scaffold材料一般都会在表面裹上厚厚的matrix 吧。如果光是用个硬塑料的话大部分
细胞根本就不乐意往上长,而且就算长了也会被诱导成错误的细胞吧。

我现在有Cas9的老鼠,想敲掉一个基因,用HDR模版引入点突变。准备直接注射卵;因
为有Cas9老鼠,所以只需要注射gRNA+HDR template。
设计好了gRNA,是必须构建载体用T7加酶体外转录获得吗?
为了简便,我考虑直接合成gRNA+scaffold RNA。加起来有104 mer,是不是不可行?
这个长度的RNA能直接合成吗?是不是会很贵?

现在难得看到这么激烈的有关生物学的讨论了!
支持这个答主一下。不过结构生物学的意义多数还是很基础科学的,另外一方面,所谓
biomedical research大多数也是这样的,研究一些蛋白在疾病中的作用是现在NIH很喜
欢的,但是实际上多数的蛋白质并不能作为药物靶点。还有,结构是功能的基础是没错
的,但是知道结构对理解功能有多大的作用,就是case by case的事情。各种protease
enzyme的结构对于功能的研究显然是critical的,不然根本不知道为什么类似的蛋白
可以有不一样的底物选择性和不同的催化活性,这方面指导药物开发的例子很多,比如
HIV的各种逆转录酶的基于结构的药物设计,一些超大分子,比如叶绿体,呼吸链复合
物,yigong的splicesome,结构对于功能的理解是决定性的,比如结构出来之后大家才
知道呼吸链里电子是怎么传递的,各种小分子是怎么结合的。至于在细胞内做scaffold
的很多蛋白,结构对于功能的意义在我看起来比较的弱,同样还有各种跟免疫相关的比
如BCR TCR,精细的结构信息可能对功能指导意义没有很强。这方面看看结构生物学本

作为一个没做过膜蛋白的伪structure biologist,赞一下专业回答。
再补充一点生物大分子的。结构可能对于大分子比如抗体的指导作用没有receptor/
ligand或者enzyme/substrate那么看起来那么直接,但也有很大帮助。比如知道target
的结构,就可以有针对性地display更感兴趣的epitope,尤其是那些non-linear的
epitope单单根据序列是没法预测的。反过来说也可以突变掉unwanted epitope来避免
screen到不想要的抗体。拿到antibody/antigen complex的结构,可以根据结构做有针
对性的affinity maturation来进一步优化抗体。design bi-specific甚至tri-
specific antibody,需要考虑几个target epitope在空间上的位置。做vaccine的,可
以根据结构来design engineer的antigen,使之更有效地产生neutralizing antibody

You must be good at both execution of experiment and people skills.
Don't panic about the job.
HPLC is not rocket science, you just need a bit training and hands-on
experience. I have seen many ppl learned on the job and performed well.
Best way to learn is to search for established method/protocol in your
company. know your analyte and understand every detail in the method. This
will give you a solid foundation and scaffold to build your own mehtod. It

Piaget 和Vygotsky最大的区别是P认为人的学习是根据个人构建知识的不同而不同,外界
无法起任何作用,最多就是给孩子提供书,但是要他们自己去看(self-directed/initia
ted);而且在不同的年龄阶段孩子的思维能力是不同的(stage dependent);V认为人的学
习不能脱离社会环境,只是脱离了社会环境就没有任何意义(scaffolding),在老师的帮
助下孩子可以达到更高的水平(instruction dependent,ZPD)。(汉语太滥了,翻译的一

其实scaffold应该归到cellular materials的范畴,有很多东西可以借鉴cellular
materials的已经发展很好的理论
relationship between surface area and blood vessel density应该很早就有人作了吧
。他的新颖之处是不是在于老鼠in vivo, 我见到的研究angiogenesis 大多都是在鸡蛋
上做

Any big name or company in west coast? Stanford, Berkeley, Companies?
Right now, the commercial focus is at artificial skins. How about the area of
tissue implants? I remember some time ago, there was a small artificial liver
grown out of a microfabricated scaffold. Knew about this work?

Car's speed or fabrication speed? :-)
It is quit slow because it is based on 3D scanning method instead of 2D
planery fab techniques. But I believe with the advances of technology,
multiple scan head would be possible so the throughput can be improved.
Anyway, it is still too early to talk about production.
3D scaffolds would be very useful in tissue engineering.
Still in China now? :-)

具体的情况不太清楚,我觉得细胞有保护的措施的,例如包裹在
hydrogel 里面
活体指living 生物,如肾脏损伤,就在腹部切开一个口子,直接
在患处生成scaffold和cell 的三维混合体
感应器感应人体的活动的原理就如坦克在行进中瞄准目标,所以
不用怕手术中其他器官跳动的影响
但具他们说不同layer生成不同tissue结构还有困难

artificial vascular graft
的研究由来已久。看这段中文我猜大致意思是biodegradable
polymer作scaffold, 即3毫米ID的channel, 用“多微孔的膜”创造一个ECM,
这个"多微孔的膜“的膜估计也是porous polymer, hydrogel之类的,然后把EC注入,
不懂”肝脏或者肾脏的细胞注入“的具体理由,大概就是多提供点cell吧。
我想他最后还要把硅晶片抽出来。
光看中文科技消息,看不出来这个除了用个硅晶片外有什么闪光点。

当然,不然怎么“绝对可以作出一篇science的文章”。嘿嘿。我们用的是latex体系
,可以把growth
factor包在latex颗粒里,然后把降解酶也包在latex颗粒里,初期scaffold降解很慢,当
降解酶被缓慢释放出来后,降解速度就会显著增加。这样你可以把growth factor, 降解
酶,和ink放在不同的syringe里,就可以实现局部和时间上的调控.

最近看了一系列的相关文章,想和大家聊聊。
大体上大家伙都差不多,找种细胞,
找个scaffold,体外养会儿,植入体内看看,做一堆表征。
fancy点的无非就是加点growth factor(controlled release就更酷),
或者折腾细胞(机械锻炼以增强强度和细胞alignment等等)或者某种
3D culture设备(不是指boss的仪器,boss不要误会:)
总体感觉是现在的TE真是容易发文章,而且感觉总体内在科技含量都不高,
大牛也一样,not very scientifically sound,Harvard MedScool的
一帮MD的文章尤其惨不忍睹,那个stain做的差呀。
可是还是觉得TE是未来一个非常重要的发展方向。搞BIO最终不就是
为了治病么,所以个人觉得将来在这上面NIH和NSF会投更多钱,
不象drug delivery有大药厂的研发撑着。纯分子生物学,基因调控和
蛋白质表达最后都可以在TE上找到落脚点。
再就是Engineer在其中起的作用。常有人argue说现在什么BioEngineering,
不就是分子生物学和医学吗?其实是低估了en
