definitive diagnosis requires catheterization to inject some stimulant to
induce coronary spasm, which is not done very often. In practice, most of it
are diagnosed by a thorough history taking. Prinzmetal's usually respond to
nitrites during attack. If it does not respond well, then he should go back
to the doctor to have another look.

我读了母其文先生《关于所谓“放弃45万美元年薪，留美博士南充“安家”
的回复》不禁用功细读了几遍细想了几通，仍有诸多不理解和不敢苟同之处。我
虽认为自己对美国社会颇知一二，且不时给别人发几个 offer letter, 而今读
母文却是万丈的和尚别说头，连脚趾甲都摸不到了。请允许我一一列举心中不明
之处：
1. 母其文先生用了那么多的排比句辱骂 Fred 有欠妥当。Fred 是在反驳
《南充日报》的报道，并且提供该报文章的网址，Fred 并没有编造。胡说八道
该挨骂的应该是那个叫陈梁的见习记者。至于陈梁再去骂谁，还请母先生明断。
2. 我不赞同母先生把诸如“异国他乡丢脸啊” “中华民族是个伟大的民民
族哟” 之类话题扯如这种一般性问题的争论交流。
3. 我不敢苟同母先生对 Fred真名实姓的索求。他的真名和我们的争论无关。

以没做CT。就当dehydration治了， 结果大量输液，全部third spacing到腹腔。
他们当时可以CT without contrast, AXR, 哪怕一个稍微认真点的，physical exam 都
检查得出来， 13L 啊！这么多积水造成compartment syndrome, which blocks the
renal perfusion, creatinine 怎么能不高？
这里有很大的疏忽和失误，所以他们现在仔细很多。。。
医院和医院差距太大了。。。

用的protocol是４％PFA perfusion fix, overnight post-fix at 4C.
70%EtOH x 2, 95%EtOH x 2, Butanol x 3 (several hours, then overnight, then 3
days)
paraffin x 2
固定应该够了，难道是脱水的问题？protocol是实验室以前的，现在没有人做这个．
不知道这儿又没有人经常切能有些经验．谢谢． |

so bright and GFP is so weak, hard to tell...not sure whether my sections

理论上，最接近生理情况的，应该是体外perfuse后，flash frozen，对吧？

donor bone marrow. Then perform vessel injury by denudation or ligature(
possibly by perfusion ischemia).
我还是没明白二者在根本上有什么区别。一种方法，不用放射性，直接注射BMMC,做动
物损伤模型。 另一种方法，就是我用的方法，用放射性，注射的也都是donor的骨髓（
全骨髓）没有在体外分化的，然后做动物血管损伤模型。既然第一种方法也挺好，为什
么我老板让我用第二种方法，多花6个礼拜的时间等donor bone marrow repopulate
into the recipients bone marrow site?
你明白我说的吗？我自己都懵了 |

gene. 目前得到了Cre Hetero/flox hetero还有Cre homo/flox hetero.
请问如何检测knock down efficiency呢？
1. take forbrain, extract genome, RT PCR
2. perfuse brain, immunostain brain sections with antibody against my gene
of interest. Can I costain with anti Cre antibody? If so, where can I buy a
good one?
3. DAB staining brain sections using antibody against my gene of interest.
请问一般Cre 的knock down efficiency 是多高？ Cre homo or hetero 会有很大区别
吗？

老鼠用DiI perfuse，然后flatmount。
retina has three vascular layer, one is the most superficial layer where
lies the veins and arteries，the second is the superficial capillary bed and
the deepest is called the deep capillary bed.
Enjoy.
When I see all X'max lighting trees, I always image them as vascular trees
with mural cells on them. lol |

实验室学生切的片子，是PBS perfuse的小鼠脑子，然后在4%PFA 4度过夜fix，然后放
到pbs里，室温切片，40UM厚。我第一次觉得片子太厚了，用了漂片法，荧光染色，染
完了再mount到slides上，效果还马马虎虎。可是老板不喜欢荧光，喜欢DAB染色。说是
同时有人的脑染色，人脑用的是DAB，鼠脑也要用DAB才行。我也不知道是什么理论。我
只好又做DAB染色。可是我没做过DAB，完全照实验室的protocol做了两次，没做出来。
用的是贴片法，我想是不是片子太厚了，所以染不出来。可我老板偏认定是抗体浓度的
问题，让我继续试抗体浓度。我觉得真不是浓度问题，因为啥也没出来。
这个实验室染色用的protocol和我以前lab的很不一样。我们以前都是pbs+PfA灌流，然
后fix过夜，蔗糖沉降，oct包埋冷冻切片，从来没超过10um厚，然后荧光染色。这个

following procedures like IHC.
I have never done on livers. What I may do, is to perfuse with 4% PFA and
post-fix in 4% PFA for a bit longer. |

For example, you have mouse sections, and your primary antibody is rabbit
anti-mouse
for your sample: dilute your primary antibody in your incubation solution (
normally PBST + 5%-10% horse/goat serum), followed by washing. then your 2
nd antibody will be horse/goat anti-rabbit antibody in incubation solution.
For your control: just use your incubation solution without primary antibody
