Hi,
I tried a couple of transfections with the NgAgo plasmid from Addgene and
the 5'P oligo against GFP from the paper, but I did not see a dramatic
reduction in GFP+ as seen using Cas9 and a GFP sgRNA.
Anybody tried as well and came to the same (dead) end?

under supervising, he demonstrates he can reproduce the test.
It is dangerous for people to draw conclusion that the result is faked since
70+ cannot reproduce it. The rest labs (80 something) may have some kind of
results that might show some promise. They just want to keep the secrete to
themselves for the benefit of priority and intelectual rights. The labs,

- 还有, 用microSD卡放MP3, 有时一开车就马上可以听, 有时刚停5分钟, 打火后车要
"reading from memory card" 一段时间才能听. 这又是什么原理...

能开到30miles以外的现场去试,附近的坡开到五十mile没法reproduce.
然后dealer说车子高速下坡松开油门加速的时候电脑会自动降档来减速,我自己不会踩
油门啊?
每次自动降档后,转速急剧加快,车子震动发抖,更糟的是到了平底以后档位还不给我升
上去,开上相当长一段路后才会给我想起来应该上五档. |

既然是说故事, 首先来介绍一下这位女主, 她叫小保方晴子，31岁，日本千叶县人士,
1983年6月29号出生, 巨蟹座, 2006/2008年早稻田大学应用化学系本科/硕士，2011年
早稻田大学生命医学博士。其中2009-2010年其博士研究是作为访问学者在哈佛大学医
学院进行的。2013年开始以30岁的年龄，被任命为日本理化学研究所（RIKEN）细胞再
造实验室（Lab of Cellular Reprogramming）的研究团队负责人。不是内行也能看出
，这种简历优秀到顶天。 这么逆天的简历也就算了，晴子酱还是个美女，当然美不美
这么主观的事情不好评价，总之她长这个样子。

发帖数: 24 | 43 9年经验的Testing + QA, 国内985本，美烂校硕，CS出身的大妈，给QA正个名吧。。。
首先，现在QA工作分2类，一种是automation，写代码，语言是TCL,Python，Perl比较
多，FLG找的都是这种类型的。面试就是刷题，比面马工简单，但基本功的要求还是比
较高的，基本上LC刷题几遍还是需要的，架构设计不需要准备了。
还有就是纯QA，这种要靠经验，各个领域相关知识要全面。比如做network的，各种
protocol要非常熟，TCP/IP,SSL这些，然后用python，perl写个client/server，做个
测试脚本这些都是基本功。
工作日常也不是前面同学说的照着测试要求按步骤输入这么简单，呵呵。一般来说有几
块内容，首先是reproduce customer bug，根据customer提供的有限信息在local重现
，这个听起来简单，其实很难，没有几年经验很难做好。专职做这个QA叫TAC，但是
Feature QA也经常要做，因为对feature内容比TAC更熟悉。然后是new feature test，

职位的主要要求是QE mindset和testing 经验或者兴趣，然后要懂基本的编程，如果有

. If you can reproduce the pain by pressing on your back, it likely comes
from muscle, tendon or bones, which is likely benign. If you cannot
reproduce it and pain persists, especially associated with breathing, you
