2. prepare 15ml falcon tubes with 1ml of LB + antibiotics;
3. pick a colony using a sterile tip; stir it in a the water
-containing PCR tube for several times and then throw the
tip in the falcon tube to inoculate the LB medium.
4. use a primer on the vector and a primer on your insert
to prepare 15ul master mix for each 20ul reaction.
By using this primer set, you will be able to confirm
both availibility of insert and its orientation. |

是试验不work，觉得是自己的责任， 我反正经常跟他说if its not going to work,
it not going to work no matter i do.
我发现试验这东西就是这样， 其实细节根本就不中要， 比如要2mls of 500mM
edta, 你就拿个pastuer pipitte 随便挤一下，其实都没问题， 知道那个大概就是1.8
-2mls
还比如， 你inoculate e coli 的时候用不用bunson burner 其实都没问题， 反正要
加antibiotics。
我的意思是说，一个东西要是那么convincing, 大手大脚， 随你怎么做， 都能做的出
来。 那种要技术才能出得来的data, 就不牢靠了。 |

inoculate的时候小心些，不要用什么牙签之类的，也不要用wire loop。。试试用长的
sterile的pipette

inoculate我都是用sterile pipette tips。
如果是bacilli，为什么一开始会是正常的cloudy的菌液，然后慢慢变清出现这种悬浮
纤维状物质呢？我没养过bacilli，不知道它的生长状况是如何的。
第二张图里那种一团团的菌体是什么呢？除了它之外，那管菌液本身的生长状态和浊度
倒是挺正常的。 |

the overnight culture into the final culture. Then wait until OD is 0.6-1.0,
add low concentration of IPTG such as 0.1 mM IPTG for 16-20 hours at 300rpm
. I just tried this protocol, it increased a lot the ratio of GST fused
protein in soluble fraction compared with that at 37 C. |

using 18 C to shake ( cell grows much slower than 37C) until OD is 0.8 and
then I added IPTG at final 0.1mM. The speed of shaker when I started using
18 C is 350 rpm.

After IPTG was added, the culture was shaked at 18 C for 20 hours in order
to get enough cells.
The big difference based on your protocol is that I started low temperature
the next morning when I did final inoculation, while you still used 37 C.
I compared the soluble and the pellet fractions at 18, 28 and 37 C,so I know
18 C is the best one. |

a OFP-encoding gene into E. coli sm10 cells. I inoculated cells both on
plates and liquid. The cells from agar plates can glow very well under
microscope, but not for cells from liquid. The cells from liquid medium few
cells can give signal. Does someone here know what cause this?

也可以用stirred bioreactor
你可以做10x的scale up。比如 1L 到10L
CHO-S（invitrogen）的话比较皮实，1E5/mL都没太大问题
293F (invitrogen)starting conc. 3-5E5/mL
media的话，如果你不要求高密度，DMEM/F12, rpmi都可以
如果加血清的话更好
很多公司都卖serum free medium
拿CHO-S 做例子，有invitrogen, hyclone, irvineSci等等
wave cellbag不用灭菌，传统的glass flask用蒸汽就可以了
inoculation的话 上youtube看看教程吧
