该是7周了，平时的离家都是很准的，30天周期。
医生说约下周一再看B超。
其实我怀孕以来除了嗜睡，胸胀疼，尿频，易累，这一周开始早上刷牙会恶心，其他没
有如恶心等症状。
也没有腹疼，和bleeding。
就上周有几天发现白带稍微带有一点深色，真的很浅。现在没有了。
大家说7周看不到胚胎正常吗？下周一需要再看B超吗？担心频繁阴超会导致流产。下周
二见OB。 其实觉得今天的B超师什么都没告诉我，就说看不到embryo。让我去抽了血。
我因为紧张害怕，忘了问更多的信息了，大家给出出主意我应该打电话回去问具体结果
吗？我是否明天要再去抽血看HCG是否翻倍？

一上去看不到embryo 就不看了，什么都没跟我说，就说看不到embryo，后来要伸头过
去看，他说yolksac在那里。然后就去找OB了，她从OB室出来，就跟我说，你去验血，
可能现在太早了，看不到，叫我去约下周一在B超。
我当时可能也是闷了，啥都没问。
不知道明天是否可以去找他们问问。 |

10月底，11月初作过几次功课。11月中旬用wondo查怀孕。显示没中。 11月26日用
clearblue，first response和另一个牌子的测。clearblue有很浅的第二根线。first
response 和另一个牌子的几乎看不出来第二根线。11月29日用clearblue, first
response再测。first reponse 测怀孕的那条线比control的线深。clearblue虽然策怀
孕的那条线颜色比上次有所加深，但依然没有control那条线深。我也有腰酸，胸涨的
反应。却不像怀老大一样有恶心，呕吐。到此，我觉得我怀孕了，可hcg水平也许不够
高，所以之前测的颜色一直较淡。今天看了医生。要说也8周了，医生说没有找到
embryo.要监测hcg.今天抽了血。结果明天才出来。然后周5再抽血。看hcg是否翻番。
我觉得，我hcg水平可能不会太高。离我最近一次做功课，也近一个月了。不可能说怀
孕时间太短，找不到embryo. 我这样是不是极大可能宫外孕，或是根本没怀孕？

不到就要过两周再约，不过运气不错，当天就看到那颗小花生豆了。我还特意问了ob，
做b超会不会对宝宝不好，我的ob很坚决地说没有事，而且每次都很快，不超过3分钟，
不会伤害宝宝的。 |

加了这么多！在网的中央有一个绿豆大小的“血块”，仔细一看竟然在啪啪啪地跳动，
原来是胚胎的心脏，我数了一下，每分钟大约120次。
下面贴一张candling照片，是这么拍的，我在鞋盒子上挖一个蛋大小的洞，在盒子里点
一个灯泡，然后放将蛋放在洞中，站着往下拍。不太清楚，不过还是能看到里面的血管
，没有embryo的蛋是透明的。
今天我又去了一次Petsmart，向他们要鹦鹉的inventory，打算猜测鹦鹉父母的种类，
要等几天他们才能给我，不过他们给我一个惊喜：又有一个新蛋。这个蛋与前面两个很
像，只是短了一截，重量也轻了一点，这个是12.4克，前面两个是13.2和12.8克，应该
是一母所生，鹦鹉妈的营养用尽，所以蛋小了。 |

Guess most will be natural cycle plus observation. If you don't ovulate
naturally in the cycle, then you might have some medicine to stimulate (
shall not be high). After ovulation, then fronzen embryo will be transfered.

我们是用小鼠胚胎的软骨和骨头染色的protocol。染软骨的话只用Alcian Blue即可。
最后胚胎用BABB处理就会变得透明。BABB危险，而且会腐蚀显微镜，使用时要小心。很
多动物的胚胎都可以用类似的方法染骨头（红色）和软骨（蓝色）：鼠，龟，蛇，鸡，
鱼，蜥蜴，变色龙，蝙蝠。。。而且都很漂亮！

话说回来，STAP cell是否真的存在过？我读那两篇文章时，有一点觉得很奇怪，他们
的细胞竟可以形成placenta。要知道，包括inner cell mass在内 mESC是pluripotent
cell, 一般只能形成embryo,而不能形成extra-embryonic tissue（比如placenta）。
小保方究竟是用了什么细胞，具有totipotency,可以同时形成embryo和placenta? 如果
他们用blastocyst之前的embryo(比如2-cell， 4-cell，8-cell， morula，这些细胞
是totipotent)，倒是可以同时形成embryo和placenta,但这种实验从genetic
manipulation (将GFP reporter整进所用细胞genome)到generate chimeras,不可能是
一个人可以做的，至少得是几个人参与其中。合作者难道就没有质疑吗？
小保方又是怎样收到启发，从而打破常规，检测STAP cell的totipotency的？完全凭空
造假的话，这样的胆子真是难以想象。 |

杂志上了，不管如何，这下子这个open access的杂志算是出名了一把。
我觉得国内还是什么都敢做啊，这种东西是不是ethical真的就是一线之隔，他们号称
用的是因为基因缺陷不viable的embryo的cells，但是大家都知道国内那些小医院去搞
一个viable的embryo是多么容易的事情，再加上crispr的方便容易，在正常embryo上做
gene editing真是没什么特别的难度，除非法律有明确的规定。那篇nature的comment
里还特别提了，国内好几个组在做这个，至于用的是什么样的embryo，估计大家都不知

will get tens of sections from a single embryo for you to try. If the
antibody works, then move to whole mount. Just remember the time for
fixatives, antibodies and etc. to get into the center of the embryo will be
a lot longer than it is needed for sections. |
