ug 呢，还是指针对一个 signaling event，还是针对一种 bio-function？
我过去一直理解为，能跟某种 ligand 的 receptor binding 的东西，如果跟这种 li
gand 引起的作用相同，并且作用的程度类似或者更高的，叫 agonist；如果 binding
但不引起后续的 signaling events 的，也就是说，block 了 receptor 的，甚至引起
相反的作用的，叫做 antagonist；能引起类似的效果，但是不如 ligand 有效的，叫做
semi-agonist，又因为这种 semi-agonist drug 和 ligand 竞争在 receptor 上的 b
inding site，如果 ligand 过量而 receptor 不足时，或者这种 drug 的 affinity 高
到严重阻碍 ligand/receptor binding 时，这种竞争就会导致 signal 减弱，所以这时

做这个报告的是Robert Margolskee, MD, PhD
我这里简化、普及化一点。
taste receptor，就是辨别各种味道（比如甜味，苦味，酸味等等。这里就只讲甜味了
）的蛋白质。舌头上有taste
receptor（地球人都知道），但是不光舌头上有，肠子里也有！肠子里面的L-cell，一
种分泌荷尔蒙的细胞上面发现了
taste receptor，如果有甜味物质刺激的时候，会分泌GLP-1 (glucagon like peptide
-1)，这个荷尔蒙会刺激胰岛素分
泌。进一步的研究发现，不光肠子里有，胰岛里也有，有甜味物质刺激时，会分泌胰岛
素（促进糖，脂肪酸uptake，当然

就行了 简单来说就是搞一个没有receptor的cell line，把有receptor表达的cDNA
library转进去 然后加ligand，看下游的phenotype就行了 可以是binding activ
ity，也可以是downstream signaling,等等
2 没有 但是如果你有个范围可以大概用agonist/antigonist猜猜看

抑制作用？

因为Olfactory receptor也算G protein-coupled receptors这个大类的，是不是也可
能和比如听觉或者别的sensory有关系？

我现在在做DR internalization, 试了好几种办法，
不管是Membrane biotinylation,
或者是用Flag M2在没有fix之前去label外膜上的receptor
或者是用Radio ligand来做，
都不是很成功，最近试了一下用Sucrose gradient (35%)来分heavy membrane 和light
vesicle, 应该是有看到了一些internalization.
不过最大的问题是用ＦＬＡＧ Ｍ２做完ＷＢ后看到的并不是一条单一的带，而是一片
的smear,
应该是某些地方没做好，所以Ｄ２Ｒ都聚集了
用的是Biorad的ＳＤＳ loading dye 配方直接溶解膜
或者也试过ＲＩＰＡ和ＴＢＳ＋１０％甘油＋１％triton
效果都不是太好
但是看了不少ｐａｐｅｒ貌似人家都是做得出来了
这个事情搞了大半年，一直没能搞定，老板又特别楞，从来不给些建设性的意见，每隔
一段时间都要威胁一下做不出来就开除，已经被威胁了十数次，真是有点窘境穷途了,,

这个receptor occupancy是在 clinical pharm area. 个人觉得偏临床的receptor
occupancy工作是不是好些阿？
问了几个身边朋友，有人觉得receptor occupancy很乏味，每天做routine的工作，PD-
1类型的工作应该更有挑战性，但是我担心工作的稳定性。

1. iv is more for acute reponse. ip is slow but maybe last longer. it
depends on what receptor you are looking at and what tissue are they located
2. somebody probably has done that, or the data sheet of your mAb suggest a
starting dose to try. you might need to do a escalating dose to find out the
optimal dose
3. again it depends on your specific receptor, their turnover rate, the
halflife of your mAb, etc...

I finished the step crosslink and IP, and sent the sample for MASS.
But to confirm the receptor is the protein reported by MASS, still need a
lot of time.
I prefer the method you recommended, transfer cDNA library to a cell line
which does not express the receptor. Screen the positive clone. |

它的protein level下降了，但它其它的磷酸化形式都没有变化（WB结果）。根据其它
的条件，实验中的cell migration应该增加，但我查了很多文献，几乎所有的
reference都说这个non-receptor tyrosine kinase的protein level下降的话是因为
cell migration减少。
难道是我的观察有误吗？但实验重复来重复去，结果都高度一致，真是想不通啊！上来
问问大家，看看有没有遇到类似情况的？ |

It seems 2010 FA is correct. But UW has an explanation about the target
organ of beta-2 adrenergic blockade is glucagon secretion. Kind of confused.
