phosphate buffer (pH around 7) after overnight storage at 4 degree. The
precipitate will be soluble again when warming up solution to room
temperature. Addition of NaCl will prevent precipitation. Other common
buffers are OK not causing precipitate.
Hypothesis is ionic cross-linking. Phosphate ion has intense negative charge

before u propose protein-ion interaction, I think u should make sure the
precipitation is ur protein and it is phosphate-dependent.
use HEPES as buffer and add series conc. of Pi to test dose-response. Only
difficulty is to quantify protein precipitation, but u could spin down it
and determine protein conc in supernatant. |

cells grew by several-fold without phosphate or arsenic. Can I claim that a
special plant cell culture can grow without phosphate, only five elements
needed. The real trick is that the plant cells store a lot of phosphate in
the initially-tested cells. The NASA paper may have the same situation.
Some one has commented this possibility in the same issue of Science.

最主要的是As妨碍普通细胞的呼吸oxidative phosphorylation，普通细胞没法在As下生存，不可能繁殖
但是这种几年前就被发现了的细胞有酶可以中和As，还可以靠As呼吸， 所以可以靠As生长，所以在用As替换P的环境下繁殖，就丝毫不足为奇了
所以更有意义的发现不是As替代P， 而是细菌可以在As里繁殖。这是个老消息了，可怜NASA不要脸，又玩花样骗钱
早就说过了，NASA是个很可怜的机构，在资本主义社会中没有任何存在的意义，所以要向好莱坞学习，不停搞出新闻来，才可以吸引缴税人民的注意力，搞研究经费

你给的link只指出bacterial phosphate transport system (Pst and Pit)能够将
Arsenate import到细胞内,很可能负责import的酶无法distinguish Arsenate and
Phosphate,但并不代表所有酶都区分不了,也没有证据表明Arsenate能够取代phosphate
加入到DNA(首先是nucleotide)和蛋白(比如phosphorylated protein)里.
至今没有在DNA和蛋白里发现Arsenate,有可能是你所说的Arsenate的对大多数生物的毒
性作用,导致其死亡.但是也有可能是砷原子的原子半径和化学性质和磷的不同导致的.
至少你给出的证据没有否认这一可能性.
有一点很奇怪,既然你说在As中繁殖的细菌早就发现了,为什么没有关于这些细菌中DNA
和蛋白质是否含As的研究呢? |

我们实验室现在一般不用phosphate，直接用H2O+NaCl,然后pH the sample,据说得到的
谱图比较好，所以我从来没有试过加phosphate。
不过这样做可能的确有问题，pH 也不太准，我试试用5mM的phosphate，不加盐离子看
看吧
sample concentration 大约0.9-1mM.
请问这个浓度能做natural-abundance N15-HSQC么？
