looks like some tumor cell transporting systems are specific to these
fluorescent dyes. PET imaging depends on tumor-specific characterirstics
such as higher metabolic rate (FDG probe), or tumor markers. This paper is
different in that the fluorescent dyes are supposed to be non-selective.
However no mechanism was studied in this paper. Can anyone think of any
mechanism?

另外，我不知道你具体用哪个dye, 但是在我印象中cell tracker这个dye
是通过和细胞膜表面上的分子连接而染色的，没有必要让细胞去“吃”。

不能像是抗体一样 想看什么就看什么 想看一的分子还得先找到可以用的dye才行

fluorescent dyes and quenchers, and I want to digest the antibody to small
fragments to break the FRET between dyes and quenchers. It would be best if
I can digest the antibody into the smallest fragments. Papain may not be the
best choice as it only cut antibody into three pieces. Maybe I can use some
kind of protease mixture? Any suggestion is welcome. |

如果某种一抗经常用，就去标记那个一抗，alexa dye那么多，想500nm就500nm，想
800nm就800nm，还可以用几种不同的一抗标记不同波长的dye去看同一块membrane，最
后用个imaging的机器去看就行了，sensitivity，contrast，用啥波长都随便调，根本
不用二抗，WB从跑胶到最后看结果小半天绝对搞定。话说，现在licor odyssey难道不
是很多人买吗？
如果一抗经常换，二抗去标记fluorophore也很方便啊~~~ |

单了，说出去不被人scoop 才怪。她本身的研究方向不是搞材料的（虽然她有这个能力
），从她的文章看来自从独立以后都是做单分子成像的，而且她以前发展过的另外几个
dye 都是用来在不同用途进行成像的。很难说她居然对optical storage这个方向本身
会有什么兴趣。而且如果这么一个dye 是她们顺手发展的话居然那么巧合就能用来做这
个技术未免有点太巧了。 |

compensation，但是信号不够强。因为compensation只和dye有关系。可否这样，用于
compensation的group加dye后在四度时间放长一些，或者在室温放置，而样品还是放置
在4度30min。
这样compensation的信号应该会强一些，而且不会影响compensation的效果。这么说对
不对? 有没有这么做过？或者有什么其他的办法可以增强compensation的信号？

想起以前的一件事, 是十多年前了, 在 Confocal email list 上. 有个ID发了个图,
是某个组做PH-sensitive dye. 用这种dye 的 强度来测样品里的PH value. 图里面几
个 control 的样品, 被认为是PH 值恒定来做校准的. 看起来是很不恒定.
ID 就问: 这样的测量有多大可信度? 有人附和.
文章作者也出来了, 回复了一下. 下面争论就更多了. 文章作者也不再出来了.
这个时候来了一个ID, 自称老ID, 说你们这样不对, 也不好. 网上瞎指责谁不会呀?
科学讨论不应该这样, 觉得有问题直接发信的原作者, 或者杂志.
结果炸出来了一个元老级ID, James Pawley. 做 Confocal 的人多少也知道这位, 那
本号称”BIBLE” 的 “handbook of biological confocal microscopy” 就是这位主
编的.

t have much choice of stokes shift.
if you need that big Stokes shift for sure, quantum dots are your only hope.
but i don't believe it is the case in your initial problem.
check your fluoremeter setup. make sure you are using the correct settings
including illumination and filters. |

米表面吸附能力是比较强的, 你要是怕不保险, 弄个有2,3个苯环的dye试试, 亲油加共
轭应该能吸附上吧? (其实这个已经有好多人干过了的).
离心洗洗就纯化了, 先拿个紫外灯裸照看看有没有荧光, 有就继续, 没有的话, 也不要
找我的麻烦?//////
很好得建议；不过水体中别的有机质不可避免得也会和dye结合产生干扰，如何解决？

处理效果还是处理后的残余?
发文章的话, 你就用纯的炭纳米先接上dye,然后.... 最后看水中残留炭纳米多少, 离
心然后测荧光. dye就相当于一个指示剂, 用来说明炭纳米的去向?
做项目想监测水处理后的炭纳米残留? 还是建议离心浓缩先.但是弄出来不好分析. 拉
曼可以检测有没有, 定量好像比较难, 反正我从没弄得它工作过.
还有一招, 炭纳米的尺寸比较大, 一般的颗粒检测仪器(就是最便宜的都可以)因该可以
检测到它的散射信号. 可以把要处理的水先过滤一下,去除大颗粒,再用炭纳米处理其它
的, 然后离心浓缩, 再悬浮在水中测光散射强度. 准备纯的炭纳米不浓度做标准曲线.
应该可以大致定量.
土人土办法, 您自己斟酌吧 |

其实 dye-sensitized solar cell 也算一种hybrid，不过现在搞Organic solar cell
很时髦（不知道为啥这么低的效率，Organic solar cell 还能开公司）。现在就有人
搞起来了Hybrid solar cell。
看目前样子，dye-sensitized solar cell 快到头了，效率很难提高到10%以上了。

机dye labeled到一个生物分子上,有一些imaging, 看到了FRET应用到单分子上的可行
性,而完全没有study任何生物方面的内容,因为很多技术问题(比如CCD能够real time的在程序控制下做不同dye之间的colocalization)都还没有解决.而后面的1999年的PNAS才study了RNA的conformational change. 是在Chu lab完成的,而后Steve很快解决了很多技术困难下,推广到更多的体系,包括protein-nucleic acid interactions. 你可以问任何一个single molecule领域的人,Simon Weiss和Steve Chu谁在smFRET的contribution更大.

我前不久贴出了别人用中古拟音念唐诗的视频。虽然这里的一些北方人表示质疑，但是
中古汉语的发音和后来它在各地发生的变化过程都是比较清楚的。
相比之下，语言学家对上古语音的还原就存在非常大的分歧了，连界定什么是上古汉语
都有分歧。（一般来说，上古汉语是那些秦前后书面语可以对照的口语。）现在比较主
流的几种方案都通过汉藏语系内部的横向比较，外加对汉字声部的推理，还有侗台语系
和其他语言的音译文，得出一些难以置信的结论，比如：
- 上古有复辅音，也就是kl gl bl hm hr这种重声母开头的发音
- 承上，上古有一字对多音节现象，有称“角落”原本是“角”一个字的发音
- 上古无声调，但有类似侗台语的紧喉音和s韵尾之类
各人觉得，上古时代中国境内的语言情况一定非常复杂。后来周朝的“雅言”口语，估
计也早不是什么纯汉藏语系语言了，而是一种以商代某汉藏语系语言为主体底层的混合

Until the 19th
century, most pigments were based on naturally occurring colored minerals
and dyes, with three significant exceptions: Egyptian Blue
(CaCuSi2O10), Chinese Blue (BaCuSi2O10)/ Purple
(BaCuSi2O6) and Maya Blue. The former two are
alkaline-earth copper silicates, and because of
this similarity it has been proposed that the
Chinese pigments were derived from Egyptian
