Amgen 公司试图开发的单抗药物宣布失败。该药品是抗 type 1 insulin-like growth
factor receptor，在细胞和动物模型中有理想的效果。而且，在动物
和细胞模型里面过表达IGF-1会导致癌细胞行为并能被该抗体抑制。
但是在phase III clinical trial中得到的数据表明该药物对病人身上的
多种癌症都没有疗效。
其他类似的单抗药物目前看来效果也不理想，研究者们认为大部分癌症（尤其是固体瘤
）的机理比较复杂，用单一靶点可能是很难抑制癌症的。而且在初期研究中使用的癌症
模型也不能被视为是真正的癌症例子。
另外几个公司目前在试图把 IGF-1R 和 insulin receptor一起阻断，看看能不能得到
更好的效果。

受体亚型receptor, NMDA和AMPA receptor. Glu gate 钠通道还是第一次听说， 猜你
是想说Glu receptor gated 钠通道。 但是Glu 有三种受体亚型， AMPA, NMDA和Kainite
对应受体介导的离子通道都不一样。

may via D1 receptors stimulate "Go" neurons in the striatum to facilitate
the section of "Go" action in rewarding behaviors;simultaneously via D2

ligand要通过polyvalency来提高apparent kd，就要ligand和receptor的
positioning和density恰好match，像齿轮一样嵌合，才能有效。可是ms
在NP上做polyvalency的人很少会去根据receptor的具体情况做一堆计算然后
决定如何incorporate ligand，polyvalency带来的好处也是从几倍到几万倍
不等。我的问题是，到底有没有组做rational polyvalency design？理想
状态下（比如3个ligand跟3个receptor同时完美结合），polyvalency对
apparent kd的贡献有没有个公式？最后如果可以很容易的通过polyvalency
提高kd和specificity，为什么还要强调antibody/aptamer的kd？搞几十个
phage display选出的peptide行不行？

GPCR的C terminus都是结合arrestin重要的位点，磷酸化与否是arrestin结合的关键因
素，虽然有些例外。receptor和arrestin结合不稳定并不是指affinity，而是整个过程
非常的dynamic，传统的观念，也是这篇文章的point，就是arrestin结合C terminus，
然后再构象变化结合到G protein结合位点上。但实际上，很多receptor结合arrestin
但是不internalize，或者结合arrestin但是没有arrestin signaling，这些都不是这
个简单的模型可以回答的，实际结合过程可能更为复杂，不是一个单一或均一affinity
的状态，甚至还有可能需要其它蛋白的帮助。如果真affinity很高，这篇文章里也不会
把他们fuse在一起。另外lipid肯定是有作用的，很多细胞外研究他们相互作用的都要

这类东东可以是天然的（植物激素）也可以是合成的（制药公司疯狂再删选的东东，市
场价值大到吓人，因为涉及乳腺癌，前列腺癌等等）
机理太复杂，同一种分子，可以在一种组织细胞中是拮抗（譬如说雌激素），再另一种

经过一个中间宿体性的其它动物才能传染给人。所以，病毒发生后，就故意误导全国人
民去海鲜市场找证据，找其它野生动物的麻烦。 而且还是病毒所去找的，找完了还装
模做样化验呀，分离呀什么的。最后把责任全部推给了海鲜市场的动物。可是那种动物
，一直不敢说，说了其它相关已经在人就会去早那种动物监测。 所以压根不说，打马
虎眼。
事实上，早在2013 年，就是这个武汉病毒研究所，已经从来自云南的蝙蝠身上所携带
的corona virus中分离出第一株蝙蝠SARS类似样的冠状病毒的活病毒，其中就包含了类
似于S类型的基因。从而证实这株病毒能够使其接受和SARS病毒相同的受体，并能够感
染人的细胞。对此新发现，武汉病毒所还把它以武汉病毒研究所的英文简称命名“WIV1
”，以彰显这一发现的重要价值和属于自己第一个发现的巨大研究成果。这个成果刊载
于2013年11月的《自然》杂志。
