看了几篇paper，是关于potassium depletion的。 其中potassium depletion buffer
的地方，我不太明白. 几篇paper中，buffer的组成是：Hepes, NaCL, CaCl2, MgCl2,
glucose (PH 7.4)，对于不同的paper（都是HEp-2 cells），组成和concentration都
不不一样，比如有的不含MgCl2 和glucose，但共同点是都有Hepes这个buffer。其中一
篇paper解释加入CaCl2是因为他们实验中的某种protein binding需要Ca+，但关于其他
成分，没有说明。
我的问题是：如果我做一个不同cell line的实验，那么这个potassium depletion
buffer我应该怎么做？比如能用PBS代替Hepes吗？要怎么加入各种成分才能最大程度上
保持和普通实验的一致性呢？（普通实验就是incubated with growth medium， RPMI
1640）。

希望各位对PBO聚合了解的给我些建议并帮我分析一下我问题出在哪.
我想通过n-butanol与 potassium naphthalene THF 溶液反应后的O-K+引发BO的开环聚
合。试验前所有试剂都蒸过，并且potassium naphthalene 溶液也是在手套箱里配置的
，所以试剂中含水量多肯定不是失败的主要问题。
还有配制的potassium naphthalene溶液浓度不是百分百精确，所以无法最后保证OH完
全变成O-K+，但是我尽量让OH过量些，形成的OH/O-K+混合引发剂。因为最早这个反应
就是用OH/O-K+引发的原位聚合，但是非常慢。后来就有人改用溶液聚合，并将OH全部
转换成O-K+。
接下来是我的实验过程了：
干燥烧瓶抽真空通氮几回，注入n-butanol与 溶剂THF，然后注入potassium
naphthalene 反应，反应很快，墨绿色的K盐很快消失成无色，并有白色沉淀析出（O-K
+在THF中溶解度小），十分钟后注入30倍量于n-butanol的BO聚合（希望得到BO30，分
