，我常常都希望身边也多一些华人，尤其最近觉得自己也有责任扩大这个圈子。
心血来潮先将这么多年面试的几样大家要避免的：
1. 不知道自己要做什么。这个在tax或audit的问题会少一些，但是advisory因为杂，
所以很多同学都不是很清楚，所以也是为什么要在這里简介一下。面试中常常有同学说
，我什么都能做，这其实是减分的答案，因为这说明你没有做足够的研究。你可以给两
个option,但是一般还是选择一样比较有胜算。当然，我们也碰到过实在是很喜欢的人
，就算对方不清楚自己要做什么，我们也会替他考虑什么更适合他/她。
2. nagative attitude。不管在什么情况下，对过去的事和人，甚至自己的国家，不要
有抱怨一样的negative compliant.你可以将一些事实，然后趁机将自己如何应对。
3. 背书。这点老美的面试官会感受比我明显。我常常觉得还行，老美却觉得有script,
或者rehearsal过的嫌疑。我一回想，果真是。有时我问一个问题，同学答着答着就扯

是不是有点呆傻了，以至于不会和别人打交道了。
我之前在国内工作过两年，带我实习的那位和我说，和上司打交道你有两个选择，一是
negotiation，另一个是argument。你如果擅长其中一种，你就会和老板相处的不错。
我本身不擅长后者，于是修炼了两年negotiation。
我老板是个30来岁的女PI，按照版上众多坏PI标准，就是个“nagging bitch”
，和这种PI相处，以前修炼的negotiation重要就体现出来了，你必须把众多实验里她
认为最重要的都做完，即使你自己认为是bullshit的你也要按时做完，甚至多做几遍，
让她觉得她的建议是有价值的，即使是阴性结果她也会觉得很满足。这样你就可以找机
会和她谈论一些你认为可能可以改进的地方，甚至你自己设计的实验。要学会不要被她
牵着鼻子走，甚至在适当时候牵着她的鼻子。
所以版上好多人应该想想怎么在满足老板的前提下去争取自己的利益，怎么和上司相处。
而不是只会抱怨，觉得自己英雄无用武之地，因为跟了一个熊老板。 |

发帖数: 214 | 14 谨慎猜测你们是不是刚开始做de novo design？如果你们是业界大牛，请原谅我的唐突。
de novo design预测出来的结构，最后表达不出来的，或者折叠不了的，可以说是十有
八九。只是这样的nagative result是发不出来文章的，所以外行看了几篇N/S文章，会
有结构预测已经很成熟的错觉。比较稳妥的做法，还是要参考自然中已经存在的蛋白，
最好是结构已经测定的蛋白来做骨架，然后再通过计算修正。100aa的小蛋白或者长多
肽的折叠已经非常复杂，如果没有homologues,几乎不可能直接准确预测。
另外，de novo design很大程度是依靠统计意义的成功。一般要有上百个预测的结构，
然后在实验室做筛选。最终序列还要做上数轮的定向进化修正。只设计一个序列，然后
就运气好爆直接成功的，我是没见过。
如果有公司在1000美元提供高质量（>80%纯度）的100aa的多肽，我建议你们直接定，
这个价格非常便宜。如果你们想继续试生物表达，除了前面的建议外，你还可以试试

wrong.
HW is affine term structure models. If you use 1 factor HW, it doesn't
capture non parallel shift of the term structure. Hence the TS you forecase
could only show paralell movement, which result in persistance positive or
nagative bias.
If you use 2 factor models, it can allow non parallel shift. But there are
many format of 2 factor models |

they say he died in his sleep. he was bedridden for couple years, they had
to hire someone to feed him, help him pee and poo.
i can't say that he and i were super close, my grand parents never raised me
or anything. but me and cousin would go and play at their old house for a
day on the wkend. i liked my grandpa, my grandma would nag him a lot, but
he would always be like, ok, ok, and never really change. kind of like me.
