发帖数: 28562 | 2 你说得很有道理。我们那片文章确实没有所谓的mechanistic insight,
这个我们也知道。只不过一开始我们以为我们做的东西非常新，而且在组织里面
找到了直接证据，所以以为很重要。相比之下，那个发了上千篇文章，以及超过50篇
CNS的pathway A, 其实就是在培养的细胞里面最明显而已，在组织里很难找到支持证据
，以至已经有人开始质疑那个pathway A在动物体内到底有没有用。
所以我们觉得我们找到的那个蛋白对pathway B的关系可能才是真正在体内有作用的关
系。在此前提之下，我们天真的认为，那些什么mechanistic insight 是follow-up 的
文章做得。
不过，真的要找mechanistic insight 其实也不是不可以，就是看要花多少时间的问题
了。
所谓mechanistic，就是A蛋白作用B蛋白，B蛋白作用C蛋白，C蛋白是公认的教科书功能
，无需多说。这就是我们这个时代的研究路数。
比如说，C是myosin，解释了宏观肌肉收缩，B是myosin kinase，A是某蛋白，能在某个

相同，我不太清楚改怎么改。program manager 说在第一页Specific Aim中，要
“Discuss the mechanisms through which the independent variables affect the
dependent variables”
同事说，reviewers look for novel hypotheses, mechanistic insights, or
analytical tools.
不太明白mechanisms，mechanistic insights指的什么意思。

发帖数: 296 | 8 相反，是因为太难，所以敢认真去碰的人很少。mechanistic study不是随便什么
人都能做的，既烧钱又周期长。很多时候还没有定论，全是crosstalk，牵一发动
全身，不太容易purturb one specific signaling。stem cell research也是这
样，一篇篇的好paper，说白了大都也是灌灌observations，鲜有碰触mechanisms
的。有人做stem cell differentiation期间的expressionprofiling，已经很算是
想干些mechanisms的事儿了，那又如何？profiling这种工作，其结果好比给你张
“错误概率未知，可能很大”的地图，除了他们自己人，谁敢拿它去指路？扯远了，
总之俺是尽量回避纯粹的mechanistic study，等天才们做出来的时候，俺在旁边鼓
鼓掌就行了。 |

尤其是translational或者preclinical study
至少在我做的这个领域，我看到大牛也常发PLOS one，
要么就是做到很牛的那种，上science translational级别
如果没那么牛，投那种经典的杂志，比如Journal of Neuroscience
reviewer就盯着mechanistic, novelty看，要么要求补一堆机理的实验，要么直接
novelty悲剧
translational的有多少mechanistic， novelty。
这次投稿之前老板还说过，最近做AD 的translational 或者 preclinical的
development的发PLOS one 越来越多，我们也应该凑个热闹，嘿嘿
我手上的这个study，投Journal of neuroscience 基本没戏，投neurobiology of
aging 不完全沾边，而且特别慢， 投Neurobiology of disease和PLOS one没啥区别 |

发帖数: 11893 | 14 我们就是把负责做那个修饰的酶给KO了，然后pathway A 和 B 同时出问题了。
然后我们把修饰好的蛋白扔进去，发现该蛋白和pathway B colocalizes, 而且recue
了pathway B，但是对pathway A就没有这个效果。
其实这个东西对pathway A到底有没有作用我们不太关心（阴性结果不说明就没有关系
）因为针对于那个蛋白和pathway A的文章超过一千篇，所以是一个没有什么意思的方
向，所以我们那个文章里面没有对pathway A下任何结论。
不过对于pathway B的结论应该是没有问题的吧？因为能够很明确的rescue, 而且很明
确的colocalizes.
批注：全部症结在此，这个叫Involved，确实没有mechanistic，
所谓mechanistic，就是A蛋白作用B蛋白，B蛋白作用C蛋白，C蛋白是公认的教科书功能
，无需多说。这就是我们这个时代的研究路数。

excluded as the stoichiometric oxidant for the reactions according
to their reaction procedure. (THF has been distilled from Na and the

之前主要是基于X衍射或者核磁共振的技术开发和应用，拍一些单分子的裸照。82，91
，02年颁给X衍射和核磁共振的技术开发，剩下11个的给了应用。
而冷冻电镜，这次算是第一个（可能是第二个），直接给了技术。用它可以看更加复杂
、分子量更大的大分子复合体。这些大个头的结构当然更有复杂也更有意思。
这个技术开启了一个新时代，细胞里重要的复合体实在太多。接下来可能拿奖的那就是
：真核细胞的细胞核通道复合体；一一公搞的剪接体（这个结构无论在生命进化和疾病
相关都非常的重要）；G蛋白偶联受体（跟N多疾病有关）等等。
彦宁的工作好像确实没有什么希望，葡萄糖转运蛋白跟上面这些不是一个量级的。

HGT is a two-step process with mechanistically and bio- chemically distinct
phases. Firstly, there is passage of donor genetic material across the cell
membrane(s) of the recipient cell, including other envelope structures such
