knockdown的细胞里，也没发现有increased apoptosis。
原来这是个common issue吗？ 我做knockdown的细胞的一个主要目的是收RNA做
microarray，好看看这个gene影响到哪些genes/pathway。 为严谨起见，我从不同的
passage收RNA，而不是一个passage好几个盘子。 可因为这种loss of knockdown最后
我都收的RNA都不能用了。
如果用inducible knockdown的话，用多长的time window为好呢？ 3天？ 一个星期？
还是各有利弊只能做做看？ |

对reactive oxygen species很敏感，由此引起的DNA damage激活p53以及其他信号（正
常生长因子和matrix缺失等）激活p16-Rb通路，引起senescence。你应该问一下他们给
你的MEF的passage，最好要到分离后第1或第2代的细胞，这样你还能用几代。
但是如果你把这些停止生长的细胞保留着（不要传代），过几个星期你会发现细胞又长
起来了，而且越长越好，这就是自发immortalized的细胞。很多MEF细胞系就是这样建立
起来的，例如著名的3T3系列。但是我不建议你用这些immortalized的MEF做实验，因为
上述ROS引起DNA damage的原因，这些自发immortalized的MEF已经产生太多基因变异（
因为需要bypass p53和p16-Rb两个重要通路的抑制），即使你看到表型的差异，也很难
说是你的gene KO的结果。
如果确实需要长久使用MEF，一是拿到老鼠，分离新鲜的MEF做实验。二是利用低氧（3-

repeat）位点
大概三个月前测的是这个位点是homozygous有6个repeat
然后最近重新解冻另外一个stock，start cell culture，测量同样的位点，居然成了
heterozygous，一半仍然6个repeat，另外一半成了10个repeat；反复做了很多遍，换
了所有的reagent生怕是contamination，但结果还是一样。
问了lab technitian，这个neuroblastoma在我们实验室都是一样的来源，他也怀疑这
个repeat是不是dynamics的
三个月前的cell line的passage不记得了
最近这个cell line的passage大概是p10
我想问下，cancer cell line的genome microsatellite难道真的会传代传着传着就发
生了变化？如此不稳定？ |

is 90% A and 10% B, that's what you mean by "并不是完全homogeneous"
Then keep cell passage and wait until P20, it becomes 10% A and 90% B, this
is genome instability that lead to more heterogeneity?
Is it because allele A itself directly being "mutated" into allele B, or
cancer cell carrying allele B compete better than allele A so B could
survive over A? |

原来，两人投宿旅馆后关掉手机，与其失去联系的同学误以为他们走失才报警。“夜间
的灵山已经零下几度了，我们就在上面找了一晚上。”参加搜山行动的清水派出所冯所
长介绍，前晚6时50分，他们接到一名自称北大学生的女生报警，她的两名同学(一男一
女)早晨去龙口涧风景区游玩，但到现在却跟他们失去联系，怀疑他们走失。
昨日中午，冯所长接到同样参加搜山的北大保卫科通知，说两名学生已经跟他们联系上
并汇合。
据门头沟警方掌握的情况，这两名学生游玩后当晚在旅馆投宿，随后关掉手机。
第二天，两人开机后发现学校正在寻找他们，立即与校方取得了联系。
“这属于一个误会，虚惊一场。”昨晚，北大新闻发言人赵部长证实了该消息。他说，
学生周末出去旅游是个人权利，学校无权干涉。

此次被召回的商品为葛莱Passage?、Alano?和Spree?婴儿推车及旅行组合套婴儿推车。
次被召回的商品仅限于配有锯齿状塑料接合铰链的婴儿推车，生产时间为2004年10月～2008年2月，产品型号和日期代码标在婴儿推车背面。该推车自2004年10月～2009年12月在全美销售，其中简便型推车的售价为80～90美元/辆，旅行组合套婴儿推车的售价为150～200美元/辆。该婴儿推车自2004年10月～2010年1月在加拿大的Elfe儿童用品专卖店销售。
此次被召回的商品数量约为1528430（其中美国约150万辆，加拿大约28430辆）。召回原因为，在折叠或打开婴儿推车时，固定婴儿推车遮阳篷的铰链有割伤或划伤车内婴幼儿手指的危险。截至目前，葛莱儿童用品有限公司在美国已收到7起婴幼儿手指被铰链割伤或划伤的事故报告；加拿大卫生署尚未收到任何事故报告。

据报道，这艘触礁的游轮为“冒险号”(Clipper Adventurer)，由多伦多西边米西索加市(Mississauga)“冒险旅运公司”(Adventure Canada)所经营。该公司专营偏远地区的旅游项目。
“冒险号”27日在北极触礁后，旅客并未发生混乱。大家都很平静地等待救援。游轮工作人员曾向旅客保证，游轮虽触礁，但并无沉没的危险。
报道称，加拿大海岸防卫队在收到“冒险号”求救信号后，立即派一艘破冰船前往营救。在破冰船未抵达前，船上旅客或下棋消遣，或听船上对北极十分熟悉的旅客演说。
73岁的旅客杰佛(Ed Jeffer)30日在埃德蒙顿市国际机场接受加拿大广播公司(CBC)访问时表示，游轮触礁时十分恐怖。他说，他听到船壳发出极为尖锐的声响，当时航速为15至18节的游轮，在3至4秒间完全停止。
由于游轮突然停止，一些人摔倒，架子上的物品也纷纷掉落地上。当时游轮上共有128名旅客及69名组员，所幸无人受伤。
