能是因
为里面含有genomic DNA的原因，会比较sticky，虽然跑SDS-PAGE胶的时候sample不是很
sticky，但是跑出来之后做western发现整条lane城smear状，特别是大size的protein
，有人建
议我做一下sonication把genomic DNA震碎，我做了但是貌似不起作用，还有人建议我
treat
with DNase，但是我从来没做过，不知道这个会不会起作用，这里有没有人做过类似的
实验，如果
DNase treatment有作用的话，要加多少量并且在什么条件下treat多长时间呢？
如果这个方法也不行的话，请问还有什么别的办法可以解决smear的问题？

这玩意是可以直接取细胞进行检查，其他的都要等到发病了后才会检查，要是胰腺癌也
像pussy那样，每年取个细胞smear下，估计夹不死也不用换那肝 |

都不如 CsCl 的高。我用 CsCl 纯化的 BAC 放很久都不会有降解，跑电泳也不会看到
smear，但是 kit 纯化的 DNA 就不行，放两三个月跑个胶就能看到降解的 DNA smear

域，普通PCR跑胶没有见到可见的smear或者dimer。用ABI的SYBR green master mix
dissociation curve能见到多个peak，amplication curve也很奇怪。跑胶可以看到
smear和dimer...最简单的方法是换引物。。。但是俺要扩的区域CG含量很高。。。换
了primer估计还会有类似问题。。。
对付high CG常用的DMSO/Betaine啥的，在real-time PCR中有没有类似应用？
求做过high gc real-time PCR的大侠指点迷津啊 |

看到smear，然后切下比如450-500bp的区间做PCR。但可能是50bp的interval太小或者
自己技术不好，所以最后PCR出来的亮度比较低。（当然也不排除ligation efficiency
不好）
PCR的结果如图。
还没拿去测浓度。只想问问有经验的前辈这样的情况能拿去测30 coverage的WGS吗？
另外还有一个问题，就是，我们切胶的smear每一个区间（300-400，400-500，500-600
）这些区间里都一定cover了genome的每一个部分吗？我总担心genome的有的部分没有
被涵盖到，那岂不是就人为引入了很多bias？

但我接着这里也有俩问题：
1. 从板子上挑选不同的colony摇菌，提质粒，测序，发现有的成功引入突变，有的没
有引入突变，有的甚至出现其他地方（不应该出现的）什么deletion之类的。。
这如何解释呢
2. 8kb的target of interest大概是2.5kb；我现在是设计10对引物覆盖这2.5kb，等于
把整个2.5kb都测一下。。。需要这么做吗？因为虽然大家说Pfu很高保真，但我依然害
怕引入其他的乱七八糟的东西，毕竟PCR的产物是一堆smear。。。其实我都好奇这种
long-range PCR到底发生了什么。。。 |

经历了跨州搬家，CS, OB, 第二个宝宝出生，申请，面试。
去年6月就买了uw ， 但只做了mcq, 后来肚子太大懒得去考，就搁下了。直到差不多面
完，12-1月才pickup。这次又买了3个月的uw mcq+ccs, 重新做题。MTB是上次看的，这
次没看，但是看了一遍CK first aid, 因为很多基本的东西都忘了。Family practice
看了一半。从考试内容来看，如果说什么东西还占上点边，那就是uw了。还有CK基础很
重要。我可以这么说，pony要是明天就去考step3, 假设她ccs已经练好了，肯定是99，
因为实在没有什么额外的东西。我考的不好是因为基础就不扎实，再让我看上一年也就
这样了，因为脑容量有限，记不住就是记不住。
第一天7个block, 5个office觉得还能handle。那2个 ER, 题目又臭又长，很多看了2遍
也不知所云，勉强做完。前一晚又没睡，考时觉得恶心，然后跳出来的图又增加了恶心

内科第一年intern
从去年开始，traditional call system（每3、4、5天call一次）不符合现在的要求，
现在新规定出来后intern连续工作不能超过16or24小时（不确定）
自己所在program，有night float，从前天晚上（偶不确定是晚上还是早上。。。）六点到第二天早上七点半，每人每年轮一个月
不值night float的时候，就值白班
一、Intern之前的准备：
（一）Limited license：
根据所在州的决定，工作一、两年后，可以申请full license，正式去program上岗前，必须拿到limited license，最好在7月1日前， 有了这个之后，才能有以下的两个号：NPI（你的全国的编号）、DEA（你签名开药需要）
tele所在州board of medicine一个月开一次会，review一次你所交的资料
tele是七年制：毕业证书是五年的时候拿到，但在五年之前没有实习，是七年的后两年实习，类似这种情况，任何时候有discrepency，就要自己所在的医学院出具文件说明

查，没事。预约了医生，2周后，终于找到一个华人医生，但我真担心到时医生就给做
个pap smear 就完了。前2次去见Np都是做个pap smear 就完事，告诉结果阴性，也不
检查到底是什么原因导致的。B超都是我自己要求才做的。
另外暂停剧烈运动，就去健身房跑跑步和游泳算剧烈运动吗？？
另外，关于粉红色分泌物都有一段时间了可能从去年10月份出现，当时量很少，就没太
在意，今年大概2月份开始出现分泌物有味道，我也不知道是不是正常的味道，但以前
没这么重，也没放心上。还有就是在3，4月份月经期刚完就有去游泳，后来出现过一段
时间下体瘙痒，但也没吃药也没检查就自己好了。 |

，医生怀疑是活动期结核，但我没症状。有人说也有可能是以前留下的scar。痰培养是
指sputum culture吧？（对不起我是美国毕业的）我那个痰培养要两个月后才能知到，
现在知道的是sputum smear， 不知中文怎么叫，三个smear都是negative。医生说要

我以前和人讨论，有人总说中医千变万化，因人而异。所以无法标准化，无法做双盲等
一系列实验，不仅仅拒绝科学，连逻辑都拒绝（这玩意不是科学）。
实际上这里面有个问题。如果每个人的病情是不相关的，完全各异，那么这意味着中医
理论体系过于复杂，其状态相空间非常大。这有个坏处是医生不可能通过统计积累经验
，中医也就不存在合格的医生，因为医生遇到的下一个病人很难和前面经验相同，他将
无法做出正确判断，在3分钟诊断时间内无法做出无数逻辑判断。
如果不同人的病情不是完全无关的，是可以通过逻辑演绎的。（比如知道y=x+5后，可
以推导出x为任何数时y的数字）那么意味中医诊断的状态相空间有限，即中医诊断可以
标准化，可以进行统计测量（比如拟合y=x+5），也就无所谓因人而异，千人千方了，
出现千人千方，很可能只是其中有不起作用的干扰（smearing）。医生看病时，一定只
做了少数观察和逻辑判断，就有结论，而不需要对可能存在的所有变量进行考察，这意
味相空间是有限的。
