disrupt the dimer but failed. 因为
protomer不是整个helix在interact, 而是有一定角度的，所以不知道为什么两个

1.你的gel是native or denatured?如果是denatured,一般非共价键的homo-dimer会分
离成monomer.
2.似乎不太明白你的描述left lane and right lane哪个蛋白被过表达?
3.可以跑个简单的MALDI-TOF质谱检测一下那个>50 kD的band是不是确定存在
4.如果你的gel是native gel without reducing reagent,你的蛋白质(们)会不会在空
气中氧化形成共价disulfide bond,可以很简单地加点还原剂做对比实验.
5.条件允许的情况下用分析超高速离心sedimentation velocity analytical
ultracentrifugation跑几个不同浓度的样品,看一看有没有reversible dimerization
进行,或者这两个条带分别是两个不相互作用的蛋白质,这个方法比SEC柱子resolution
高很多.
6.co-IP的artifect不少,最好用其他方法检验来确认

我是天天在玩PHREEQC，不过PHREEQC的那个TRANSPORT Module是个简化版的1D
reactive transport，只能用于一些简单流场的地下水化学计算，我一般拿它来试算一
下，然后用USGS开发的相应的一个3D的版本PHAST来做更大domain的3D model。PHAST类
似于PHT3D，个人认为比PHT3D还强一点，虽然没有像PHT3D那样被整合到了最新版的
VMODFLOW里，但USGS的维护和开发还是很不错的，而且最关键的是免费以及source
code是公开的。欧洲有人把PHREEQC和Hydrus1D合在一起做了个HP1http://www.sckcen.be/hp1/index.shtml，其实就是把transport这部分用Hydrus1D来算了，本质上还是operator splitting的原理来couple化学反应和transport的。类似的还有一个叫CoTReM，据说还可以考虑sedimentation过程。
至于你要做的东西，我不知道你所谓的动态模型究竟指的是什么。如果认为河水不动，

雨水的pH值在任何时间都小于河流的pH值;降雨时RUNOFF中的H+总量和河流增加的H+总
量比较接近...?
还有,"下雨对河流的pH影响很大",这既可能与overland flow, interflow有关,也可能
与河床的sediment resuspension有关.

only, dirty water will come out of sediments naturally and slowly. Basically
you have to throw away your mop/paper towel after cleaning kitchen sink. If
leave it in the sink and refill with fresh water, after a while, the sink
