dealer service 检查的结果出来了, 说是油燃烧生成的碳的积累, 导致什么东西坏掉
了, 要换个throttle body, fuel injector. 收费239+tax.
由于3年的保修昨天到期,今天就不算了,正跟ford 公司和dealer协商, 他们会review这
个case, 要4个business day才给回复是否保修这次的问题.

我05年的时候，买了个03年六万五迈的malibu（以前是budget租车公司的，可能这个把
车耗得特别狠吧）。
买到手没多久就觉得刹车抖动得厉害。送去修车铺检查，说后轮的刹车鼓有问题……换
了，大概三四百吧。
第二年，某日车喇叭忽然长鸣近一个小时，随即哑了。送到车行去检查，先花了100块
做电路probe。probe结果是喇叭坏了，修好要一千五，重新拉一条线装个新喇叭则只要
两百。于是装了个新喇叭。
第三年，开车去大雾山玩，发动机在半山腰过热冒烟，害得我们狼狈到极点。幸好当时
有好心人过路，一个在Autozone上班的老兄教我们把暖风开到最大，或可避免过热问题
；另一位则一直开车跟着我们，说如果我们的车再死菜他就带我们去山脚的旅馆。到了
地方之后首要任务是修车，修车铺说发动机里的温度sensor坏了，趁机宰了我们五百刀
。结果大雾山三天全部窝在旅馆，哪里都没去成。
第四年，空调又出了问题，漏氟利昂。忘记修掉多少钱了。
……
一年一年下来。这车大毛病倒没有，发动机和变速箱还算OK，但是小毛病没断过，七七
八八修下来也修了好些钱去了。

我从种子提nuclear extract, 想做EMSA, 用的是DIG-Gel shift kit. 开啥有了
TATABOX （5 repeats) 25bp in length, 试试看蛋白是不是有功能的，结果没加蛋白
的free probe也有带，所以认为我那加了蛋白后产生的带不是真的binding，因为2 带
的位置是一样的。但是有些情况用其他大一点的探针，50bp, 会得到smear. 还有我跑
电泳的胶很小，8x8cm（bio-rad)， 5% native gel. 0.25x TBE buffer, 4C, 80V.
问题：
1）怎么能证明蛋白是用功能的？现在没有binding， 不知是不是蛋白的原因，SDS-
PAGE 检测蛋白还行。抽的几次都是结果差不多，尽管有些带很强且集中在2个区段，但
总体来讲，蛋白的分布大小都有。
2)能不能找个其他probe 做postitive control。 有人可以推见一下啥探针？
3）我是用的nuclear extract, 探针可不可以用200bp 长的。
4)调整些啥才能有好 |

我答的是这样的，请大家指正。
我谈了做实习时给图做优化，sign extension的问题。eda的东西，他听了似乎不是太
有兴趣。
hashtable O(1) access time， no order，通过key来map
bst O(logn) time, but can retain the order of stored item
我一开始说bst是balanced tree. 他指出不是。又问bst不balance会怎么样，worst
time analysis. 以及有啥方法balance. 我跟他说avl tree 或者红黑树，但没要求写
具体代码。
接着还追问了hash collision怎么处理，我说可以弄个list append上去，或者probing
(然后稍微解释了下probing)
我首先想到的是debug statement的副作用，可能里面执行了什么函数。其他我说想不
出来。他说加了一行code会改变什么？我说executable大小会改变，load到内存位置也
会不一样。我说可能是内存某一块坏了，刚好load到了那块。接着他问还会改变什么？

我是即将毕业的博士生（第五年），做的方向是RF/microwave的硬件。具体做过THz的
Probe，THz的测试，以及相关的高频器件。
几年前流过一次片，2.4GHz的RFIC（LNA+MIXER），不过不成功。所以board level
design没做。但是课程的project吧最后的这一步补上了。给定chip做两种放大器（LNA
, small signal），根据chip的s-parameter用ADS设计不同的input/output matching
network。
RF on-wafer test做过， probe station也都用过。RF的器件的测试懂一些，带过TA，
基本的gain, IIP3, NF, P1dB, s-parameter带着小本测了一学期。。。
Sensor以及sensor外围电路也懂一些，我的探头就集成了自己设计fab的strain sensor
，用来测定探头的接触力和接触角度。
Semiconductor fabrication经验还可以，大概四五年clean room experience了。常用

1）Broan E60E42SS 炉罩（这个是不是也就包括了15楼贴图里的hood flue了？）
2）Broan 336 1500CFM 外置blower（用这个就不用再买roof cap了，是吧？）
3）MD8TU 补气装置（这个就包括了pressure switch，probe了，是吧？然后pressure
switch，probe是安在抽油烟机附近，damper要安到别的地方，是吧？）
4）exhaust duct，跟MD8TU配套的应该选8寸的管道是不？是不是买MD10TU的补气装置
加10寸管道效果更好？还是说没啥差别，就用8TU和8寸管就行了？
一共就这些了，是不？

工作上的事情，我是薄厚，工作上要用一个仪器，今天叫管仪器的人帮我换了一个
probe,因为老板要求做一个实验，很简单的，在电脑上敲了几个命令，都是平时用的，
用完就走了，突然下午受到关仪器的人的Email，说要ban me, unless my supervisor
talk with her.
我太惊讶了，因为我只用了最简单的两个命令（专业上的东西，学有机的mm知道）测得
谱图，它的仪器我什么都没碰
这个关仪器的也是个妈妈, 可是平时对我说话就特别挑剔，在换probe的时候要我把
sample 拿出来，我告诉她sample is ejected, 可能口音不好，被他反问eject 还是
inject。总之觉得她看我不顺眼，老是挑毛病。有一回她周末测了3天的结果不知道怎
么弄没了，tmd 第一个打电话给我，问我动了没有。我那天都没用那个仪器。
真是烦。在这里，博后真是受欺负。做这个仪器都作了8年了，第一次碰到。 |

1.probe是不是就合成目的基因互补的一段单链DNA或者RNA? DNA好还是RNA好
呢?
2.probe的label是自己用试剂盒做么?看到很多人做地高辛标记,但是我们实验室要做
FISH,荧光标记是自己标还是公司可以合成? 如果有好的公司,可否给个链接,谢谢.
3.section是冰冻的好还是石蜡的好?要不要蛋白酶K处理?

这俩各有侧重，很难互相取代吧。尤其是 NGS 的数据在 accuracy 上还远不能和
microarray 竞争。不说别的，shotgun of RNA 从原理上就使得其测试结果有很大的
bias，使得测 isoform 时不如 isoform array 精确。而 sequencing depth 又直接决
定了 alignment 的可靠性。对于低表达的 RNA，远不如 microarray 更靠谱。
NGS 的作用是 explore for novel transcripts。定量还得 microarray，又便宜又准
。所以我觉得 NGS 的大量使用反而会进一步促进 microassay 的 probe design。
学术界喜新厌旧是常态。但是要想得到靠谱的结果来指导实验和 hypothesis，

PARIS — France's government on Wednesday asked a health watchdog to carry
out a probe, possibly leading to EU suspension of a genetically-modified
corn, after a study in rats linked the grain to cancer.
Agriculture Minister Stephane Le Foll, Ecology Minister Delphine Batho and
Health and Social Affairs Minister Marisol Touraine said they had asked the
National Agency for Health Safety (ANSES) to investigate the finding.

You may have vague memories of this guy. He went from New Jersey Governor to
Obama bundler to a guy who made over a billion dollars disappear. Then the
media completely ignored him since he’d make their God King look bad during
his re-election campaign. Well, at least some financial regulators won’t
