rt 我用的是Phenomenex Aeris widepore C18 column, 是专门用来分离protein的，
mobile phase 用ACN/H2O 0.1%TFA, 一开始只用两种standard antibody试条件，无奈
尝试了很久都没有分离出来，Gradient slope大了solute就全部一起洗出来，小了peak
就宽到不行，昨天试了一个long gradient用0.1%B/min结果peak宽到几十分钟几乎看不
到...
有没有大侠有过类似经验的给点指导，感激不尽。。。。 |

Dr. Xu was a student/postdoc in David Scheinberg's lab at Cornell/Sloan
Kettering, but I don't think he ever got a faculty position there. Dr.Xu might have made some contribution to the CD33 projects, his main work was about the regulation of CD13/aminopeptidase N.

这几种单抗是可以在很多公司定制的，以前只听说过老鼠的怎么生产，知道要花很长的
时间，刚刚看了一下兔子单抗的生产工艺，也不是那么简单，基本上也要半年左右吧。
不明白为什么新冠出来几个月已经到处都有各种单抗在卖了，哪位大牛能给解释一下吗？
另外想问的一个问题是抗原是怎么来的呢？

other way around.
Monoclonal antibody is against one specific epitype of antigen. If
it works, it works. If not (for example, under immunohistochemistry
conditions, methanol, paraglutahyde...), it won't. Polyclonal antibody
is against a great variety epitype of the antigens. Some clones of
antibody might only work for western, the others might work for immunohist,

stroma cells， lymphoid cells， endothelial cells.....
即使只看tumor tissue 中的tumor cells， monoclonal 和 heterogeneous 也并不矛盾。
tumor tissue 里的monoclonal 的 tumor cells， 即使有相同的DNA 背景，
但是在transcription 水平，不同的cell populations 的 gene expression patterns
会很不一样。

treat cancer. That line blurs a bit because there are newer medications for
cancer that are not in the same classes of typical "cytotoxic"
chemotherapies. These new treatments, for example, monoclonal antibodies

2019年末，武汉暴发“不明原因肺炎”。2020年1月9日，以中国疾控中心徐建国院士为
首的团队向公众发布消息称，在武汉的不明原因肺炎患者体内分离并检测到了一种新型
冠状病毒。发稿时的最新数据显示，截至1月19日22时，武汉累计报告新型冠状病毒感
染的肺炎病例198例，其中治愈25例，死亡3例。
经历过2003年非典疫情的人对冠状病毒不会陌生。那场可怕传染病的病原体正是一种冠
状病毒。如今，与SARS病毒相近的新型冠状病毒在中国大陆卷土重来，我们应该怎样应

经过一个中间宿体性的其它动物才能传染给人。所以，病毒发生后，就故意误导全国人
民去海鲜市场找证据，找其它野生动物的麻烦。 而且还是病毒所去找的，找完了还装
模做样化验呀，分离呀什么的。最后把责任全部推给了海鲜市场的动物。可是那种动物
，一直不敢说，说了其它相关已经在人就会去早那种动物监测。 所以压根不说，打马
虎眼。
事实上，早在2013 年，就是这个武汉病毒研究所，已经从来自云南的蝙蝠身上所携带
的corona virus中分离出第一株蝙蝠SARS类似样的冠状病毒的活病毒，其中就包含了类
似于S类型的基因。从而证实这株病毒能够使其接受和SARS病毒相同的受体，并能够感
染人的细胞。对此新发现，武汉病毒所还把它以武汉病毒研究所的英文简称命名“WIV1
”，以彰显这一发现的重要价值和属于自己第一个发现的巨大研究成果。这个成果刊载
于2013年11月的《自然》杂志。
就是说，从云南弄回来的这种蝙蝠所携带的类似于sars的 corona virus， 可以不经过

摘要
sars和mers的出现表明跨物种传播病毒对人类具有威胁。
这里我们研究一种在中国马蹄蝙蝠里流行的SARS-相似病毒，SHC014。
使用SARS冠毒的反向遗传系统，我们把SHC014的spike蛋白装在了一个可以在老鼠里复
制的SARS病毒框架上，并对它进行了一些研究。这些结果显示，SHC014所属的2b类病毒
可以使用SARS的人类受体，ACE2。这种病毒可以在来自人的呼吸道的原代细胞里高效增
殖，达到跟SARS病毒类似的病毒滴度。而且，体内实验表明这个人工病毒可以在老鼠肺
里产生病理效果。
对现有的抗SARS免疫疗法和其他疗法的测试显示，这些方法都不太有效。抗SARS的单抗
没有中和病毒，抗SARS的疫苗没有产生有效的免疫效果。
（总结）我们造了一种全长的SHC014重组病毒，并证明它可以在老鼠体内和细胞培养的
条件下有效复制。我们的工作提示SARS有可能再次从蝙蝠病毒传到人类。
