BL21(DE3) or Rosetta2(DE3) strains for future or should we always start from
a new transformant?
How often does mutation occur in these strains?

It's usually recommended to renew those stocks every 6 months. But I've used
frozen stocks of several years old and still got satisfactory expression. I
guess it may vary from case to case, though.
I would do small scale expression test each time before a large scale
expression culture if I'm worried about the quality of the frozen stock.
Just use a 5-ml culture and induce as how you would with a 1-L or larger

我们是用小鼠胚胎的软骨和骨头染色的protocol。染软骨的话只用Alcian Blue即可。
最后胚胎用BABB处理就会变得透明。BABB危险，而且会腐蚀显微镜，使用时要小心。很
多动物的胚胎都可以用类似的方法染骨头（红色）和软骨（蓝色）：鼠，龟，蛇，鸡，
鱼，蜥蜴，变色龙，蝙蝠。。。而且都很漂亮！

glycerol, because freezing and thawing cycle can kill the enzyme. For other
proteins, I do not bother to add glycerol either.
And to get rid of gluathione, besides dialysis, there is an easy way. Take a
spin-column with an appropriate cut-off filter, e.g. 30KD, spin, concentrate

Dr. Burgess 这人特有意思，喜欢吹嘘他发现的小trick。比如，他得意说他是全美国最早用
coomassie blue染蛋白胶的人。据他说，六几年的时候他看到一个澳洲的老兄的一片文章,讲述
一种新奇的染料叫考马市亮蓝, 比当时流行的染料amido black灵敏十倍以上。所以他就写了一
封信要了一些，结果效果奇好。我们听得都目瞪口呆。
不过Burgess最自豪的，还是他的“万金油”buffer。他告诉我们说，他几十年了，总喜欢用这
个buffer的配方，什么蛋白都喜欢这种buffer, 可以说到了包治百病的神奇地步。尽管他吹得神
乎其神，配方却十分简单。这个buffer叫TGE,就是Tris, glycerol 和 EDTA。配方是这样的:
50mM Tris pH 7.9
0.5mM EDTA
50mM NaCl
5% glycerol
Tris EDTA NaCl 这三样都没什么，真正关键的是5%glycerol。有了这个glycero |

发帖数: 1130 | 14 偶来补充自己的看法吧：透析不是必须的，只要imidazole不影响activity就好。一般
不用再加protease inhibitor，因为细胞裂解的时候不是已经加过了吗。不建议加EDTA
，不知道lz是要做什么activity assay，很有可能这个蛋白本身自己就是需要镁离子的。
glycerol，对于一般的蛋白我们都是加10%，然后分装后用液氮冻完存在－80度。但是
碰到很不好保存的蛋白，比如冻了一两个月就没有activity的，把glycerol加到20％会
有帮助。
DTT或者beta mecapitalmethanol,保持溶液的还原状态，视情况而定。
存也会失活，那就需要找到合适的buffer，或者每次都用fresh made的蛋白 |

1. 如果你的蛋白有aggregation，加一些Triton100(0.1，0.5or1%). 听很多人说，这
很有用。
2. 用4-12% TBE gel （Invitrogen）。如果自己配，用4%。或者3%，下层加三分之一
15%防止free probe run out。
3. 如果还不行，用垂直配胶系统（Bio-rad配PAGE gel用的， 1.5mM）配Agrose胶（1%
？ 2%？记不清了）。里面可以加Triton & glycerol。
我的蛋白80kD， oligomerization 后500 or 1000kD （FPLC测定）。6%TBE gel完全在
well上，4-12%能进一点点，3%可完全进入但很难看。最近我们lab新来的Postdoc用了
Agrose胶，效果不错。我让他加了一些Triton & glycerol，效果更好了，binding
affinity 都增加了几倍。但具体浓度，我得问问他。 |

存了一份glycerol stock放在－80, 然后剩下的菌IPTG诱导,成功表达。
可是后来用glycerol stock, 同样的条件，却再也不能诱导表达了，我也从冻在四度的
板子上再挑过菌，还是不能表达。
现在的打算是再找别人借一次（主要是有六个蛋白要表达一老借也不是个事），但还是
觉得很奇怪，菌的抗性还保存着说明质粒应该还在啊，当然这个菌不是保存质粒的好选
择，会有重组，可我还是不敢相信六个样品都发生重组。
有人知道这种第一次可以表达第二次就不行的现象是为什么吗？先谢谢大家了。

deficiency，但是在SC 2%glycerol plate上drop assay，居然长得和WT一样。。。
然后尝试用p416 gpd promoter的vector rescue，却完全rescue不了这个phenotype。
