：有的人自己看书效果好，有的人听讲效果好，有的人看电视片儿上网课效果好
：不论哪种学习方式吧，真要学通学懂了，一个人必须用心用脑要思考
你说的在理。
但另一方面，人心理学 PHD 也得灌水 paper 数量一到就毕业不是？反正心理学业界
一半以上的心理学实验 not-reproducible，你总不能让人 PHD 不能毕业做一辈子千老
不是？有没有良心你。。。
反过来说，马工这么较真，以至于测试组报个产品有 10% 概率的晃点率，马工立马一拍
桌子问你丫是人工肉测还是自动机测。人工肉测的话，测试工丫的手滑没？自动机测的
话，
机器人 script 和 log 先拿来 review，云云。。。这看起来特么的不近人情，但这其实
是马工职业为老板所迫歌，一样都是为了混饭。
这是因为马工的实验太可控了，概率错误也能做到让机器人在 sever farm 放羊跑
一万+次回来，在概率上可控。这还不算，一万+个数据点还可以 data mining 找各种
pattern。这样万一你不怀疑测试人肉工手滑、或者测试机器工逻辑漏洞，而别人怀疑
了、

背景比较弱，一年之前citation只有70，所以当时交了NIW，想着如果批了，家里ld可
以H4 EAD。结果NIW十个月也没有消息，这时候citation 涨到大概90，受了版上一些弱
case的鼓励，于是决定试试Eb1a。
背景：化学
publication: 9篇，会议五个，都是poster
citation: google schoolar 97，不区分自引他引。
审稿：30
从今年六月开始着手准备，推荐信就用了NIW的推荐信，有两封信改了改找教授重新签
了一下。结果有一位教授没理我，只好重新找推荐人，好在比较顺利，骚扰了几个教授
之后，有一位同意了，赶快打好草稿发了过去。最终推荐信六封，一个来自德国，一个
来自波兰，四个美国的。Independent的有四封。
PL花了很多时间和精力，看精华区各种帖子学习，按照mit的模板又重新写PL，毕竟NIW
和Eb1a还是有很大的区别的。

the same as your protein.所以难以用来测蛋白的绝对浓度。我见过Bradford结果和
蛋白实际浓度相差2、3倍的。
感觉楼住的问题是要reproducible results.nanodrop在这点上有时候是不怎么的。。
其实bradford 或者其他方法要reproducible results的话，都是可以做到的，当然要
正确操作以及稳定的仪器。但如果要精确定量蛋白的绝对浓度的话，我觉得这些方法都
不是很好。UV-vis是相对好些的方法，它的assumption是，你的extinction
coeffecient计算正确（通常网上一些工具的结果还是可以的）。还有就是蛋白纯度要
好。这样虽然也有误差，会比bradford好很多。
当然，前面提到的amino acid analysis最好了，不过要特殊的地方才能做。
所以，UV-vis (A280) with calculated coeffecient可能是比较方便的，比较折中的

general...there are clearly exceptions...besides what LZ and some already
pointed out, there are also reproducible junk...because people try to fit
data...or "some visionary guys" basically have a hypothesis which is likely
right and then fit data..it is also a way bull lab screw small labs with
their sloppy but happen to be right data/conclusions cause they could get
through the reviewing process... |

that's completely different from real results,
but difficult to reproduce due to technical issues.
since the PI is supposed the top expert
in the field, if she can't even reproduce
the same results in her lab;
and found out the figures are different
from the raw data, what can you say?! |

That is good. What is your concentration of CHX? you can try 100ug/ml,
although most protocol say around 50ug/ml
What is the concentration of your gradient, w/v or w/w? 15-45% w/w gradient
and 15-50% w/v gradient work good.
How did you make the gradient? Usually, a gradient marker (machine)
generates highest quality and most reproducible gradients. If no such
equipment available, you can use freeze-thaw method to make gradient.

Paper's quality is evaluated by its reproducibility.
- reproducible research
I have reservation on open source everything (e.g. software), besides that
it is difficult to define what the 'raw' data are. As long as the results
can be replicated, it would be a good paper.
- simplified paper format [less time in writing paper, more time on doing
the actual work]
- easy search [tag, keywords ...] |
