Because radioactive detection is very sensitive.
If you use cell-free in vitro translation kit, you will stick a lot of
ribosomes on the NTA-resin, and the unfinished peptides will show up as
smear on the gel.
What you need to do, is to use high speed centrifugation (at least 144,000
xg) on a 20% sucrose cushion to separate the ribosome fraction (go to bottom
) from the complete proteins (remain on the supernatant).
Then, you can use the NTA resin on the supernatant. |

如果resin没那么脆弱的话那就用第二种。
洗得干净与否在实践上和流速的关系不是特别特别的大，因为洗的方式和
洗的次数才是更重要的因素。如果你们对这个很重视，那就应该用比较小量
的液体，多次洗。 |

tagged transcription factor DNA binding domains. The protein were bound to
the beads with high efficiency. However, when I used glutathione buffer to
elute the GST-protein off the beads, they can't be eluted. I tried 10 mM ,50
mM and 100 mM glutathione. To make the buffer, I diluted 1 M Tris (pH 7.5

看看上面到底有没有蛋白.
第二，把resin 转移到小管里面去，加入elution buffer之后摇上10分钟，
然后再离心取上清。

己pack一个可以装在HPLC的柱子，用来分离血浆里面的albumin;还交代在pack之前，先
用IP Kit试试不同家的抗体，看哪家最好。
我的问题：这个pre-pack的柱子，有包含相应的 resin/bead 吗？若是分别买，怎么样
才能配套？还有,老板讲的IP需要做吗？还是直接买不同的抗体，和resin/bead 混在一
起，然后过柱子？有推荐的Vendor吗？ |

发帖数: 363 | 10 每个步骤都是别人都知道的,我只是组装了一下. 最大不同是别人是先Urea变性,提出来
再复性.我是利用Triton X-100可以洗脱SDS的特点利用SDS变性,然后Triton X100洗脱
复性,然后过柱. 肯定不会对所有蛋白适用,但是起码我遇到的都work (包括一个>100
Kd的酶).
我是用的sonication water bath, 也可以用probe sonication再加SDS,都OK,但是注意
dilution的比例和体积.

很高，电泳后蛋白大小正常，与理论值相符。
可是：
1. 和cobalt 或者镍resin混合后，混合物中出现很多块状物（聚集在一起，RESIN很难
沉下来，与溶液清晰分层）
2. 过柱子后，蛋白莫名其妙就消失了，看到的那些带比理论值小很多。比如，理论值
90kDa，那些小带只有40左右。
同时我们纯化去掉跨膜区的同一个蛋白，一点问题都没有。 |

purification变性条件来第二轮纯化（此顺序是实验需要，不要问为什么）。目前结果
是第一轮IP很好，但是His-purification后蛋白都在unbound fraction. 我用的是
Qiagen Ni-NTA Magnetic Agarose bead,据说适合纯化含量比较少的比如mammalian
cell的蛋白。IP后的蛋白都经过 buffer exchange，也没发现有与Ni resin
uncompatible的成分。请帮忙trouble shooting。会不会因为蛋白浓度太低导致与Ni
resin结合不好？His-purification有没有target protein concentration threshold?
是否需要改用binding capacity更大的Qiagen Ni-NTA agarose (不是magnetic bead)？ |

纯化拉下来。所以你的蛋白不能在如此多的背景蛋白中被看到。
如果你真的要用这个方法纯化蛋白的话，我的建议是：
1。用大量Hela 细胞，悬浮培养。
2。用high glucose medium.
3. 在你的蛋白上再加一个tag, 进行双重纯化。如果实在不能再加一个tag的话，也必
须至少在NTA-NI resin之后进一步用ion-exchange column + size exclusion column
进行再纯化。

比方说你应该可以用DEAE resin (positively charged) 来收集DNA啊。
比方说QIAGEN Anion-Exchange Resin。

cysDML and cPILOT, 相关工作在ASMS会议报道过。他们所用的方法基本还是基于传统
方法改良如Thiopropyl Sepharose 6B resin和Cys TMT，这些基于抗体和resin方法的
富集效率比较低，从鉴定数目就可以看到。我们的富集效率可以达到97%，很轻松鉴定
到10000+ unique peptides。
目前没有同时含有iodoacetamide+phosphate的tag。phosphate可能会被phosphatase酶
切掉，phosphonate不能，因此更加稳定。
至于为什么要富集Cys peptide,因为其low abundance,essential for the biological
activity and structure, highly sensitive and reactive.

任何加工方式现在基本都是电脑控制，怎么电脑就变成3d pr的专利了。我做数控这么
多年，还真没见过谁把这些雕虫小技用到实际加工的。
3D pr只是additive 中间的一个很小的分支，另外楼主把复合材料简单定义的好象只是
几种金属的混合，这也是复合材料其中最简单的一种。军迷们carbon fibor 总听说过
吧，请问3d pr怎么做贴片怎么做resin. 大多数CP不是单一材料的成形。很
多材料和模型只能靠切削加工。这就好
比东邪西毒谁都取代不了谁。很多工艺不需要也不适合做additive,特别是批量化生产
的情况下。另外精度也是一个问题，现代切削加工的精度可以达到纳米级别。考虑材料
热变形和表面热处理3d pr最多只能当玩具骗骗经费。不知道这里讨论的有几位在制造
业干的，学校里发垃圾文章的不算。咱们有点机械方面的基本常识好不好 |

不知道“制粒”是什么意思，如果是把回收的塑料做成resin pellets，再运到大陆，
连粉尘污染都替大陆解决了。美国公司不可能这么少，美国政府也应该不允许这么做。 |
