serine突变成alanine没有表型。reviewer认为这个位点在正常生理情况下不怎么重要
，并要求解释原因。我的解释是这个位点在in vivo 被高度调控，可能有phosphate不
停的去磷酸化；也可能只有在特定的条件下才被磷酸化。
有人知道有这样的例子吗？只有D mutation 有表型而A mutation 没有表型的。

导致水解的。
一般认为DNA酶的作用是把镁离子放在一个最有利的位置从而加快这个水解过程。如果
用一个比方的话，如果说DNA酶是枪，那么镁离子就是子弹。
其实从一个化学的角度来说，大部分酶的作用无非就是把氨基酸侧链上的基团（COOH,
SH, OH,NH3, etc.)或者蛋白结合上的小分子（离子或者有机小分子)放到合适的地方让
他们进行水解或者合成（水解倒过来）反应而已。
比方说什么serine-protease, 无非就是用serine的侧链上的-OH来搞搞氢键什么的来促
进水解而已。 |

理工的庆宏教授发表的LRRK2能够直接结合a-Synuclein并且磷酸化Serine-129位点。
但是后续大量的综述讨论的时候提到他这篇文章时说this is the only one paper
show LRRK2 can phosphorylate S129-synuclein。我看他发表的这篇BBRC已经被引用
快超100次了，如果文章的结论有问题，那岂不是严重阻碍了关于LRRK2-G2019S的研究

Wiki上的解释好像whey protein是在牛奶里占20%的溶解性不受pH影响的蛋白呢。我没
有具体查过成分，不过你的营养师推荐，那估计是因为这个蛋白里含你儿子需要的氨基
酸多？看分子结构Serine确实是一个溶解性不受pH影响的氨基酸。
你也别把Whey蛋白粉给妖魔化了啊，咱们喝这个还不是因为喝着方便。我们喝牛奶也行
啊，不过相同的蛋白摄入量我们要喝6-7杯牛奶才够；我们也可以吃肉啊，不过肉里面
很多胆固醇脂肪什么的，也不能多吃。对了，Whey是用牛奶制造奶酪后的副产品，相同
的蛋白含量应该比牛奶要便宜吧。
退一步说，你家的孩子是儿子，长点肌肉也不是啥坏事啊，这边的小男孩不是越壮越受

一点EDTA, 1mM, 绝大部分serine protease没有Mg离子都没有活性了，有的蛋白要加DTT或者beta mecapitalmethanol,保持溶液的还原状态，视情况而定。
然后分装，用液氮进行迅速冷却，然后保存在-80, 有些比较脆弱的蛋白即使这样保存也会失活，那就需要找到合适的buffer，或者每次都用fresh made的蛋白
推荐你去读一下一个科大师兄写的从纯化到星辰，里面提到了很多蛋白提纯跟保存的技
术，对于做蛋白的很有帮助 |

我要检测的酶是血浆中的某种serine 蛋白水解酶,血浆中类似的酶很多.我现在知道的
就5种.我的短肽里面 有6个HIS(是连接片断,我写第一帖时没算进去) 和6 个GLYCINE,
血浆中是否有识别这些位点的酶,我还不清楚( 请大家不要BS我) 其他的有丝氨酸,异亮
氨酸,谷氨酸和精氨酸. 都可能被非特异切割.
我的酶识别位点主要是精氨酸和它前面的甘氨酸,这个在我合成片断的中间.
这回说清楚,各位看看是不是更无解了?

如果你表达的非kinase, 有少许可能被修饰，可能在Serine, Threonine, Tyrosine,
Histidine, Aspartate被修饰。
一旦被修饰而你又很介意的话，可以co express一些phosphatase, 或者invitro
dephosphorylation. |

这个药最早是由 Schering-Plough开发的，这个公司在２００９年被 Merck 买下.
Boceprevir 属于蛋白酶抑制剂，特异性的抑制hepatitis C virus 的一个蛋白酶NS3
protease ，而导致病毒蛋白不能被完全处理而无法组装和成熟.
由于丙种肝炎病毒的寄主特异性和在体内的感染过程的特殊性。该病毒目前没有有效的
体外细胞培养系统，也缺乏可以大规模筛选的动物模型，所以无法采用小分子筛选方式
。该药的开发几乎是纯粹由蛋白结构研究和binding prediction 推导开的，在对NS3
protease进行结晶之后，研究组在一些已知信息的基础上，对蛋白酶的活性基团serine 的侧链进行了抑制物设计
。在得出了最初的蛋白多肽lead 之后进行了进一步的优化，
最终得出了一个最有效的分子结构。

非常非常多的通路都可能有关系， 所以很难直接推测。
一般是把磷酸化的氨基酸突变了，看看会引起什么表型，然后再推测。
可以突变成不能磷酸化的集团，或者把serine, thronine变glutamate 来
模拟磷酸化后的结果 |
