ |

单纯从STING的重要性来说，目前还不知道啊~ cGAS是个新gene， 希望以后genome
study把cGAS划进去，如果cGAS真的跟尼玛autoimmune disease很有关联，那就牛b了，
