amplified DNA has similar sequence that can hybridize with the probe. It
really depends on how the probe is designed.
As for Sybr Green assay, it is necessary to check melting curve every time.
If any non-specific amplification were present, the curve wont be a single
peak.

我做In situ hybridization，设计了3对probe（500bp左右），3对proble都是发现
sense和anti－sense有相似的pattern。
我一开始觉得是non－specific binding。后来越做，我越觉得signal是对的。我做了
transfected和non transfected cell pallet，只有transfected有信号，而且sense和
anti－sense都有信号（sense弱一些）。
因为我的基因有的tissue表达，有的tissue不表达。我又多做了很多tissue，发现我的
probe有表达的tissue才亮，说明probe是好的，看起来信号也挺好。
可是怎么解释sense和anti－sense都有信号的问题？有ISH的大牛能帮忙trouble
shooting吗？ |

the surface of Mars on Wednesday has been imaged by an American satellite.
The Mars Reconnaissance Orbiter (MRO) has identified a large dark patch in
the robot's targeted landing zone consistent with a high-velocity impact.
Schiaparelli is widely thought to have crashed and been destroyed.
Data transmitted from the probe before it lost contact indicated that its
descent systems did not work properly. |
