scv比drone和probe多5血，因为drone边打边回血，所以都可以被打9下，probe的护盾
只有在不受攻击超过5秒的时候才能回复，所以只能抗8下，不过scv和drone单挑，除非
scv明显先出手，不然drone胜，因为最后一下攻击，drone伤害判定比scv快
probe单挑其他农民其实很简单，等护盾没了就跑，回满了再A回去，打SCV完全不亏，

targetting DNA. Generally I believe the concentration of your probe should be
enough for the detecting because 10ng/ml of a 16mer oligo aproximately equals
to 1.5nM by molar concentration, which could capture 1.5pmol targetting DNA if
it bound at a ratio of 1 to 1.
To label or not to label, it depends on the method you used to detect the
targetting. If you use, for example, biotin to lable to probe, then label the
probe |

making non-radioactive probe incorporate only a small unit, like a biotin,Dig
or fluorescein into the probe via UTP or others instead of directly incorporate
the enzymes for detection,like HRP, AP. Since it is not easy to keep the full
enzymetic activity during the long experiment procedure.
So if you use klenow to label the probe, you can deactivate the klenow by
heating the reaction at 75C for 20min in presence of 10mM E |

DNA-protein complex. 400 bp probe is a lot big. I quite agree with the
suggestion of adjusting your gel concentration or Acr/Bis ratio. I think it
might be easier to use the probe only to optimize the gel first, at least to
let the free probe run to the bottom. You don't need to label the DNA. Instead
you can use EB to stain the DNA. To my personal opinion, I think 5% gel is not
the best choice. Gernerally I believe 3% |

probe. Could have some reasons for this, first of all, probe denaturing and
annealing is not good, I bought oligonucleotide, because it is highly
purified. the free probe is also smear which is about half length of the
smear with protein.
secondly, when I extract nuclear protein, I just use 250M sucrose in the
extraction buffer for nuclei and no further gradient centrifuge thereafter.

1.可是在做竞争实验时，加入400x的unlabelled probe （与biotin labelled probe
探针一样） 就是看不出binding complex 减少的迹象， positive control 都 很好。
我用的是 LIGHTSHIFT 试剂盒。
请问是不是 nuclear extract 太多了，过量了，导致足够的factor 剩下后给label 的
probe binding。
2. 形成的binding complex 总是跑不下来，一直就是在最上面， 担心都没有地方做
super shift 了。

首先，这两者的理论sensitivity的确差不多，而且在某种程度上来说，Sybr green因
为信号强度大，其实在检测上更容易被仪器看到。不过，你要是真的两者都做一次你就
知道了，对于含量特别低的样品，Sybr green的信号强度根本没有什么用，因为不同重
复孔跑出来的差别太大了（类似楼主看到的这个问题），导致那些跑出来的常常都是废
数据，所以真实的有效敏感度没有那么高。
另外，你可能对Taqman 的特异性的理解有些偏差。那个fluorescent probe, 要产生荧
光必须有两个条件：1，必须在template 上有PCR reaction 在进行； 2。probe 必须
能够结合在template 上面。必须两个条件同时满足，缺一不可，如果仅仅是probe非特
异结合是没有荧光的。
但是对于Sybr-green, 只要满足第一个条件就可以有信号了。
这样一来，Taqman qPCR 和Sybr-green 的特异性向比较就类似于Nested PCR 和普通
PCR的差别，前者在理论上肯定比后者更特异。

感觉应该是，否则管他热传导如何干啥，直接给dunk到液氦里不就得了？所以我觉得他
是应该是在设计一个真空probe，并且可以用heater和少量exchange gas来控温。这个
需要有一段比较绝热的材料把样品托跟环境隔离开。
一般来说，用铜做样品托，用brass或者不锈钢做insulating的那一段。密封方式有
probe头的indium密封和cone密封，还有采用长套筒在probe顶端用一般的o-ring密封。
当然，还有一种方式是采用控制He vapor的温度再加上一个样品的heater控温的，这种
情况倒是不用密封的probe，但是可得到的温度范围在液氦温度以上。 |

波士顿，8月27日，又一件华人社区的大事： 来自波士顿当地，麻省各地以及邻近的华
人同胞在这里聚会，抗议亚裔细分。
为了我们的合法权益，为了我们的孩子，为了我们的未来，8月27日，数百人聚集在波
士顿，举行了一次抗议亚裔细分的集会游行。 这是两个月以来规模最大的一次抗议亚
裔细分的集会。
